NHP gene expression after 4 Gy total body irradiation.

ABSTRACT: In the current study, we investigated the gene expression response of blood cells of non-human primates that were total body irradiated with a 4 Gy dose. We isolated total RNA from peripheral blood at 3 days before irradiation, and then from the same animals on days 3, 5 and 7 days after irradiation. Using Agilent Rhesus microarrays, we identified genes that were significantly differentially expressed across the 7-day time course of this study. We identified common biological functions affected that persisted across the 7-day study.

Project description:In the current study, we investigated the gene expression response of blood cells of non-human primates that were whole thorax irradiated with a 10.1 Gy total dose. Partial irradiation of the NHP in the upper half of the body allows study of late radiation lung injury while avoiding acute respiratory syndromes related to hematopoietic and gastrointestinal injury. In this study, we report gene expression changes in the peripheral blood, an easily biopsied tissue, up to a month after radiation injury to the lungs. We isolated total RNA from peripheral blood at 3 days before irradiation, and then from the same animals on days 2, 5 and 30 after irradiation. Using Agilent Human Whole Genome microarrays, we identified 1187 genes that were significantly differentially expressed across the 30-day time course of this study. We identified common biological functions affected that persisted across the 30-day study, such as immune response. Response to oxygen-containing compounds and bacterial molecules were implicated by the gene expression changes at both the earliest day 2 and last, day 30 time-point and suggest that although cells are being recycled through the body in a 30-day time course after exposure to irradiation, the damage to blood cells and immunity, specifically the response to infections might persist throughout the study .

Longitudinal multi-omic changes in the transcriptome and proteome of peripheral blood cells after a 4 Gy total body radiation dose to Rhesus macaques.

Project description:Background: Non-human primates, such as Rhesus macaques, are a powerful model for studies of the cellular and physiological effects of radiation, development of radiation biodosimetry, and for understanding the impact of radiation on human health. Here, we study the effects of 4 Gy total body irradiation (TBI) at the molecular level out to 28 days and at the cytogenetic level out to 56 days after exposure. We combine the global transcriptomic and proteomic responses in peripheral whole blood to assess the impact of acute TBI exposure at extended times post irradiation. Results: The overall mRNA response in the first week reflects a strong inflammatory reaction, infection response with neutrophil and platelet activation. At 1 week, cell cycle arrest and re-entry processes were enriched among mRNA changes, oncogene-induced senescence and MAPK signaling among the proteome changes. Influenza life cycle and infection pathways initiate earlier in mRNA and are reflected among the proteomic changes during the first week. Transcription factor proteins SRC, TGF and NFATC2 were immediately induced at 1 day after irradiation with increased transcriptional activity as predicted by mRNA changes persisting up to 1 week. Cell counts revealed a mild / moderate hematopoietic acute radiation syndrome (H-ARS) reaction to irradiation with expected lymphopenia, neutropenia and thrombocytopenia that resolved within 30 days. Measurements of micronuclei per binucleated cell levels in cytokinesis-blocked T-lymphocytes remained high in the range 0.27-0.33 up to 28 days and declined to 0.1 by day 56. Conclusions: Overall, we show that the TBI 4 Gy dose in NHPs induces many cellular changes that persist up to 1 month after exposure, consistent with damage, death, and repopulation of blood cells.

Transcriptome analysis of LT-HSC isolated from WT and CD169DTR/+ mice 5 days after a 2 Gy-Total Body Irradiation

Project description:A 2 Gy-total body irradiation (TBI) of WT mice induces a second wave of ROS and a second wave of apoptosis in hematopoietic stem cells starting from 6 days after irradiation. In CD169DTR/+ mice, which are depleted in CD169+ resident macrophages after a diphtheria toxin injection, these two waves do not occur and a total recovery of the LT-HSC pool is observed, contrarily to the LT-HSC pool from WT mice, which is decreased. Our goal is to understand why these secondary waves of apoptosis and ROS occur in LT-HSC from irradiated WT mice and not in LT-HSC from irradiated mice depleted in resident macrophages, allowing a recovery of the LT-HSC pool. We used microarrays to compare the global gene expression in LT-HSC isolated from WT and CD169DTR/+ mice 5 days after irradiation, i.e. just before both secondary waves of ROS and apoptosis occurred in WT mice.

6 Gy gamma-irradiation of mouse small intestinal organoids at 24h, 48h and 96h after irradiation

Project description:The intact intestinal epithelial barrier protects our body from a range of immune mediated diseases. The epithelial layer has an impressive ability to reconstitute and repair upon damage and this process of repair is increasingly seen as a therapeutic target. In vitro models to study this process in primary intestinal cells are lacking. Therefore we established and characterized an in vitro model of intestinal damage and repair by applying γ-radiation on small intestinal organoids. We then used this model to identify novel regulators of intestinal regeneration. Small intestinal single crypts were isolated from C57BL/6 mice and cultured in matrigel. We treated the organoid cultures, 3 days after seeding, with 6 Gy of gamma-irradiation. RNA was isolated at 24, 48 and 96 hours after irradiation. Non-irradiated controls were collected per time point.

Whole blood gene expression profiling at day 1, 3 and 7 after total-body irradiation in Gottingen minipigs

Project description:Gottingen minipigs mirror the physiological radiation response observed in humans and hence make an ideal candidate model for studying radiation biodosimetry for both limited-sized and mass casualty incidents. We examined the whole blood gene expression profiles at day 1, 3 and 7 after total-body irradiation with 1.7, 1.9, 2.1 and 2.3 Gy doses of gamma-rays. Blood taken one day prior to radiation served as control. Six animals per dose group were used The minipigs were monitored for up to 45 days or time to euthanasia necessitated by radiation effects. Survival correlative signatures were identified from the data.
