Diethyldithiocarbamate, an anti-abuse drug, alleviates steatohepatitis and fibrosis in rodents

ABSTRACT: C57BL/6 mice received methionine/choline deficient (MCD) diet to establish the model of non-alcoholic fatty liver disease (NAFLD) with or without Diethyldithiocarbamate (DDC) treatment. DDC improves hepatic steatosis, ballooning, inflammation and fibrosis in rodent models of NAFLD through modulating lipid metabolism and oxidative stress.

Project description:The pathogenesis of non-alcoholic fatty liver disease (NAFLD) is associated with abnormalities of liver lipid metabolism, especially with the accumulation of saturated fatty acids (FA). On the contrary, a diet enriched with n-3 polyunsaturated FA (n-3-PUFA) has been reported to ameliorate the progression of NAFLD. The aim of our study was to investigate the impact of dietary n-3-PUFA enrichment on the development of NAFLD and liver lipidome. Mice were fed for 6 weeks either high-fat methionine choline-deficient diet (MCD) or standard chow (two groups fed MCD, two control groups, both with or without n-3-PUFA). Genome-wide transcriptome analysis of liver tissue was performed and revealed differences in liver mRNA transcriptomes after MCD as well as n-3-PUFA administration.

The critical and diverse roles of CD4-CD8- double negative T cells in non-alcoholic fatty liver disease

Project description:Methods: RNA transcription profiling of intrahepatic CD4-CD8- double negative T cells were compared between C57BL/6 mice fed with control diet and methionine-choline deficient diet (MCD) for 5 weeks. Results: RNA sequencing data showed that intrahepatic DNT in NAFLD mice exhibited markedly decreased Prf1, Gzmb but increased Il17a expression.

Project description:We report the effect of NAFLD on liver gene expression changes that could impact tetrachloroethhylene metabolism Methods: We fed male C57Bl/6J mice a base diet (BD), high fat (HFD), or methionine/choline/folate deficient high fat diet (MCD) for 8 weeks and then treated them with vehicle or tetrachloroethylene (PERC, 300 mg/kg ig). We only report "basal" differences herein (aka vehicle-treated). Results: We report that there were diet-specific differences in xenobiotic metabolizing genes, and that these genes may be responsible for NAFLD-induced disruption in PERC metabolism

Transcriptional profiling of mouse liver tissue following control, choline and methonine/choline deficient diet intervention

Project description:Choline and methionine/choline deficient diets are widely used to generate severe rodent hepatic steatosis and steatohepatitis in an attempt to reflect stages of human non-alcoholic fatty liver disease (NAFLD). The underlying mechanism of hepatic injury in these models, and how this reflects human disease remains incompletely understood. We used detailed transcriptional analysis to interrogate the molecular mechanisms of this intervention and its similarity to human disease. Adult C57Bl/6J mice were maintained on control, choline deficient (CDD) or methionine/choline deficient (MCDD) diets for 4 weeks. Isolated liver RNA was used for transcriptional profiling by micro array analysis.

We performed transcription factor binding experiments and show that PPARs are dysregulated in the contect of PC-TP depletion We then performed gene expression profiling analysis using data obtained from RNA-seq of 3 Mice for WT and KO fed either normal chow or methionine and choline depleted chow.

TSPO ameliorated the liver fibrosis in non-alcoholic fatty liver disease mice

Project description:The 18-kDa translocator protein TSPO is a conserved ubiquitous high affinity cholesterol binding protein localized in the outer mitochondrial membrane. It plays a critical role in the segregation and transport of cholesterol from the outer to the inner mitochondrial membrane in steroidogenic cells. Imaging studies in humans, using specific TSPO ligands, showed that TSPO is highly expressed and accurately mirrors the histological picture of non-alcoholic fatty liver disease (NAFLD)/nonalcoholic steatohepatitis (NASH). However, the functions of TSPO in simple steatosis (SS) and NASH are unknown. In the present study, we examined the TSPO functions with in vivo NAFLD model. To induce NASH in vivo, we fed the WT and TSPO KO rat with MCD (methionine-choline deficient) diet. Loss of TSPO ameliorated the liver fibrosis through down regulation of bile acid synthesis by reduction of CYP7A1 and CYP27A1 and increase of farnesoid X receptor (FXR).
