Project description:We found genetic deletion of IKKβ in mdx cardiomyocytes improved cardiac function and normalized calcium transients, and that differential gene expression favored a repressive function for NF-[kappa]B in these mice. We suspected that NF-[kappa]B was conferring a more restrictive chromatin conformation in a subset of genes. ChIP-seq was performed for H3K27 acetylation to show this repressive function for NF-[kappa]B was a global effect and to identify specific genes with more permissive chromatin conformation in hearts of mdx mice with IKKβ-deficient cardiomyocytes compared to mdx mice with intact IKKβ signaling.

Project description:G protein-coupled receptor kinase 2 (GRK2) has emerged as a key regulator of cardiac function and myocardial structure. Cardiac GRK2 is increased in heart failure and ischemia in humans, whereas genetic inhibition of GRK2 is cardioprotective in animal models of these pathologies. However, the mechanistic basis underlying these effects are not fully understood. We have used adult GRK2 hemizygous mice (GRK2+/-) as a model to assess the effects of a sustained systemic inhibition of GRK2 in heart tissue with age. We used microarrays to determine the global programme of gene expression underlying cardioprotection in GRK2 hemizygous mice.

RNA-seq analysis of hearts from control and cardiomyocyte-specific PRMT1-deficient mice

Project description:We found that cardiomyocyte-specific PRMT1-deficient (PRMT1-cKO) mice showed dilated cardiomyopathy and aberrant cardiac alternative splicing. To identify novel cardiac splicing events, we performed a comprehensive analysis of gene expression changes in hearts of wildtype (WT) and PRMT1-cKO mice using RNA sequencing (RNA-Seq). To investigate differentially expressed genes (false discovery rate (FDR) p<0.05, fold change >2) between control and PRMT1-cKO mice, we performed pairwise comparisons of RNA-Seq data using the CLC Genomics Workbench software.
