A human TH17 population with a tissue-resident signature in healthy and inflamed oral mucosal tissues [Bulk RNA-seq]

ABSTRACT: T cells in mucosal tissues fulfill a complex array of duties to ensure maintenance of barrier immunity. In oral mucosa tissue, we found that increased inflammation altered CD4 T cell subsets in a spatially-dependent manner, although it had a modest effect on the frequency of tissue-resident memory T cells (TRM) and the CD4 T cell transcriptome. In contrast, localization to the tissue profoundly altered the transcriptional profile, emphasizing the importance of studying healthy tissue to understand disease-specific changes. Our data revealed the existence of a TH17 cell population that is predominantly found in the tissue-resident, but not transient, CD4 T cell compartment in mucosal tissue.

Influenza-trained mucosal-resident alveolar macrophages confer long-term antitumor immunity in the lungs [ATAC-seq]

Project description:Respiratory viral infections reprogram pulmonary macrophages with altered anti-infectious functions. However, the potential function of virus-trained macrophages in antitumor immunity in the lung, a preferential target of both primary and metastatic malignancies, is not well understood. Using mouse models of influenza and lung metastatic tumors, we show here that influenza trains respiratory mucosal-resident alveolar macrophages (AMs) to exert long-lasting and tissue-specific antitumor immunity. Trained AMs infiltrate tumor lesions and have enhanced phagocytic and tumor cell cytotoxic functions, which are associated with epigenetic, transcriptional and metabolic resistance to tumor-induced immune suppression. Generation of antitumor trained immunity in AMs is dependent on interferon-γ and natural killer cells. Notably, human AMs with trained immunity traits in non-small cell lung cancer tissue are associated with a favorable immune microenvironment. These data reveal a function for trained resident macrophages in pulmonary mucosal antitumor immune surveillance. Induction of trained immunity in tissue-resident macrophages might thereby be a potential antitumor strategy.

Nanoengineered STING Mucosal Adjuvant Activates Coordinated Mucosal-Systemic Immunity for Durable Cross-Protective Defense

Project description:A longstanding barrier in vaccinology has been the inability to reconcile potent mucosal immune activation with systemic tolerability, hindering adjuvant development for over a century. Here, we present NanoCF501, a nanoparticulate STING agonist engineered with respiratory-tuned, clinically safe polymers, which overcomes this fundamental limitation. When co-administered with a multivalent pan-β-coronavirus subunit vaccine, NanoCF501 at 1/20th the systemic adjuvant dose elicits coordinated mucosal-systemic immunity, inducing robust and durable cross-protective responses. These include mucosal secretory IgA and tissue-resident memory T cells alongside systemic neutralizing antibodies, memory B cells, and long-lived plasma cells persisting >12 months in murine models. Non-human primates validate the dual-axis mucosal-systemic coupling, while toxicology studies in rats demonstrate more than 18-fold safety margin and undetectable systemic exposure, underscoring translational viability. Mechanistic interrogation via single-cell transcriptomics reveals that NanoCF501 drives STING-dependent innate immune activation, orchestrating broad transcriptional reprogramming in lung antigen-presenting cells and efficient priming of adaptive T and B cell responses. Notably, NanoCF501 converts conventional quadrivalent influenza subunit vaccines into mucosal immunogenic formulations, demonstrating platform versatility. By integrating rational nanocarrier design with innate immune targeting, this work establishes a universal mucosal vaccination strategy with broad implications for pandemic preparedness and mucosal vaccinology.

Human tissue-resident memory T cells are defined by core transcriptional and functional signatures in lymphoid and mucosal sites

Project description:Tissue resident memory T cells (TRM) mediate optimal protection to site-specific pathogens in mouse models and are key targets for vaccines. In humans, the existence and defining properties of Positive remain unclear. Here, we performed in-depth transcriptome profiling and phenotypic and functional analyses of circulating and resident memory CD4+ and CD8+T cells from lungs, spleens, blood and intestines of 72 individuals, revealing a core gene set conserved between tissues, species, and T cell lineages. Human Positive exhibit increased expression of specific adhesion and regulatory molecules, increased capacity for pro-inflammatory and regulatory cytokine production, and downregulation of molecules controlling responses to chemokines, tissue egress, and cell cycle control compared with circulating Negative cells. We further identify subsets of genes exhibiting tissue-specific expression patterns for mucosal and lymphoid sites. Our results indicate that human Positive constitute a subset distinct from circulatory Negative, with important implications for promoting anatomic localization of memory responses.
