Transcriptomic changes induced by a new Src Family Kinase Inhibitor and proton radiation in glioblastoma cells

Project description:Glioblastoma multiforme (GBM) is a central nervous system tumour. The current standard care for GMB is surgical resection, followed by conventional radiotherapy (RT) often combined with daily drug (temozolomide) administration. However, these treatment modalities are not currently curative and the resistance to both chemotherapy and RT plans is the main cause of GBM care failures. In this scenario, proton therapy (PT) could be used as a successful strategy for GBM treatment, being able to regulate the balance between tumour control and the normal tissue tolerance. In addition, new drugs could be used as radiosensitizing agents combined with PT in order to optimize GBM care. In this work we analyzed the GEP of the U87 MG human glioblastoma cell line, after treatment with PT alone or in combination with a new targeted compound, named Si306 (property of Lead Discovery Siena, Siena, Italy). We described GEP induced by both treatments, highlighting for the first time, the cell pathways induced by Si306. Summarizing, our results suggesting this compound as a novel possible candidate to treat GBM in combination with PT.

Single-nucleus and spatial landscape of the sub-ventricular zone in human glioblastoma

Project description:Glioblastoma (GBM) clinical management is challenging due to its heterogeneous nature, invasive potential, and poor response to radio- and chemo-therapy. As a result, GBM inevitably recurs and only a minority of patients survive 5 years post-diagnosis. We were the first to show that in the majority of GBM patients, the sub-ventricular zone (SVZ) of the lateral ventricles is a reservoir of cancer stem-like cells (CSCs) that show distinct patterns of treatment resistance when compared to matched CSCs from the tumor mass and contribute to the seeding of the recurrent tumor. However, the sampling and the characterization of the SVZ in GBM patients pose several challenges, as this area is extremely small and needs to be objectively identified during tumor surgical resection.

Project description:The invasive nature of glioblastoma (GBM) represents a major clinical challenge contributing to poor outcomes. Invasion of GBM into healthy tissue restricts therapeutic access and surgical resection. Therefore, effective anti-invasive strategies of GBM cells can be key to increase the efficacy of chemotherapy against this devastating disease. As cancer stem or initiating cells are considered to retain the tumorigenic potential in a number of tumors including glioblastoma, we studied the invasion capabilities of glioblastoma initiating cells (GICs) that were isolated from the peritumoral (PT) tissue, which surrounds the tumor mass (TM) and remains in the brain after tumor removal. Project description:Glioblastoma multiforme (GBM), is the most malignant and common tumor among glial neoplasms. It is characterized by a peak of incidence between 45 and 70 years and a patient median survival time of 14.6 months. In addition, GBM undergoes malignant progression under hypoxic conditions that contributes to strong radioresistance and chemoresistance; alters the metabolism of the tumor cells; generates a strong genome instability, increases the angiogenesis, vasculogenesis, and contributes to the formation of the Cancer Stem Cells (CSCs) and the infamous Circulating Tumor Cells (CTCs) involved in metastasis formation. Moreover, GBMs have a poor prognosis due to treatment (such as surgical resection, conventional RT and chemotherapy) plan failures. In this sense, since standard GBM treatments are not effective, new promising strategies such as Proton Therapy (PT), are under investigation because supported by encouraging results. Considering these assumptions, the main aim of this study was to analyze GBM response to PT innovative treatment during induced acute hypoxia, by using a whole gene expression analysis. Thus, in this work we have reported in a descriptive way, the most statistically and biologically relevant pathways deregulated in U87 GBM cell line, according to specific PT plans during hypoxia condition, and in addition, some interesting biomarker networks were following discussed.

Proteome profiling of extracellular vesicles from neurosurgical aspirates identifies CCT6A as a potential glioblastoma biomarker with prognostic significance

Project description:Glioblastoma, WHO-grade IV glioma, carries a dismal prognosis owing to its infiltrative growth rates and limited treatment options. Glioblastoma-derived extracellular vesicles (EVs; 30-1000nm membranous particles) influence the microenvironment to mediate tumour aggressiveness and carry oncogenic cargo across the blood-brain-barrier into the circulation. As such, EVs are biomarker reservoirs with enormous potential for assessing glioblastoma tumours in situ. Neurosurgical aspirates are rich sources of EVs, isolated directly from glioma microenvironments. Quantitative LC-MS/MS compared EV proteomes enriched from glioblastoma (n=15) and glioma grade II-III (n=7) aspirates and identified 298 differentially-abundant proteins (p-value<0.00496). Along with previously reported glioblastoma-associated biomarkers, levels of all eight subunits of the key molecular chaperone, T-complex protein 1 Ring complex (TRiC), were higher in glioblastoma-EVs, including CCT2, CCT3, CCT5, CCT6A, CCT7 and TCP1 (p<0.00496). Analogous increases in TRiC transcript levels and DNA copy numbers were detected in silico; CCT6A had the greatest induction of expression and amplification in glioblastoma and showed a negative association with survival (p=0.006). CCT6A is co-localised with EGFR at 7p11.2, with a strong tendency for co-amplification (p<0.001). Immunohistochemistry corroborated the CCT6A proteomics measurements and indicated a potential link between EGFR and CCT6A tissue expressions. Putative EV-biomarkers described here should be further assessed in peripheral blood.
