ABSTRACT: Endothelial cell-selective adhesion molecule (ESAM) is a life-long maker for hematopoietic stem cells (HSCs). Although we previously elucidated the functional importance of ESAM on HSCs in stress-induced hematopoiesis in adults, it remained unknown how ESAM affects hematopoietic development during fetal life. To address this issue, we analyzed fetuses of conventional ESAM-knockout mice.

Project description:We previously identified endothelial cell-selective adhesion molecule (ESAM) as a novel functional marker of hematopoietic stem cells (HSCs) in mice. To examine ESAM expression in human, we analyzed diverse HSC sources using flow cytometry. From mononuclear cells of collagenase-treated human hip bones, we obtained two ESAM positive populations in CD34(+) CD38(-) fraction, referred to as ESAM(High) and ESAM(Bright). Then we conducted microarray analyses comparing gene expression signatures between these two populations. Trabecular tissues of the femora were treated with collagenase IV and DNase. Mononuclear cells were collected, and CD34(+) CD38(-) ESAM(High) or ESAM(Bright) cells were sorted.

Microarray analysis of WT ESAM+ and ESAM- cDC subsets

Project description:Defense against attaching and effacing (A/E) bacteria requires the sequential generation of IL-23 and IL-22 to induce protective mucosal responses. While the critical source of IL-22 has been identified as CD4+ and Nkp46+ innate lymphoid cells (ILCs), the precise source of IL-23 is unclear. Here, we use genetic techniques to deplete specific classical dendritic cell (cDC) subsets and analyze immunity to the A/E pathogen Citrobacter rodentium. We find that Zbtb46+ cDCs, and specifically Notch2-dependent intestinal CD11b+ cDCs, but not Batf3-dependent CD103+ cDCs, are required for IL-23 production and immunity against C. rodentium. Notch2 controls cDC differentiation at a terminal step mediated by lymphotoxin signaling. Importantly, these results provide the first demonstration of a non-redundant function of CD11b+ cDCs in vivo. Analysis of differentially expressed genes in ESAM+ and ESAM- CD11b+ and DEC205+ splenic classical DC subsets. Splenocytes were harvested from WT C57Bl/6 or WT Cx3cr1-gfp mice and cDC subsets sorted to >95% purity on the FACSAriaII.

Inflammatory signaling regulates fetal hematopoiesis

Project description:Multiple signaling pathways contribute to blood cell formation in the fetus, but a role for innate immune/inflammatory signaling has not been described. Here we show that mouse fetuses deficient for either interferon gamma 1 (IFNg) or its receptor have decreased numbers of lymphoid progenitors and hematopoietic stem cells (HSCs) in the aorta/gonad/mesonephros region and placenta. The role of IFNg signaling was evolutionarily conserved, as morpholino knockdown of IFNg and its receptor reduced the number of hematopoietic stem and progenitor cells in the dorsal aorta and caudal hematopoietic territory of zebrafish embryos, and rag2 expressing cells in the thymus. ChIP-Seq analysis demonstrated that interferon regulatory factor 2 occupied genes in human fetal liver CD34+ hematopoietic stem/progenitor cells, and Gene Ontology analysis identified innate immunity and interferon signaling as enriched processes. Thus inflammatory signaling in the fetus, in the absence of pathogenic challenge, regulates the production of lymphoid progenitors and HSCs.
