Project description:Hydrogen sulfide (H2S), a metabolic endproduct of sulfate-reducing bacteria in the large intestine, represents a genotoxic insult to the colonic epithelium, which may also be linked with chronic disorders such as ulcerative colitis and colorectal cancer. The present study defined the early (30 minutes) and late (4 hours) response of non-transformed human intestinal epithelial cells (FHs 74 Int) to sulfide. We measured the genotoxicity of Na2S using the single cell gel electrophoresis (comet) assay. Changes in gene expression were analyzed after exposure to a genotoxic–but not cytotoxic–concentration of sulfide (500 μM Na2S) using pathway specific quantitative RT-PCR gene arrays.

Different race pacing strategies among runners covering the 2017 Berlin Marathon under 3 hours and 30 minutes.

Project description:The purposes of this study were 1) to analyse the different pacing behaviours based on athlete's performance and 2) to determine whether significant differences in each race split and the runner's performance implied different race profiles. A total of 2295 runners, which took part in Berlin's marathon (2017), met the inclusion criteria. 4 different groups were created based on sex and performance. Men: Elite (<02:19:00 h), Top 1 (<02:30:00 h), Top 2 (<02:45:00 h) and Top 3 (<03:00:00 h); women: Elite (02:45:00 h), Top 1 (<03:00:00 h), Top 2 (<03:15:00 h), Top 3 (<03:30:00 h). With the aim of comparing the pacing between sex and performance the average speed was normalized. In men, no statistically significant changes were found between performance group and splits. A large number of significant differences between splits and groups were found amongst women: 5-10 km Top 2 vs Top 3 (P = 0.0178), 10-15 km Top1 vs Top 2 (P = 0.0211), 15-20 km Top1 vs Top 2 (P = 0.0382), 20-21.1 km Elite vs Top 2 (P = 0.0129); Elite vs Top 3 (P = 0.0020); Top1 vs Top 2 (P = 0.0233); Top 1 vs Top 3 (P = 0.0007), 25-30 km Elite vs Top 2 (P = 0.0273); Elite vs Top 3 (P = 0.0156), 30-35 km Elite vs Top 2 (P = 0.0096); Top 1 vs Top 2 (P = 0.0198); Top2 vs Top3 (P = 0.0069). In men there were little significant differences based on athletes' performance which implied a similar pacing behaviour. Women presented numerous differences based on their performance which suggested different pacing behaviours.

500 metagenome-assembled microbial genomes from 30 subtropical estuaries in South China.

Project description:As a unique geographical transition zone, the estuary is considered as a model environment to decipher the diversity, functions and ecological processes of microbial communities, which play important roles in the global biogeochemical cycle. Here we used surface water metagenomic sequencing datasets to construct metagenome-assembled genomes (MAGs) from 30 subtropical estuaries at a large scale along South China. In total, 500 dereplicated MAGs with completeness ≥ 50% and contamination ≤ 10% were obtained, among which more than one-thirds (n = 207 MAGs) have a completeness ≥ 70%. These MAGs are dominated by taxa assigned to the phylum Proteobacteria (n = 182 MAGs), Bacteroidota (n = 110) and Actinobacteriota (n = 104). These draft genomes can be used to study the diversity, phylogenetic history and metabolic potential of microbiota in the estuary, which should help improve our understanding of the structure and function of these microorganisms and how they evolved and adapted to extreme conditions in the estuarine ecosystem.

Project description:Identifying the effect of the natural agent curcumin on global transcriptional activity of PShTert-AR prostate fibroblast cells over time with a view to understanding mechanisms of action over time. Treatment of PShTert-AR cells for 4, 8, 12 and 16 hours in PShTert-AR cells significantly altered the expression of approximately 1200 genes. Through non-hierarchical cluster analysis, these genes were classified into 3 distinct groups depending on their pattern of temporal response.

Renal tissue thawed for 30 minutes is still suitable for gene expression analysis.

Project description:Some biosamples obtained from biobanks may go through thawing before processing. We aim to evaluate the effects of thawing at room temperature for different time periods on gene expression analysis. A time course study with four time points was conducted to investigate the expression profiling on 10 thawed normal mice renal tissue samples through Affymetrix GeneChip mouse gene 2.0 st array. Microarray results were validated by quantitative real time polymerase chain reactions (qPCR) on 6 candidate reference genes and 11 target genes. Additionally, we used geNorm plus and NormFinder to identify the most stably expressed reference genes over time. The results showed RNA degraded more after longer incubation at room temperature. However, microarray results showed only 240 genes (0.91%) altered significantly in response to thawing at room temperature. The signal of majority altered probe sets decreased with thawing time, and the crossing point (Cp) values of all candidate reference genes correlated positively with the thawing time (p<0.05). The combination of B2M, ACTB and PPIA was identified as the best choice for qPCR normalization. We found most target genes were stable by using this normalization method. However, serious gene quantification errors were resulted from improper reference genes. In conclusion, thirty minutes of thawing at room temperature has a limited impact on microarray and qPCR analysis, gene expression variations due to RNA degradation in early period after thawing can be largely reduced by proper normalization.

Untreated controls were labeled with Cy5 red dye, and human skimmed milk treatment were labeled with Cy3 green dye. 4 pairs of control-Cy5/treated-Cy3 labeled samples were hybridized to a 4*44k Agilent human array (G4845A, AMADID 026652 Human V2)
