Gene expression analysis in fly ovaries growing from somatic tkv-knockdown gonads driven by c587-GAL4

ABSTRACT: Purpose: The goal of this study is to analyze genes affected by Tkv knockdown in the developing ovarian soma using RNA-seq. Ovarian mRNA prolife of one-day-old control flies and flies carring tkv-knockdown ovarian somatic cells duirng development by deep sequence, in duplicate, using Illumina NextSeq 500. The sequence reads that passed quality filters were analyzed at the transcript isoform level using TopHat followed by Cufflinks.

Project description:Purpose: The goal of this study is to analyze microRNA affected by wuho and mei-p26 mutations in Drosophila ovary Ovarian microRNA prolife of 7 day old control flies and flies bearing wuho and mei-p26 mutations, in duplicate.Sample libraries were prepared using the QIAseq miRNA Library Kit (QIAGEN) according to the manufacturer's protocols. Adaptors are ligated sequentially to the 3' and 5' ends of miRNAs. Subsequently, universal cDNA synthesis with UMI assignment, cDNA cleanup, library amplification and library cleanup are performed. Libraries were sequenced on an Illumina instrument (75-cycle single-end read, 75SE). Sequencing data was processed using the Illumina software BCL2FASTQ v2.20.

Gene expression during head development in a sexually monomorphic stalk-eyed fly

Project description:Stalk-eyed flies (family Diopsidae) are a model system for studying sexual selection due to the elongated and sexually dimorphic eye-stalks found in some species. These flies are of additional interest because their X chromosome is derived largely from an autosomal arm in other flies. To investigate how sex-biased expression arose on the novel X we compared gene expression between males and females using oligonucleotide microarrays and RNA from developing eyestalk tissue in the sexually monomorphic diopsid, Teleopsis quinqueguttata. We use probe sequence divergence to evaluate cross-species ascertainment bias and chromosome assignment to determine if sex linkage influence expression. Microarray analysis revealed sex-biased expression for only 1.9% of 3,748 genes expressed in eye-antennal imaginal discs. Analysis of probe sequences between species indicates that the lack of sex-biased expression is not due to ascertainment bias. These findings indicate that the the majority of sex-biased gene expression observed in developing heads of the dimorphic species, T. dalmanni, is causally related to development of dimorphic head shape. Project description:Canonical protein phosphatase 3 (Ppp3) / calcineurin signaling is central to numerous physiological processes. Here we provide evidence that calcineurin plays a pivotal role in controlling systemic energy and body weight homeostasis. Knockdown of calcineurin in Drosophila melanogaster led to a decrease in body weight, and energy stores, and increased energy expenditure. D. melanogaster piggyBac transposon insertion mutants (Thibault et al., 2004) (PBac{WH}CanA1[f01787] (Calcineurin A1) and PBac{WH}CanA-14F[f02374] (CanA at 14-F)) and the isogenic wildtype w1118 strain (BL-6326) were ordered from Exelixis Drosophila Stock Collection at Harvard Medical School. RNA was isolated from the thorax of 50 flies per sample. Whole genome tiling gene arrays were utilized to reveal distinctly deregulated gene patterns that appeared in both mutants (n=4, respectively) as compared to the WT controls (n=4).
