Thyroidal gene expression changes in tamoxifen inducible Foxe1 knockout mouse at two weeks post-treatment

Project description:To directly examine the role of Foxe1 in the adult thyroid we generated a model of tamoxifen-inducible Cre mediated Foxe1 deletion in mice of C57BL/6J background (Foxe1flox/flox/Cre). Tamoxifen (0.05g/kg) was administered intraperitoneally to Foxe1flox/flox/Cre mice or Cre control mice aged 6-8 weeks, once daily for five consecutive days. Thyroids were harvested two weeks post-treatment, total RNA extracted and transcriptomic analysis performed using Clariom S Mouse arrays.

Effect of cardiomyocyte specific KO of AKT1 + AKT2 on gene expression profile of the heart

Project description:In the mammalian heart AKT1 and AKT2 are the isoforms of the protein kinase AKT (protein kinase B) which are predominantly expressed. AKT isoforms exert common and specific functions in the field of metabolism, cellular growth, apoptosis and cell migration. To identify specific and common functions of AKT1 and AKT2 isoforms, we generated tamoxifen-inducible, cardiomyocyte-specific AKT1+AKT2 double knockout mice (iCMAKT – KO mice). Inactivation of AKT isoforms was achieved by application of 4-OH-tamoxifen, which activates an OH-Tx inducible cre-recombinase expressed under the control of the αMHC-promoter (αMHC-mercremer). Transgenic mice expressing only the αMHCmercremer construct were also treated with OH-Tx and served as controls. To identify alterations in cardiac gene expression due to AKT deletion we analyzed gene expression profiles of control hearts and iCMAKT1+2 hearts.

Decoupling astrocytes in adult mice impairs synaptic plasticity and spatial learning

Project description:To study the role of the gap junction coupled astrocytic network, we generated inducible double knockouts to selectively delete Cx30 and Cx43 from astrocytes in adult mice. Mice carrying loxP-flanked Gjb6 (Cx30fl/fl mice) (Boulay et al., 2013) and Gja1 (Cx43fl/fl mice) (Theis et al., 2003) alleles were crossbred with mice expressing the tamoxifen-sensitive Cre-recombinase CreERT2 under the endogenous GLAST(Slc1a3)-promoter (Mori et al., 2006). Adult, 8-10 week old mice (Cx30fl/fl:Cx43fl/fl:GLASTCreERT2/+, termed cKO) and littermate control mice (Cx30fl/fl:Cx43fl/fl:GLAST+/+) were injected with tamoxifen for 5 consecutive days and hippocampi were isolated for subsequent TMT-based proteomics analysis 90 days after tamoxifen treatment, to study the consequences of astrocyte decoupling and connexin deletion.
