Eumyc and WT NK cells

ABSTRACT: Eumyc lymphoma was inoculated i.v. into C57BL/6J mice. Spleens were harvested from healthy or Emyc lymphoma-bearing mice at day 14 post-inoculation, and splenic NK cells were purfied using flow cytometry.

At 10 days after tumor injection, mice (5/group) were treated with adoptive transfer of 2.5 × 106 Th1, Th9 or Th17 cells, followed by i.v. injection of 2.5 × 105 peptide-pulsed BMDC. Total RNA was extracted with RNeasy Mini kit (Qiagen) from CD45.1+CD4+ Th cells sorted from spleens of tumor-bearing mice 12 days after transfer.

Aging-triggered iron-rich insoluble protein aggregates in the spleen originate chiefly from red pulp macrophages.

Project description:Both iron homeostasis and erythropoiesis are known to be affected by aging. Iron needs in mammals are met primarily by iron recycling from senescent red blood cells (RBCs), a task chiefly accomplished by red pulp macrophages (RPMs) in the spleen. Given that RPMs continuously process iron, their cellular functions might be susceptible to age-dependent decline, a possibility that has been unexplored to date. In our project, we identified a formation of undegradable iron- and heme-rich extracellular aggregates in the spleens of 10-11-month-old female mice. To better understand the origin of these aggregates, here, we performed: i) protein identification and intrasample quantification (iBAQ) of proteins of magnetically-isolated red pulp macrophages from spleens of two female 8-weeks-old C57BL/6J (maintained on a standard diet) and ii) label-free quantification of proteins of the splenic protein aggregates formed in the mouse spleen 24 hours after intraperitoneal iron dextran injection, using dextran-injected mice as a control. Two 8-week-old C57BL/6J mice per group were analyzed. This dataset is related to the project PXD032900, which describes quanytitative analysis of proteins in aggregates magnetically isolated from spleen of aged (standard or iron-reduced diet) and young mice (standard diet).

Total RNA was extracted from the spleen tissues, and we used the RT² Profiler PCR array-mouse common cytokines (SABioscience) array panels to quantitate gene expression of cytokine genes from the spleens.

Lymphoma-induced regression of high endothelial venules impairs homeostatic lymphocyte trafficking

Project description:Lymphoma growth is facilitated by the well-balanced infrastructure of the microenvironment in lymph nodes (LN). LNs undergo rapid expansion during lymphoma progression, often accompanied by excessive blood vessel growth. Here we report that aggressive lymphoma cause severe high endothelial venule (HEV) regression which impairs lymphocyte recirculation. In this study, we used transferred (i.v.) Eµ-Myc tumor cells as a mouse model of aggressive lymphoma. Blood endothelial cells from tumor bearing and control LNs were isolated by FACS and processed for single-cell RNA sequencing. The results revealed a detrimental mechanisms causing HEV-dedifferentiation during lymphoma growth with potential implications on tumor-targeting immune surveillance and strategies of immune therapy in LNs

TCR repertoire analysis by Next-generation sequencing of mouse CD8+ T cells in spleen or tumor

Project description:We have reported that intra-tumoral treatment with 1V270, a phospholipid-conjugated TLR7 agonist,induces local expansion an systemic dispersion of oligoclonal tumor-specific T cells by TCR repertoire analysis using next generation RNAseq methodology. Here, we examined whether systemic 1V270 therapy also induced oligoclonal expansion of tumor-specific T cells. Two groups of BALB/c mice (n=4/group) were i.p. treated with 1V270,a phospholipid-conjugated TLR7 agonist. One cohort of mice was i.v. injected with 4T1-GLF cells (2×104) on day 0. Another cohort did not receive i.v. tumor injection (no tumor-exposed mice). 4T1 cells were orthotopically inoculated on day 21. To examine clonal specificity of tumor-specific T cells, CD8+ cells were isolated from the spleens and the tumor infiltrating lymphocytes of secondarily challenged tumors after initial 1V270 therapy. The TCR repertoires were assessed by next generation RNA sequencing of both TCRαand TCR β genes.

Microarray analysis on spleens and livers of mice inoculated subcutaneously with J558 myeloma cells and supplemented with oil palm phenolics (OPP)

Project description:OPP (1500 ppm gallic acid equivalent (GAE)) was supplemented to BALB/c mice inoculated subcutaneously with J558 myeloma cells at the dorsum of the neck in order to observe possible immune responses in cancer suppression. Spleens and livers were harvested at three designated time points, eighteen hours, one week and four weeks after inoculation of the cells for gene expression studies. Results show that the expression of immune-related genes in the spleen was delayed, while those related to inflammation in the liver were down-regulated across time, thus suggesting that OPP may have anti-inflammatory properties in suppressing cancer. Total RNA obtained from spleens and livers of BALB/c mice given OPP (four weeks after subcutaneous inoculation of J558 myeloma cells) were compared to controls given distilled water at three time points, eighteen hours, one week and four weeks after inoculation of the cells (three replicates per group per time point)
