Project description:This project is meant to document the effect of the negative energetic balance on the embryo quality and as it happens, to identify ways to improve the fertility of dairy bovines. Measurement of the β-Hydroxybutyric acid (BHB) has been taken between 45 and 60 days post-partum on twelve Holstein cows. Cows have been ranked according to the sanguine measurement as high or low in BHB in order to get 6 cows per group. After having synchronized the heat with hormones and an ovarian stimulation, cows have been inseminated and the embryos were selected as so to be transferred in at least two heifers. With this transfer it is possible to isolate the impact of the energetic level of the mother before and during the gestation on the performances of the future progeny.

Alterations of the Lipid Metabolome in Dairy Cows Experiencing Excessive Lipolysis Early Postpartum.

Project description:A decrease in insulin sensitivity enhances adipose tissue lipolysis helping early lactation cows counteracting their energy deficit. However, excessive lipolysis poses serious health risks for cows, and its underlying mechanisms are not clearly understood. The present study used targeted ESI-LC-MS/MS-based metabolomics and indirect insulin sensitivity measurements to evaluate metabolic alterations in the serum of dairy cows of various parities experiencing variable lipolysis early postpartum. Thirty (12 primiparous and 18 multiparous) cows of Holstein Friesian and Simmental breeds, fed the same diet and kept under the same management conditions, were sampled at d 21 postpartum and classified as low (n = 10), medium (n = 8), and high (n = 12) lipolysis groups, based on serum concentration of nonesterified fatty acids. Overall, excessive lipolysis in the high group came along with impaired estimated insulin sensitivity and characteristic shifts in acylcarnitine, sphingomyelin, phosphatidylcholine and lysophospholipid metabolome profiles compared to the low group. From the detected phosphatidylcholines mainly those with diacyl-residues showed differences among lipolysis groups. Furthermore, more than half of the detected sphingomyelins were increased in cows experiencing high lipomobilization. Additionally, strong differences in serum acylcarnitines were noticed among lipolysis groups. The study suggests an altered serum phospholipidome in dairy cows associated with an increase in certain long-chain sphingomyelins and the progression of disturbed insulin function. In conclusion, the present study revealed 37 key metabolites as part of alterations in the synthesis or breakdown of sphingolipids and phospholipids associated with lowered estimated insulin sensitivity and excessive lipolysis in early-lactating cows.

Text mining datasets of ?-hydroxybutyrate (BHB) supplement products' consumer online reviews.

Project description:The current dataset is obtained by text mining of ?-hydroxybutyrate (BHB) supplement products' consumer online reviews. The text data of 71 BHB products' consumer reviews were extracted with the aid of the Web Scraper Chrome extension. Then, a lexicon-based sentiment analysis approach was developed to classify the sentiment or polarity of BHB products' consumer reviews. Both word-level and sentence-level sentiment analyses were conducted to score the analyzed text snippets. In terms of word-level sentiment analysis, word clouds of selected BHB products' reviews were generated to give direct observation, and the statistics of high-frequent sentiment words were listed for comparison. In terms of sentence-level sentiment analysis, two factors such as flavor and package were taken into consideration to map the products' polarity distributions. Besides, the complex analysis provides us with the basic statistics of the analyzed BHB customer reviews data.

β-Hydroxybutyrate Effects on Bovine Caruncular Epithelial Cells: A Model for Investigating the Peri-Implantation Period Disruption in Ketotic Dairy Cows.

Project description:Ketosis is a metabolic disorder arising from a negative energy balance (NEB). It is characterized by high β-Hydroxybutyrate (BHBA) blood levels and associated with reduced fertility in dairy cows. To investigate the impact of BHBA on bovine caruncular epithelial cells (BCEC) in vitro, these cells were stimulated with different concentrations of BHBA. Cell metabolism and motility were examined using an MTT assay and Live-cell imaging. RT-qPCR was used to examine mRNA expressions of TNF, IL6, RELA, prostaglandin E2 synthase (PTGES2) and receptor (PTGER2) as well as integrin subunits ITGAV, ITGA6, ITGB1 and ITGB3. Stimulation with 1.8 and 2.4 mM of BHBA negatively affected cell metabolism and motility. TNF showed increased mRNA expression related to rising BHBA concentrations. IL6, RELA, ITGAV, ITGA6, ITGB1 and ITGB3 as well as PTGER2 showed no changes in mRNA expression. Stimulation with 0.6 and 1.2 mM of BHBA significantly increased the mRNA expression of PTGES2. This does not indicate a negative effect on reproductive performance because low BHBA concentrations are found in steady-state conditions. However, the results of the study show negative effects of high BHBA concentrations on the function of BCECs as well as an inflammatory response. This could negatively affect the feto-maternal communication during the peri-implantation period in ketotic dairy cows.
