Project description:Microarray analysis was used to compare the gene expression profiles of Deaf-1-transduced mouse mammary epithelial cells (MECs) relative to Deaf-1-deficient MECs. Experiment Overall Design: To investigate potential target genes of Deaf-1, primary mammary epithelial cells (MECs) were isolated from a Deaf-1-null mouse and immortalised by transduction with a retrovirus encoding the human papilloma virus (HPV16) E6/E7 proteins. Two clones were selected and transduced with either an empty retrovirus or one encoding Deaf-1. RNA was subsequently isolated from cells at 24 and 48 hours after transduction for Affymetrix analysis. High levels of Deaf-1 expression were demonstrated in cells transduced with the Deaf-1 retrovirus relative to controls following a 48 hour selection in puromycin. This time-point was therefore selected for further analysis. Experiment Overall Design: To compare the gene expression profiles of Deaf-1-transduced MECs relative to Deaf-1-deficient MECs, Affymetrix analysis was performed using the GeneChip® Mouse Expression Set 430 2.0 array which comprises 39,000 transcripts on a single array. Total RNA was harvested from two independent clones (C1 and E1) infected with either control or Deaf-1-expressing retrovirus making 4 samples in total. Clones C1 and E1 were treated as biological replicates in the subsequent analysis.

Project description:The jmjC-domain containing H3K4 histone demethylase JARID1B/KDM5B/PLU1 is over-expressed in human breast cancer and is a potential target for breast cancer treatment. To investigate the in vivo function of JARID1B, we developed a new strain of Jarid1b knockout mice and characterized the phenotypes in detail. Unlike previously reported knockout strains, the majority of our Jarid1b knockout mice are viable beyond embryonic and neonatal stages. Nonetheless, these mice exhibit decreased body weight, higher incidence of adult mortality and decreased female fertility. Furthermore, Jarid1b knockout mice show delayed mammary gland development. Mechanistically, loss of JARID1B leads to decreased serum estrogen levels and reduced proliferation of mammary epithelial cells in early puberty. In addition, in mammary epithelial cells, loss of JARID1B diminishes the expression of key regulators of mammary morphogenesis, including FOXA1, estrogen receptor α (ERα), and GATA3. Taken together, these results indicate that JARID1B positively regulates mammary ductal development through both extrinsic and cell-autonomous mechanisms. Project description:During puberty, robust morphogenesis occurs in the mammary gland and mammary stem- and progenitor- cells develop into mature basal- and luminal-cells to form the ductal tree. The receptor signals that govern this process in mammary epithelial cells (MECs) are incompletely understood. Here we focused on the EGF receptor and its downstream pathway component Rasgrp1, expressed in MECs. Rasgrp1-/- or point-mutated Rasgrp1Anaef MECs demonstrated increased activation of ERK-, AKT-, and S6- kinase pathways. Rasgrp1 perturbation delayed ductal tree maturation, accompanied by aberrant MEC proliferation and AKT signals. In colony formation- and 3D Matrigel- assays, loss of Rasgrp1 function leads to elevated progenitor cell proliferation and gain-of-function branching, both in EGFR-dependent manners. Our studies revealed that unbalanced EGFR signals in Rasgrp1-/- or Rasgrp1Anaef females did not impact the intrinsic ability to form basal and luminal cells, but that a Wnt-driven stem cell gene signature disappears when EGF is added to organoid assays.

Here, we show that a small molecules cocktail enables the in vitro generation of chemically-induced mammary epithelial cells (CiMECs) with milk-secreting properties from goat ear fibroblasts. The properties of CiMECs were highly similar to those of naturally isolated MECs. Notably, the CiMECs lineage reprogramming process may partially imitate the MECs developmental process from embryonic to milk-secreting stages. Interestingly, fibroblasts were firstly reversed back to embryonic ectoderm/surface ectoderm-like state. Our findings constitute a substantial step toward further promoting the application of transgenic mammary bioreactors, moreover, also offer a new strategy for obtaining other cell types by reprogrammed fibroblasts.
