Transcriptome analysis of Bacillus cereus strain F837/76 in response to porcine gastric mucin, accomplished by RNA sequencing

ABSTRACT: In the intestine, B. cereus comes in contact with the epithelial mucus layer. To investigate the overall response to that contact, total transcriptome analyses were performed using porcine gastric mucin (PGM), which is a commonly applied model for mucus interactions. Differential regulation of genes encoding enterotoxins and further virulence factors was of special interest.

Project description:The iron-binding glycoprotein lactoferrin exhibits anti-bacterial activity. To investigate the overall response of Bacillus cereus to lactoferrin, total transcriptome analyses were performed. Growth conditions were mMOD minimal medium, minimal medium plus 10 mg/ml lactoferrin, as well as minimal medium without iron source for comparison. Differential regulation of genes involved in iron acquisition as well as genes encoding enterotoxins and further virulence factors was of special interest.

Campylobacter jejuni Utilization of Mucin

Project description:While growing in the human intestine, C. jejuni grows within the mucus layer. The largest constituents of this layer are the large mucin glycoproteins. A transcriptomic profile of C. jejuni NCTC11168 growing in a mucin-containing minimal medium seeks to describe the effect of the presence of mucin proteins on the transcriptome of C. jejuni. Microarray data was collected from three independent biological replicates and 9 technical replicates for each biological replicate.

Comparative Genomics of Bacillus cereus and Bacillus anthracis

Project description:The aim of the study was to carry out a CGH study utilizing a set of 39 diverse Bacillus isolates. Thirty four B. cereus and five B. anthracis strains and isolates were chosen so as to represent different lineages based on previous characterizations, including MLEE and MLST (Helgason, Okstad et al. 2000; Helgason, Tourasse et al. 2004). They represent the spectrum of B. cereus phenotypic diversity by including soil, dairy and periodontal isolates in addition to virulent B. anthracis strains.

Transcriptional profiling of C. albicans in the presence of mucin versus absence of mucin

Project description:Candida albicans is the most prevalent fungal pathogen of humans, causing a variety of diseases ranging from superficial mucosal infections to deep-seated systemic infections. Mucus, the gel that coats all wet epithelial surfaces, accommodates Candida albicans as part of the regular microbiome where C. albicans resides asymptomatically in healthy humans. Through a series of in vitro experiments combined with genome-wide transcriptional profiling, we show that mucin biopolymers, the main gel-forming constituents of mucus, induce a new oval-shaped morphology in C. albicans in which a range of genes related to adhesion, filamentation, and biofilm formation are down-regulated. We also show that corresponding phenotypes are suppressed, rendering Candida incapable of forming biofilms on a range of different synthetic surfaces and human epithelial cells. Our data suggests that mucins can manipulate Candida physiology and we hypothesize that they are key regulators for retaining Candida in the host-compatible, commensal state. 3 biological replicates grown in log phase in the presence and absence of mucin for 8 hours. Experiments were performed using RPMI 1640 (Gibco 31800-089) buffered with 165mM MOPS and supplemented with 0.2% NaHCO3 and 2% glucose with and without 0.5% mucin.

New genetic biomarkers to differentiate pathogenic and clinically relevant Bacillus cereus strains

Project description:Bacillus cereus is the second leading cause of collective food poisoning in France. B. cereus is also associated with severe clinical infections leading to patient death in 10% of the cases. The emergence of B. cereus as a foodborne and opportunistic pathogen has intensified the need to distinguish strains of public health concern. In this work, by performing a screen on a large collection of B. cereus strains of varying pathogenic potential, we identified genetic determinants capable of discriminating B. cereus strains inducing negative clinical outcomes. The combination of 4 biomarkers is sufficient to accurately discern clinical strains from harmless strains. Three of the biomarkers are located on the chromosome, with a fourth one identifying a plasmid carried by most pathogenic strains. A 50 kbp region of this plasmid promotes the virulence potential of these strains and could thus be defined as a new pathogenicity island of B. cereus. These new findings help in the understanding of B. cereus pathogenic potential and complexity and may provide tools for a better assessment of the risks associated with B. cereus contamination to improve patient health and food safety.

Global gene expression analysis of Bacillus cereus to proanthocyanidin exposure

Project description:B. cereus RNA isolation were taken at OD 0.5 just before addition of 0.02 mg/mL EEC, and at 30 min of exposure. Water was added instead of EEC for control. Total RNA was isolated and we performed DNA microarray for genome-wide transcriptional analysis of B. cereus ATCC 14579 in the presence or absence of EEC. The microarray used in this study was custom-made B. cereus ATCC 14579 developed by Agilent Technologies (https://earray.chem.agilent.com/earray/). The B. cereus microarray design was based on the predicted chromosomal open reading frames (NCBI Accession No. AE016877). It targets the 5234 annotated CDS, three non-overlapping probes were designed, low-quality probes were eliminated, and finally 15242 could be spotted. Microarray data was normalized to 75th percentile and the difference was found to be significant by unpaired T-test (P = 0.05). Next, ratio changes of 2-fold (for up-regulated genes in the EEC stress condition) and 0.5-fold (for down-regulated genes in the EEC stress condition) were regarded as biologically significant. 596 were up-regulated and 495 were down-regulated.
