Project description:We compare miRNA profiling in two cell populations of mesenchymal stromal cells focusing on senescence-related changes: Human Dental Pulp Stromal Cells (hDPSCs) and human Periosteum-Derived Progenitor Cells (hPDPCs). After these cells were harvested, total RNA extraction and whole genome miRNA profiling was performed, and DIANA-miRPath analysis was applied to find the target/pathways. Our finding is consistent with the idea that the embryological origin influences cell behavior and the ageing process. Moreover, the two populations activate different downstream mechanisms. Our study strengthens the hypothesis that ageing is driven by numerous mediators interacting through an intricate molecular network, which affects adult stem cells self-renewal capability.

Project description:Cancer incidence escalates exponentially with advancing age, the mechanism of which remains obscure. Here we built up a chronological molecular clock at the single cell level for the mammary stem cell enriched population to depict the physiological ageing dynamics. We found the mammary ageing process was asynchronous and progressive, determined by the relative proportions of four distinct cellular states. The mammary ageing was initiated by a light senescence state with elevated NFkb, P53 signals, followed by a deep senescence state with reduced NFkb, P53 and enhanced PI3K-Akt-mTOR, Wnt, Notch and pluripotency activities succumb to cancer predisposition. Both of these senescence programs were regulated by a master mammary stem cell factor Bcl11b through modulation of multiple stress, longevity and pluripotency associated pathways. Depletion of Bcl11b triggered morphological, functional and molecular ageing-like phenotypes, and drastically enhanced DMBA-induced tumor formation. We further screened out a drug TPCA-1 that can elevate Bcl11b activity, rejuvenate mammary cells transcriptomically and significantly reduce the ageing-related cancer incidence. Our findings established a molecular portrait of progressive mammary cell ageing and elucidated the regulatory network bridging mammary ageing and cancer predisposition that can be modulated to control the cancer initiation, which has potential implications in management of cancer prevalence.

Project description:While much research has focussed on advanced stages (and mechanisms) of ageing, this fundamental process may commence at a relatively early age, impacting organ function and resilience throughout the adult lifespan. In male C57BL/6 mice, multiple phenotypic and transcriptional features of cardiovascular ageing were evident from 16 weeks of age, well in advance of 'middle age'. Phenotypic changes include declining cardiac and coronary reserves and resistance to ischaemic insult. Gene changes support early constitutive stress together with a plateau in transcriptome responsiveness to ischaemia, and declining induction of cardioprotective and quality control pathways vs. increasing induction of genes promoting signalling dysfunction, hypertrophy and cell death. These findings support cardiac ageing from early adulthood. Molecular changes reflect declining adaptive capacity/quality control, consistent with evolutionary theories of biological ageing.

Project description:To investigate ageing-related changes in cardiac transcriptome of FVB mice we performed differential gene expression profiling analysis using data obtained from RNA-seq of five life time points (4-, 8-, 10-, 12-, 14 months) by comparing older FVB mice (8-, 10-, 12-, 14 months) to young 4-month-old FVB mice (FVB vs. FVB analysis). Genes that were differentially expressed in FVB vs. FVB analysis were also annotated to the respective biological processes based on Gene Ontology database, assessed for their overrepresentation to indicate processes involved in cardic ageing process in FVB mice. To investigate heart failure-related as well as ageing-related changes in cardiac transcriptome of Tgαq*44 mice we performed differential gene expression profiling analysis using data obtained from RNA-seq of five life time points (4-, 8-, 10-, 12-, 14 months) by comparing Tgαq*44 to age-matched FVB mice (Tgαq*44 vs. FVB analysis). Project description:Understanding the relationship between parenchymal and supporting cell populations is paramount to recapitulate the multicellular complexity of native tissues. Incorporation of stromal cells is widely recognized to be necessary for the stable formation of stem cell-derived cardiac tissues, yet the types of stromal cells used has varied widely. This study systematically characterized several stromal populations and found that stromal phenotype and morphology was highly variable depending on cell source and exerted differential impacts on cardiac tissue function and iPSC-CM phenotype. Therefore, the choice of supporting stromal population can differentially impact the phenotypic or functional performance of engineered cardiac tissues.

Project description:Here we show that an unrestricted galactose diet in early life minimises pathology during replicative ageing in budding yeast, irrespective of diet later in life. Lifespan and average mother cell division rate are comparable between glucose and galactose diets, but markers of senescence and the progressive dysregulation of gene expression observed on glucose are minimal on galactose, showing these to be facets of ageing pathology rather than intrinsic parts of the replicative ageing process. Respiration on galactose is critical for minimising ageing pathology, and forced respiration during ageing on glucose by over-expression of Hap4 also has the same effect though only in a fraction of cells. This fraction maintains Hap4 activity to advanced age with low senescence and a youthful gene expression profile, whereas other cells in the same population that lose Hap4 activity undergo dramatic dysregulation of gene expression and accumulate aneuploid fragments of chromosome XII aneuploidy (ChrXIIr), which are tightly associated with ageing pathology.

Project description:Senotherapeutics are new drugs, which can modulate senescence phenomena within tissues and reduces the onset of age-related pathologies. Senotherapeutics are divided in senolytics and senomorphics. The senolytics kill selectively senescent cells, while the senomorphics may delay or block the onset of senescence. Metformin is used for treatment of diabetes since several decades. Recently, it has been evidenced that metformin may have anti-aging properties by preventing DNA damage and inflammation. We evaluated the senomorphics effect of metformin on the biology human adipose mesenchymal stromal cells (MSCs) treated for six weeks with therapeutic doses of metformin. The study was combined with proteome analysis of changes occurring in MSCs intracellular and secretome protein composition, this to identify molecular pathways associated with the observed biological phenomena. The metformin reduced the replicative senescence and cell death phenomena, which are associated with prolonged in vitro cultivation. The continuous metformin supplementation delayed/reduced the impairment of MSC functions as evidenced by the presence of specific pathways in metformin treated samples: i) the alpha-adrenergic signaling, which contributes to regulation of MSCs physiological secretory activity; the signaling pathway associated with MSCs detoxification activity; the aspartate degradation pathway for optimal energy production. The senomorphics function of metformin seemed related to its reactive oxygen species (ROS) scavenging activity. In metformin treated samples, the CEBPA, TP53 and USF1 transcription factors appeared implicated in the regulation of several factors (SOD1, SOD2, CAT, GLRX, GSTP1) involved in blocking ROS.
