Single-cell RNA-seq of the intestine sample of ICR mice treated with busulfan and/or Alginate Oligosaccharides

ABSTRACT: We profiled a single-cell RNA-seq on ICR mouse intestine treated with busulfan and or Alginate Oligosaccharides (AOS) from 3 weeks of age to 5 weeks of age (for 2 weeks) to study the rescue effect of AOS on the intestine development disrupted by busulfan. We collected single cell from intestine from the four groups AOS 0 (control), B+A 0 (busulfan alone), B+A 10 (busulfan plus AOS 10 mg/kg Body Weight). The the cells were further captured with 10x Genomics based single-cell system. By using single-cell RNA seq, we found busulfan damaged intestinal cells. AOS significantly improved the intestinal cell junction and microvilin formation. Besides, we provide candidate master regulons regulated by AOS. Our data here offers the opportunity for improving intestine under stress.
