Pancreatic β-cell specific Ets-1 inhibition treats type 2 diabetes via revitalized insulin production

Project description:As a transcription factor, Ets1 (E26 transformation-specific-1 transcription factor) plays an important role in cell growth, development, differentiation, etc. Our research found that it plays an crucial role in β cell dysfunction induced by metabolic stress such as high glucose. In order to further clarify the downstream target molecules of Ets1 in β cells and the potential mechanism of regulating the mRNA level of target molecules, we transfected Min6(Mouse insulinoma cells) with control virus (Ad-GFP) and Ets-1 overexpression adenovirus (Ad-Ets-1) respectively. Through immunoprecipitation of Ets1 antibody, the DNA fragment specifically bound to Ets1 was obtained and used in high-throughput sequencing. By comparing the enriched DNA fragment with mouse genome (mm10) DNA, the downstream genes that Ets1 can bind to under physiological conditions and the differential peak related genes after overexpression of Ets1 were obtained. Through GO analysis and KFGG analysis of the above related genes, it was confirmed that there was an obvious regulatory relationship between Ets1 and β cell function related genes. Project description:Fine mapping and validation of genes causing ? cell failure from susceptibility loci identified in type 2 diabetes genome-wide association studies (GWAS) poses a significant challenge. The VPS13C-C2CD4A-C2CD4B locus on chromosome 15 confers diabetes susceptibility in every ethnic group studied to date. However, the causative gene is unknown. FoxO1 is involved in the pathogenesis of ? cell dysfunction, but its link to human diabetes GWAS has not been explored. Here we generated a genome-wide map of FoxO1 superenhancers in chemically identified ? cells using 2-photon live-cell imaging to monitor FoxO1 localization. When parsed against human superenhancers and GWAS-derived diabetes susceptibility alleles, this map revealed a conserved superenhancer in C2CD4A, a gene encoding a ? cell/stomach-enriched nuclear protein of unknown function. Genetic ablation of C2cd4a in ? cells of mice phenocopied the metabolic abnormalities of human carriers of C2CD4A-linked polymorphisms, resulting in impaired insulin secretion during glucose tolerance tests as well as hyperglycemic clamps. C2CD4A regulates glycolytic genes, and notably represses key ? cell "disallowed" genes, such as lactate dehydrogenase A We propose that C2CD4A is a transcriptional coregulator of the glycolytic pathway whose dysfunction accounts for the diabetes susceptibility associated with the chromosome 15 GWAS locus.

miR-7a is regulated in beta-cell dysfunction and couples early and late stages of pancreatic beta-cell differentiation to insulin secretion

Project description:Transcriptional and posttranscriptional regulatory networks play a crucial role in the maintenance and adaptation of pancreatic beta-cell function. In this study we show that the levels of the prototypic neuroendocrine miRNA-7 are regulated in islets of obese, diabetic and aged mouse models. Using gain- and loss-of-function models we demonstrate that miR-7 regulates crucial members of the endocrine pancreatic transcriptional network controlling differentiation and insulin synthesis. Importantly, it also directly regulates key proteins in the insulin granule secretory machinery. These results reveal an interconnecting miR-7 genomic circuit that influences beta-cell differentiation, insulin synthesis and release and define a role for miR-7 as an endocrine checkpoint to stabilize beta-cell function during metabolic stress. These findings have implications for miR-7 inhibitors as potential therapies for type 2 diabetes and neurodegenerative diseases. Either miR-7a2 or miR-7b were over-expressed in MIN6 cells using an adenoviral vector. The miR-7a infection was performed in duplicates. In addition, a GFP over-expression in MIN6 using the same viral vector served as control. We also explored the consequence of miR-7a2 deletion in pancreatic beta-cells by generating a beta-cells specific miR-7a2 knock-out using the Lox/Cre system in a C57BL/6 background. We profiled gene expression in mutant and wild-type (control) islets.

Project description:Recent studies have identified dysregulation of RNA-binding proteins (RBPs) and aberrant mRNA splicing in the onset of diseases including diabetes. Here we investigated the role of RBFOX2 in the pancreatic β cell through the conditional mutation of Rbfox2 in the mouse pancreas (Pdx1:Cre; Rbfox2fl/lf) and RNAi experiments in the mouse insulinoma cell line, MIN6. We then identified the direct targets of RBFOX2 in the mouse β cell transcriptome my eCLIP-Seq in MIN6 cells.
