Primary Human B-cell Temporal Response to CD40L Stimulation

ABSTRACT: RNAseq was used to identify primary human B-cell responses to CD40L stimulation. Primary human peripheral blood B-cells were isolated by negative selection and then subjected to 0, 1 or 18 hours of mega-CD40L stimulation at 50 ng/ml.

RNA-seq analysis of neutrophils from CD40L-deficient patients and healthy controls, as well as HL-60 promielocytic cell line

Project description:We aim to characterize to effects of the absence of CD40L on neutrophil transcriptome and the effect of soluble CD40L on HL60 cells. For this purpose Total RNA of isolated neutrophils from three CD40L-deficient patients and three healthy controls as well as HL-60 cells from ATCC (HL-60 (ATCC CCL-240) were analyzed by RNAseq. Before RNA obtention, neutrophils were incubated for 2 hours in the presence or absence of 100 U/ml rhIFN- (Immukine, Boehringer Ingelheim), and HL-60 cells cultured for 6 days in the presence or absence of 500 ng/mL sCD40L and/or 1,0 % dimethyl sulfoxide (DMSO).

The T cell-DC CD40L-CD40 axis, but not platelet CD40L, drives atherosclerosis

Project description:Atherosclerosis, the underlying vascular cause of cardiovascular disease, has a strong inflammatory component. The co-stimulatory CD40-CD40 ligand (CD40L) signaling axis is a pivotal regulator of immune responses in atherosclerosis. However, therapeutic long-term inhibition of CD40L will severely compromise the immune system making it a non-viable treatment option. To circumvent this issue, cell-specific inhibition may present a better approach to target the CD40-CD40L axis. Therefore, we generated T cell and platelet-specific knockout mice for CD40L and apolipoprotein E, which were aged for 28 weeks to study their effects on immune status and atherosclerosis. Here, we show that T cell specific deficiency in CD40L signaling reduced plaque progression through hampered Th1 polarization as well as reduced antigen-dependent proliferation and oxLDL IgM production. DC-specific CD40 deficient mice displayed a similar phenotype. Platelet-specific CD40L deficiency, however, failed to decrease atherosclerosis, but ameliorated atherothrombosis. Together, our results illuminate the divergent cell-specific mechanisms of CD40-CD40L signaling in atherosclerosis, which may lead to advances in targeted therapies.

Project description:The aim of the present study was to clarify the role of IL-18 in mouse mast cell functions. Mouse mucosal mast cells (Balb/c) were differentiated in the presence of IL-3 (5 ng/ml), SCF (40 ng/ml), IL-9 (10 ng/ml) and human TGFbetaI (2 ng/ml) for >4 weeks. Cells were then stimulated by 100 ng/ml IL-18 for 2 hours to monitor the direct effect of IL-18. Treated samples were compared to the untreated ones in a paired experimental setting. Response of murine lymphatic endothelial cells to stimulation with vascular enothelial growth factor-C (VEGF-C)

Project description:The goal of this study is to investigate changes in gene expression profile of lymphatic endothelial cells (LECs) following stimulation with VEGF-C. Primary murine LECs from C57Bl/6 mice were expanded ex vivo from five individual mice (taken from digests of axillary, brachial, cervical, inguinal, popliteal lymph nodes), and stimulated with media only or supplemented with 10 or 100 ng/mL of murine VEGF-C (recombinant, expressed in HEK-293E cells) for 72 h (3 d). This timepoint was chosen because we had detected no significant changes in cell number or density in a daily check up to this timepoint, and ELISAs/Luminex assays detected no significant changes in most chemokine/cytokine biomarkers before 72 h. At this time, LECs were lysed and mRNA isolated for RNAseq analysis to determine differences in gene expression as a function of VEGF-C stimulation. Unstimulated, 10 ng/mL VEGFC-stimulated, and 100 ng/mL VEGFC-stimulated LEC transcriptomes (n = 5 per condition) are provided here.
