ABSTRACT: Y37A1B.5, a SELENBP1 ortholog in C. elegans modulates lifespan and stress resistance. Hence transcriptome profiling (RNA-seq) after depletion of Y37A1B.5 via RNAi was performed, aiming to identify targets that would explain these effects.

Project description:Human selenium-binding protein 1 (SELENBP1) was originally identified as a protein binding selenium, most likely as selenite. SELENBP1 is associated with cellular redox and thiol homeostasis in several respects, including its established role as a methanethiol oxidase that is involved in degradation of methanethiol, a methionine catabolite, generating hydrogen sulfide (H2S) and hydrogen peroxide (H2O2). As both H2S and reactive oxygen species (such as H2O2) are major regulators of Caenorhabditis elegans lifespan and stress resistance, we hypothesized that a SELENBP1 ortholog in C. elegans would likely be involved in regulating these aspects. Here we characterize Y37A1B.5, a putative selenium-binding protein 1 ortholog in C. elegans with 52% primary structure identity to human SELENBP1. While conferring resistance to toxic concentrations of selenite, Y37A1B.5 also attenuates resistance to oxidative stress and lowers C. elegans lifespan: knockdown of Y37A1B.5 using RNA interference resulted in an approx. 10% increase of C. elegans lifespan and an enhanced resistance against the redox cycler paraquat, as well as enhanced motility. Analyses of transgenic reporter strains suggest hypodermal expression and cytoplasmic localization of Y37A1B.5, whose expression decreases with worm age. We identify the transcriptional coregulator MDT-15 and transcription factor EGL-27 as regulators of Y37A1B.5 levels and show that the lifespan extending effect elicited by downregulation of Y37A1B.5 is independent of known MDT-15 interacting factors, such as DAF-16 and NHR-49. In summary, Y37A1B.5 is an ortholog of SELENBP1 that shortens C. elegans lifespan and lowers resistance against oxidative stress, while allowing for a better survival under toxic selenite concentrations.

Project description:Selenium-binding protein 1 (Selenbp1) is a 2,3,7,8-tetrechlorodibenzo-p-dioxin inducible protein whose function is yet to be comprehensively elucidated. As the highly homologous isoform, Selenbp2, is expressed at low levels in the kidney, it is worthwhile to compare wild-type C57BL mice and Selenbp1-deficient mice under dioxin-free conditions. In this study, we conducted a comprehensive analysis of Selenbp1-Knockout mice to elucidate the role of Selenbp1 in the kidneys of wild-type C57BL mice under non-dioxin-administered conditions. Data from DNA microarray analysis suggested that Selenbp1 is involved in lipid metabolism. The results of real-time RT-PCR confirmed that ablation of Selenbp1 alters lipid metabolism via the Ppara pathway.

Selenium uptake, tolerance and reduction in Flammulina velutipes supplied with selenite.

Project description:Recently, selenium (Se) enriched mushrooms have been exploited as dietary Se supplements, but our knowledge of the metabolic process during the Se enrichment process is far from complete. In this study, the uptake, tolerance and reduction of selenite in a widely cultivated mushroom, Flammulina velutipes, was investigated. The results showed that pH variation (from 5.5-7.5), metabolic inhibitor (0.1 mM 2,4-DNP) and P or S starvation led to 11-26% decreases in the selenite uptake rate of F. velutipes. This indicates that a minor portion of the selenite uptake was metabolism dependent, whereas a carrier-facilitated passive transport may be crucial. Growth inhibition of F. velutipes initiated at 0.1 mM selenite (11% decrease in the growth rate) and complete growth inhibition occurred at 3 mM selenite. A selenite concentration of 0.03-0.1 mM was recommended to maintain the balance between mycelium production and Se enrichment. F. velutipes was capable of reducing selenite to elemental Se [Se(0)] including Se(0) nanoparticles, possibly as a detoxification mechanism. This process depended on both selenite concentration and metabolism activity. Overall, the data obtained provided some basic information for the cultivation of the selenized F. velutipes, and highlighted the opportunity of using mushrooms for the production of Se(0) nanoparticles.

Selenium nanovirus and its cytotoxicity in selenite-exposed higher living organisms.

Project description:Selenium (Se) is an essential micronutrient in living organisms, having a narrow margin between essential and potentially toxic intake/exposure. Thus, the biochemistry of Se in living organisms must be studied in-depth to determine the underlying mechanism of Se cytotoxicity. In this study, we report the emergence of selenium nanovirus (SeNVs) in selenite-exposed fish (freshwater and saltwater) and plants (dryland) and its toxicity in them. SeNVs were found in both the abdomen and tail of Oryzias melastigma and saltwater Rhodeus ocellatus, which led to their death. The occurrence of the intracellular assembly of SeNVs was observed in the roots and leaves of corn Zea mays, but not in those of Limnobium laevigatum. SeNVs led to the death of Z. mays but caused chronic toxicity in L. laevigatum. SeNVs should be a system or structure that dissipates the intracellular redox gradients of the host cells, with simple information consisting Se-O, Se-N, or Se-S bond, that would ensure elemental Se ligand binding with nearly specific biomolecules in host cells, thereby maintaining their composition and stabilizing their structure. The multiple toxic effects of Se, therefore, could be the consequence of increase of entropy in the host cells caused by the intracellular assembly of SeNVs. This study may provide an insight into the underlying mechanism of Se in environmental toxicology and its applications in human health.

Highly Selenite-Tolerant Strain <i>Proteus mirabilis</i> QZB-2 Rapidly Reduces Selenite to Selenium Nanoparticles in the Cell Membrane.

Project description:The application of biosynthesized nano-selenium fertilizers to crops can improve their nutrient levels by increasing their selenium content. However, microorganisms with a high selenite tolerance and rapid reduction rate accompanied with the production of selenium nanoparticles (SeNPs) at the same time have seldom been reported. In this study, a bacterial strain showing high selenite resistance (up to 300 mM) was isolated from a lateritic red soil and identified as Proteus mirabilis QZB-2. This strain reduced nearly 100% of 1.0 and 2.0 mM selenite within 12 and 18 h, respectively, to produce SeNPs. QZB-2 isolate reduced SeO3 2 - to Se0 in the cell membrane with NADPH or NADH as electron donors. Se0 was then released outside of the cell, where it formed spherical SeNPs with an average hydrodynamic diameter of 152.0 ± 10.2 nm. P. mirabilis QZB-2 could be used for SeNPs synthesis owing to its simultaneously high SeO3 2 - tolerance and rapid reduction rate.

A human model of selenium that integrates metabolism from selenite and selenomethionine.

Project description:Selenium (Se) metabolism is affected by its chemical form in foods and by its incorporation (specific vs. nonspecific) into multiple proteins. Modeling Se kinetics may clarify the impact of form on metabolism. Although the kinetics of Se forms have been compared in different participants, or the same participants at different times, direct comparisons of their respective metabolism in the same participants have not been made. The aim of this study was to simultaneously compare kinetics of absorbed Se from inorganic selenite (Sel) and organic selenomethionine (SeMet) in healthy participants (n = 31). After oral administration of stable isotopic tracers of each form, urine and feces were collected for 12 d and blood was sampled over 4 mo. Tracer enrichment was determined by isotope-dilution-GC-MS. Using WinSAAM, a compartmental model was fitted to the data. Within 30 min of ingestion, Se from both forms entered a common pool, and metabolism was similar for several days before diverging. Slowly turning-over pools were required in tissues and plasma for Se derived from SeMet to account for its 3-times-higher incorporation into RBC compared with Se from Sel; these presumably represent nonspecific incorporation of SeMet into proteins. Pool sizes and transport rates were determined and compared by form and gender. The final model consisted of 11 plasma pools, 2 pools and a delay in RBC, and extravascular pools for recycling of Se back into plasma. This model will be used to evaluate changes in Se metabolism following long-term (2 y) Se supplementation.
