Project description:Plants regulate growth and development in different ambient environments through light signaling pathways and heat stress signaling pathways, both of which are very important for plant adaptation to the environment, but whether there is an interaction between the two is still unclear. In this article, we reported that the blue light receptor Cryptochrome CRY1 phosphorylation will be inhibited under high temperature stress, and CRY1 activity will decrease; at this time, CRY1 could not interact with E3 ubiquitin ligase COP1(CONSTITUTIVELY PHOTOMORPHOGENIC 1) and COP1 activity increased. Its substrate HY5(ELONGATED HYPOCOTYL 5）is continuously degraded under heat stress. Through further transcriptome analysis, we found that the substrates of HY5 have a class of HSFAs（HEAT SHOCK FACTOR）HSFA2/HSFA7As/HSFA7Bs transcription factors, which are rapidly induced under heat stress conditions and activate downstream HSPs (HEAT SHOCK PROTEIN)expression.Under normal conditions, HY5 protein inhibits HSFA2/HSFA7As/HSFA7Bs. Under heat stress, HY5 protein is degraded, which releases the expression of HSFAs, thereby enhancing plant heat tolerance. In this paper, we put forward the hypothesis that the phosphorylation of CRY1 seems to be a switch in the ambient environment , and through the regulation of phosphorylation level, the light and heat stress signal are transmitted downward, forming an light-heat signal interactive regulation pathway to regulate the growth and development of plants.

Project description:Plants transcriptome react to environment temperture changes profoundly. In Arabidopsis seedlings, genes response to temperature fluctuations to adopt the ever-changing ambient envrionment. We used microarrays to detail the global programme of gene expression underlying heat stress response progress in Arabidopsis. Ten-day-old Arabidopsis seedlings were selected for RNA extraction and hybridization on Affymetrix microarrays. We sought to explore the heat stress response in transcriptome, thus we treat the plants with heat stress. While in order to identify the interaction between light and temperature signaling pathways in plant , we treat Arabidopsis with heat stress under both light and dark conditions. To that end, our plant tissues are grouped as: HS-LIGHT, HS-DARK,CONTROL-LIGHT,CONTROL-DARK.

Project description:To understand plant adaptation to heat stress, gene expression profiles of Arabidopsis leaves under heat stress, during recovery and control condition were obtained using microarray. Microarray data listed responsible candidate genes for glycerolipid metabolism. Arabidopsis thaliana ecotype Columbia (Col-0) seeds were surface-sterilised and sown on an agar-solidified Murashige and Skoog medium. Plants were grown at 22ºC under a 16-h-light/8-h-dark cycle. Vegetative plants were subjected to high temperature for a day, then continued to be grown under normal condition for a day.

Project description:Adequate reprogramming of global translation under stress is of critical importance to cell survival in eukaryotes. The phosphorylation of eukaryotic initiation factor-2α is a major pathway for stress-induced translational arrest in animals. Here we report that eIF2α phosphorylation is not induced by heat in Arabidopsis. Instead, we identify an uncharacterized protein, FUST1, that can directly sense heat and initiate translational shutdown in Arabidopsis. FUST1 exhibits heat-dependent condensation both in vivo and in vitro, which is mainly driven by its prion-like domain (PrLD). Molecular dynamic simulation reveals that PrLD undergoes conformational rearrangements and engages more inter-amino acid interactions as temperature increases. Mutations that block this conformational change also diminish FUST1 condensation in vitro and in vivo and impair heat tolerance. FUST1 condensates preferentially partition mRNAs of greater length via electrostatic interactions, recruit translation repressors and RNA decapping and deadenylation factors. Importantly, disruption of FUST1 condensation dramatically compromises translational arrest under heat. As a result, FUST1 condensation precedes and is necessary for the assembly of heat stress granules. These findings thus uncover FUST1 as a molecular switch for translation under heat stress and shed light on engineering of heat-adaptable crops.

Project description:Purpose: Acclimation referes to reversible responses that plants trigger under frequently and moderate changes in natural environments, mostly light intensity and temperature. To better understand acclimation in Arabidopsis thaliana, modulators and integrators for this process should be identified. To achieve this goal, a dynamic set-up was designed to systematically cultivate Arabidopsis plants under moderate high light, heat and cold and a multiple high-throughput strategy was conducted to monitor changes in transcriptome, proteome and metabolome. As part of this comprehensive characterisation, lncRNA-Seq has been carried out to investigate transcriptome changes and further integrated in conditional correlation networks. Methods: lncRNA-Seq libraries were prepared from 14-d-old plants cultivated under standard control conditions or acclimated to high light, cold or heat for 4 d and then de-acclimated under normal conditions for 4 additional days. Standard Illumina protocols were used for lncRNA-Seq sequencing. Unless some quality issue in libraries was detected, triplicates for 3 independent plants were used. Adaptor sequences were removed with Trimmomatic and resulting reads mapped to the Arabidopsis genome (Araport11) with Tophat 2.1.1. FPKM reads were quantified with Cufflinks to provide processed data files and differential expression analysis were conducted with featureCounts/DEseq2 pipeline. Results: lncRNA-Seq libraries contained 10 milion reads each. Transcript analysis for differential gene expression was conducted according to the Tophat/featureCounts/DESeq2 pipeline. To declare significant differences, a cut-off for absolute log2(FC) ≥ 1 compared to the corresponding control and FDR ≤ 0.05 was applied. Transcripts significantly different in at least one time-point were considered for subsequent analysis. Accordingly, high light treatments caused changes in 2683 transcripts, followed by 3470 under heat and 6882 under cold. Further data analysis revealed that more than 12% of all altered transcripts were commonly miss-regulated among the three treatments. From a dynamic point of view, transcriptional changes occurred from 3h to 4d of treatment in all treatments and tend to recover to the initial state after 2 and 4d of de-acclimation. Conclusions: the reversibility of the transcriptional changes during the recovery phase validated the conditions of our set-up to investigate acclimation. Moreover, the existance of a common fraction of transcripts that responded to the three conditions unveiled a central response to acclimation.

Maize (Zea mays cv. B73) were grown in plastic pots filled with nutrient soil for 2 weeks with a 12 h light (28°C)/12 h dark (25°C) regimen (ca. 1500 μmol photons mâ2 sâ1) and were treated for 30 min at 42°C.
