Circadian modulation of genome-wide RXR binding in mouse liver tissue

ABSTRACT: RXR DNA-binding sites cluster together and toward promoter-riches region on chromatin landscape and boost gene expression.The RXR signal is highly correlated with Pol2 signal and even mimic the Pol-II profile upon some high expressed genes. Majority of RXR peaks are constant binding. Only 10% of RXR peak showed circadian rhythm, and only 10% of them associated with circadian expressed genes. The genes with and without RXR at their promoter exhibit different phases of Pol2 signal. And RXR is not the main driver for the circadian transcriptional output. Pathways analysis for target genes showed distinct pattern for genes with unique binding motif, they mainly involve in general fundamental pathways like cell cycle and cell check-point. While the genes with multiple binding motifs are involve in specific pathways like circadian and liver metabolism pathways.
