Next generation sequencing facilitates quantitative analysis of transcriptional profiling of Candida albicans wild type and cys4 mutant strains

ABSTRACT: mRNA profiles of Candida albicans wild-type (CAF2-1),cys4 mutant and CYS4 integrated strains were generated by deep sequencing, in triplicate, using Illumina HiSeq X Ten. The sequence reads that passed quality filters were analyzed at the transcript isoform level. qRT–PCR validation was performed using SYBR Green assay. We find that filament or biofilm-specific regulators such as UME6, TEC1 and BRG1 and adhesins such as hypha-specific genes ECE1 and ALS3 were downregulated and negative hyphae regulator NRG1 was upregulated in cys4 mutant. Genes invovled in cell wall synthesis and oxidative stress response were also accordingly regulated in cys4 mutant.
