Identification of novel genes involved in postnatal longitudinal muscle growth

ABSTRACT: Overall goal: To identify genes involved in postnatal longitudinal muscle growth through misregulation by neonatal brachial plexus injury leading to contracture formation. Purpose of analysis: To generate a transcriptional profile of mice given unilateral neonatal brachial plexus injuries. Experimental structure: The transcriptional profiles generated from the RNA-Seq analysis would be compared as neonatally-denervated biceps muscle versus the contralateral control to identify genes regulating postnatal longitudinal muscle growth.

Project description:Neuropathic pain is an apparently spontaneous experience triggered by abnormal physiology of the peripheral or central nervous system, which evolves with time. Neuropathic pain arising from peripheral nerve injury is characterized by a combination of spontaneous pain, hyperalgesia and allodynia. There is no evidence of this type of pain in human infants or rat pups; brachial plexus avulsion, which causes intense neuropathic pain in adults, is not painful when the injury is sustained at birth. Since infants are capable of nociception from before birth and display both acute and chronic inflammatory pain behaviour from an early neonatal age, it appears that the mechanisms underlying neuropathic pain are differentially regulated over a prolonged postnatal period. We used microarrays to detail the global programme of gene expression underlying the differences in nerve injury between along the postnatal development and identified distinct classes of regulated genes during the injury

Neonatal Genistein Exposure Causes Implantation Failure by Disrupting Uterine Developmental Patterning and Endometrial Gland Formation

Project description:Female mice exposed neonatally to the phytoestrogen genistein (GEN) at doses similar to those in infants consuming soy-based infant formulas are infertile due in part to uterine implantation defects. To determine the mechanisms by which neonatal GEN exposure causes implantation defects, female mice were exposed to vehicle (corn oil) or GEN in corn oil (50 mg/kg/day) on postnatal days (PND) 1-5. Uterine tissues were collected during early gestation for microarray analysis; gestation day 1.5 (GD1.5), GD3.5, GD4.5 and GD5.5. We conclude that neonatal GEN exposure disrupts expression of genes important for uterine development, causing posteriorization and diminished gland function during pregnancy that together cause implantation failure.

Transcription profiling of rat gastrocneumius muscle following short and long term denervation

Project description:Effect of "Sensory Protection" on the transcriptome of Gastrocnemius muscle denervated by Tibial nerve transection in the rat. Comparison is made between the muscle transcriptome in denervated muscle (D), muscle undergoing surgical repair with a motor nerve (immediate repair, or IR) and muscle undergoing surgical repair with a pure sensory nerve (sensory protection, or SP). There are 6 cohorts of rats with 4 to 6 rats in each cohort:<br><br>1 Month Denervated, 1 Month Immediate Repair, 1 Month Sensory Protection, 3 Month Denervated, 3 Month Immediate Repair, 3 Month Sensory Protection. <br><br>For each animal RNA was extracted from both the experimental operated (right) gastrocnemius muscle and the contralateral control unoperated (left) gastrocnemius. Samples were run in duplicate with fluorophor reversal experiments (i.e experimental limb RNA labelled with Cy3 and control limb RNA labelled with Cy5 and vice versa) to control for possible unequal incorporation of the dyes in the reverse transcription reaction.

Expression data from mouse neonatal hindlimb muscles

Project description:During neonatal development, skeletal muscle grows dramatically by myonuclei accretion to existing fibers and hypertophic growth of fibers with protein synthesis. To understand molecular mechanism underlying neonatal muscle growth, we used microarray to profile the global program of gene expressions especially involved in myoblast fusion, migration, and muscle fiber growth by itself. Mouse hindlimb muscles at postnatal day (P) 2, 7, and 14 were isolated and used for RNA extraction and hybridyzation on microarrays. We used three biological replicates for each time point.

A Dach2-Hdac9-Myog-Gdf5 signaling system regulates regeneration of neuromuscular synapses

Project description:Muscle denervation due to injury, disease or aging results in impaired motor function. Restoring neuromuscular communication requires axonal regrowth and regeneration of neuromuscular synapses. Muscle activity inhibits neuromuscular synapse regeneration. The mechanism by which muscle activity regulates regeneration of synapses is poorly understood. Dach2 and Hdac9 are activity-regulated transcriptional co-repressors that are highly expressed in innervated muscle and suppressed following muscle denervation. Here, we report that Dach2 and Hdac9 inhibit regeneration of neuromuscular synapses. Importantly, we identified Myog and Gdf5 as muscle-specific Dach2/Hdac9-regulated genes that stimulate neuromuscular regeneration in denervated muscle. Interestingly, Gdf5 also stimulates presynaptic differentiation and inhibits branching of regenerating neurons. Finally, we found that Dach2 and Hdac9 suppress miR206 expression, a microRNA involved in enhancing neuromuscular regeneration. RNAseq on innervated and 3 day denervated adult soleus muscle from wildtype mice is compared with that from 3 day denervated soleus muscle from Dach2/Hdac9 deleted mice to identify Dach2/Hdac9-regulated genes.
