NPA-induced vascular overgrowth in Arabidopsis

Project description:We used N-(1-naphthyl) phthalamic acid (NPA)-induced vascular overgrowth in Arabidopsis leaves to look for differential up-regulation of genes in NPA-treated tissues that may be involved in vascular differentiation. Arabidopsis thaliana Col-0 plants were grown for approximately 2 weeks on solid ATS medium (1) containing a final concentration of 10 um NPA (dissolved in DMSO) or an equivalent volume of DMSO (control). At this stage plants had approximately 6 rosette leaves. RNA was prepared from entire shoot tissues of control (DMSO) or NPA-treated plants.(1) Lincoln et al., 1990. Plant Cell 2: 1071-1080.

Project description:Phosphonate related fungicides such as neutralized phosphorous acid (NPA) are effective for the control of plant diseases, especially those caused by Phytophthora and other oomycetes. It has been suggested that phosphonate may induce plant resistance, however, the mechanism is not clear thus far. We performed microarray analysis using Affymetrix Tomato Genome Array to identify tomato genes that are differentially expressed in response to NPA treatment.

Dwarf Ad libitum

Project description:Female Ames dwarf mouse (df/df) with ad libitum access to water and standard pelleted food (LabDiet, PMI Feeds, Inc., St Louis, MO) caged with microisolator filter tops. Mice were killed between 24 to 26 months of age, tissues removed, rapidly frozen on dry ice, and stored in liquid nitrogen. Total liver RNA was isolated from frozen tissue as described (T. Tsuchiya, J.M. Dhahbi, X. Cui, P.L. Mote, A. Bartke, S.R. Spindler, Physiological Genomics, Submitted). mRNA levels were measured using the Affymetrix mouse U74Av2 array according to standard protocols. After hybridization, arrays were scanned using a Hewlett-Packard GeneArray Scanner. Image analysis was performed as described (Cao SX, Dhahbi JM, Mote PL, and Spindler SR. Transcriptomic analysis of liver in sex-linked Dwarf and wild type chickens

Project description:The sex-linked Dwarf (SLD) recessive mutation is characterized by a significant reduction of body size in hemizygous females (30%) and homozygous males (40%). The SLD chickens of the Leghorn line are affected by a Growth Hormone (GH) resistance condition due to a missense mutation at the Growth Hormone Receptor (GHR) gene causing a loss of function of the GHR. The sex-linked recessive dwarf mutation has large commercial use in maternal lines of the broiler poultry industry since these hens require less food and less housing space. The SLD chickens are also an interesting model for investigating complex traits such as energy expenditure and heat resistance for which the SLD chickens differ compared to normal size ones. In this study we performed a whole genome microarray expression profiling of liver using a 8x60K custom Agilent microarray. In total we analyzed 24 SLD and 24 wild type birds of 12 weeks of age obtained by crossing a heterozygous “dw” Leghorn cock with 12 females of an inbred Leghorn line to obtain wild type and dwarf sib and half-sib hens. The aim of the study was the fine identification of GH-inducible genes, and the characterization of the GH dependent signalling pathways.
