Project description:Willow (Salix), a dioecious plant, is an important ornamental tree species in the world. Salix linearistipularis, a perennial woody plant species naturally distributed on the Songnen Plain saline-alkali land in northeast China, has a high saline condition. To study the sexual differences of S. linearistipularis in salinity tolerance, the physiological and transcriptional responses to salinity were compared between female and male cuttings. Under salinity stress, the female leaves exhibited higher superoxide dismutase and peroxidase activities and photosynthetic capacity, and lower H2O2 contents than those of male leaves. Under salinity stress, sodium (Na+) accumulation in female leaves was lower than that in the male leaves. The non-invasive micro-test showed that the net Na+ efflux in the salt-treated female roots was higher than that in male roots. Physiological responses revealed that female cuttings were more tolerant than males, which may be mainly due to females having lower leaf Na+ accumulation and higher root Na+ efflux capacity than males. Transcriptional analyses showed that 108 differentially expressed salt-responsive genes were identified in both female and male roots; most of these showed sexual differences in expression patterns under salinity stress. RNA-seq combined with qPCR analysis showed that the salt-induced expression of four Na+/H+ antiporter (NHX) genes (SlNHX3, 5, 6, 7) in female roots was higher than that in male roots. Transcriptional analyses revealed that the higher Na+ efflux capacity in female roots than in male roots may be closely related to the differential expression of salt-responsive genes, especially NHX genes.

We found that several functional groups of genes involved in important pathways were differentially expressed, including photosynthesis-related genes which were mainly up-regulated in males but down-regulated in females. This gene expression pattern is consistent with physiological observation that salinity inhibited photosynthetic capacity more in females than in males. In conclusion, our study provided molecular evidence of sexual differences in poplar salinity tolerance. Identified sex-related genes in salinity tolerance and their functional groups will enhance our understanding of sexual differences to salinity stress at the transcription level.

Project description:Abiotic stress greatly inhibits crop growth and reduces yields. However, little is known about the transcriptomic changes that occur in the industrial oilseed crop, rapeseed (Brassica napus), in response to abiotic stress. In this study, we examined the physiological and transcriptional responses of rapeseed to drought (simulated by treatment with 15% (w/v) polyethylene glycol (PEG) 6000) and salinity (150 mM NaCl) stress. Proline contents in young seedlings greatly increased under both conditions after 3 h of treatment, whereas the levels of antioxidant enzymes remained unchanged. We assembled transcripts from the leaves and roots of rapeseed and performed BLASTN searches against the rapeseed genome database for the first time. Gene ontology analysis indicated that DEGs involved in catalytic activity, metabolic process, and response to stimulus were highly enriched. The Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis revealed that differentially expressed genes (DEGs) from the categories metabolic pathways and biosynthesis of secondary metabolites were highly enriched. We determined that myeloblastosis (MYB), NAM/ATAF1-2/CUC2 (NAC), and APETALA2/ethylene-responsive element binding proteins (AP2-EREBP) transcription factors function as major switches that control downstream gene expression and that proline plays a role under short-term abiotic stress treatment due to increased expression of synthesis and decreased expression of degradation. Furthermore, many common genes function in the response to both types of stress in this rapeseed.

Project description:Finger millet is an important cereal that is grown in semi-arid and arid regions of East-Africa. Salinity stress is a major environmental impediment for the crop growth and production. This study aimed to understand the physiological and biochemical responses to salinity stress of six Kenyan finger millet varieties (GBK043137, GBK043128, GBK043124, GBK043122, GBK043094, GBK043050) grown across different agroecological zones under NaCl-induced salinity stress (100, 200 and 300 mM NaCl). Seeds were germinated on the sterile soil and treated using various concentrations of NaCl for 2 weeks. Early-seedling stage of germinated plants were irrigated with the same salt concentrations for 60 days. The results indicated depression in germination percentage, shoot and root growth rate, leaf relative water content, chlorophyll content, leaf K+ concentration, and leaf K+/Na+ ratios with increased salt levels and the degree of increment differed among the varieties. On the contrary, the content of proline, malonaldehyde, leaf total proteins, and reduced sugar increased with increasing salinity. At the same time, the leaf Na+ and Cl- amounts of all plants increased substantially with increasing stress levels. Clustering analysis placed GBK043094 and GBK043137 together and these varieties were identified as salt-tolerant based on their performance. Taken together, our findings indicated a significant varietal variability for most of the parameters analysed. The superior varieties identified could be used as promising genetic resources in future breeding programmes directed towards development of salt-tolerant finger millet hybrids. Further analysis at genomic level needs to be undertaken to better understand the genetic factors that promote salinity tolerance in finger millet.

Project description:BackgroundBecause soil salinity is a major abiotic constraint affecting crop yield, much research has been conducted to develop plants with improved salinity tolerance. Salinity stress impacts many aspects of a plant's physiology, making it difficult to study in toto Instead, it is more tractable to dissect the plant's response into traits that are hypothesized to be involved in the overall tolerance of the plant to salinity.Scope and conclusionsWe discuss how to quantify the impact of salinity on different traits, such as relative growth rate, water relations, transpiration, transpiration use efficiency, ionic relations, photosynthesis, senescence, yield and yield components. We also suggest some guidelines to assist with the selection of appropriate experimental systems, imposition of salinity stress, and obtaining and analysing relevant physiological data using appropriate indices. We illustrate how these indices can be used to identify relationships amongst the proposed traits to identify which traits are the most important contributors to salinity tolerance. Salinity tolerance is complex and involves many genes, but progress has been made in studying the mechanisms underlying a plant's response to salinity. Nevertheless, several previous studies on salinity tolerance could have benefited from improved experimental design. We hope that this paper will provide pertinent information to researchers on performing proficient assays and interpreting results from salinity tolerance experiments.

Experiment Overall Design: Seedlings were cultured in sand and irrigated with a nutrient solution for 22 d (salt-treated) and 30 d (control) after germination, respectively. Salinity treatment was applied by adding NaCl and CaCl2 (5:1 molar concentration) in two steps over a period of 3 days (final electrical conductivity: 7.4 dS m-1) to prevent osmotic shock. All plants were harvested on day 30.

Project description:In view of the continuous salinization of arable lands world-wide, there is an urgent need to better understand the mechanisms underlying plant responses to salt stress at different stages of their development. We investigated the role of calmodulin (CaM)-binding transcription activator 6 (CAMTA6) under salinity stress during early germination in Arabidopsis. These analyses suggest that ABA signaling is involved in CAMTA6-dependent salt-responsive gene expression, consistent with the ABA hyper-tolerance phenotype and the lack of HKT1 response to ABA and NaCl in the camta6 mutants.
