Investigation of molecular events underlying the miR192-mediated modulation of inflammatory response

ABSTRACT: To understand the molecular mechanism underlying the miR192-mediated modulation of inflammatory response, we have employed whole genome microarray expression profiling of bone-marrow derived macrophages transfected with miR192. In order to distingish the effect of miR192 on inflammatory response from the effecct of IL-6, RNA was isolated from miR192-transfected and R848-stimulated macrophages in the presence or absence of anti-IL-6 Ab during the culture.

Project description:To understand the molecular mechanism underlying the IL-6-mediated attenuation of Th1 differentiation in tumor-bearing animals, we have employed whole genome microarray expression profiling of CD4+ T cells isolated from tumor-bearing mice. In order to distingish the effect of IL-6/sIL-6R signaling augmented in tumor-bearing mice from the effecct of other tumor-derived signals, we isolated CD4+ T cells that were primed in tumor free-mice and in the mice inoculated with ovalbumin (OVA)-expressing MCA205 fibrosamcoma, in conjunction of anti-IL-6R Ab injection. We found that Th2-like gene signature such as Il4, IL10, Ccr4, and c-maf was up-regulated in CD4+ T cells primed in tumor-bearing mice. However,the expression of other Th2 master regulator Gata3 gene, or Tfh-related genes (Bcl6, Cxcr5, and Batf) was not substantially augmented in tumor-bearing mice as compared with those in tumor-free mice. These expressions were abrogated by IL-6R blockade, suggesting that the expression of these Th2-like gene set and the other IL-6 signal-dependent genes like Klf1, Bcl11a, and Klf4 are candidate regulators responsible for IL-6 signaling-mediated Th1 inhibition in tumor-bearing animals.

Itaconate Negatively Regulates Innate Signaling through Modification of IRAK4, Degradation of NFκB, and the Modulation of Global Ubiquitination

Project description:A wide variety of electrophilic derivatives of the Kreb’s cycle-derived metabolite, itaconate, are immunomodulatory, yet these derivatives have overlapping and sometimes contradictory activities. Therefore, we generated a genetic system to interrogate the immunomodulatory functions of endogenously produced itaconate in human macrophages. Endogenous itaconate is driven by multiple innate signals restraining inflammatory cytokine production. Endogenous itaconate directly targets cysteine 13 in IRAK4 disrupting IRAK4 autophosphorylation and activation, drives the degradation of NFκB, and modulates global ubiquitination patterns. As a result, cells unable to make itaconate overproduce inflammatory cytokines such as TNFα, IL6, and IL-1β in response to these innate activators. In contrast, the production of IFNβ, downstream of LPS, requires the production of itaconate. These data demonstrate that itaconate is a critical arbiter of inflammatory cytokine production downstream of multiple innate signaling pathways laying the groundwork for the development of itaconate mimetics for the treatment of autoimmunity.

ZC3H13 and WTAP KO in hMDMs multi stimuli(NGS5421)

Project description:Here, we revealed an immunosuppressive role of m6A mediated by ZC3H13 and WTAP on TNF-driven inflammatory phenotype in human macrophages. In the following experiment we would like to investigate if the negative regulatory role of m6A is consistent across multiple stimuli using innate ligands (LPS, R848 and polyIC). We generated ZC3H13 KOs, WTAP KOs and NTC (WT) in monocyte-derived macrophages from 4 individual human donors and stimulated cells over night with LPS (5ng/ml) , R848 (50ng/ml) , polyIC (50ng/ml) and unstimulated cells as control. This yielded a total of 48 total samples.

Peripheral blood transcriptomics following immunization with TLR-adjuvanted nanoparticle vaccine in rhesus macaques

Project description:Transmission-blocking vaccines (TBVs) aim to disrupt malaria parasite development by targeting antigens expressed in the mosquito midgut and are considered an integral element of malaria eradication efforts. Here, we used non-human primates to demonstrate that immune responses to Pfs25, the leading TBV candidate, can be improved using unconjugated Pfs25 encapsulated in PLGA-based synthetic vaccine particles (SVP[Pfs25]) delivered subcutaneously, compared to the multimeric form, Pfs25-EPA administered intramuscularly. Pfs25-EPA given with the toll-like receptor (TLR)-based adjuvant GLA-LSQ induced robust Ab responses, but SVP[Pfs25] adjuvanted with GLA-LSQ or SVPs containing CpG or R848 increased Ab titers, Pfs25-specific plasmablasts, circulating memory B cells, and plasma cells in the bone marrow. Animals receiving R848 or CpG showed particularly strong type I IFN polarized innate activation, which correlated with increased Ab half-life. Only SVP[Pfs25] induced detectable Pfs25-specific CD4 Th1 and Tfh cells, correlating with improved Ab avidity. Collectively, multiple aspects of the responses elicited to Pfs25 indicate that the nanoparticle technology combined with TLR agonists is a promising platform for TBVs.

Modulation of macrophage inflammatory function through selective inhibition of the epigenetic reader protein SP140

Project description:SP140 is a bromodomain-containing protein expressed predominantly in immune cells. Genetic polymorphisms and epigenetic modifications in the SP140 locus have been linked to CrohnÃ¢Â€Â™s disease (CD), suggesting a role in inflammation. We report the development of the first small molecule SP140 inhibitor (GSK761) and utilize this to elucidate SP140 function in macrophages. We show that SP140 is highly expressed in CD68+ CD mucosal macrophages and in in vitro-generated inflammatory macrophages. SP140 inhibition through GSK761 reduced monocyte differentiation into inflammatory macrophages and lipopolysaccharide (LPS)-induced inflammatory activation. SP140 preferentially occupies transcriptional start sites (TSS) in inflammatory macrophages, with enrichment at gene loci encoding pro-inflammatory cytokines/chemokines and inflammatory pathways. GSK761 specifically reduced SP140 binding and thereby expression of SP140-bound genes. GSK761 inhibited the spontaneous expression of cytokines, including TNF, by CD14+ macrophages isolated from CD intestinal-mucosa. This study identifies SP140 as a druggable epigenetic therapeutic target for CD.

Modulation of transcriptional and inflammatory responses in murine macrophages by the Mycobacterium tuberculosis mammalian cell entry (Mce) 1 complex.

Project description:The outcome of many infections depends on the initial interactions between agent and host. Aiming at elucidating the effect of the M. tuberculosis Mce1 protein complex on host transcriptional and immunological responses to infection with M. tuberculosis, RNA from murine macrophages at 15, 30, 60 min, 4 and 10 hrs post-infection with M. tuberculosis H37Rv or mce1 H37Rv was analyzed by whole-genome microarrays and RT-QPCR. Immunological responses were measured using a 23-plex cytokine assay. Compared to uninfected controls, 524 versus 64 genes were up-regulated by 15min post H37Rv- and mce1 H37Rv-infection, respectively. By 15min post-H37Rv infection, a decline of 17 cytokines combined with up-regulation of Ccl24 (26.5-fold), Clec4a2 (23.2-fold) and Ppar (10.5-fold) indicated an anti-inflammatory response initiated by IL-13. Down-regulation of Il13ra1 combined with up-regulation of Il12b (30.2-fold), suggested switch to a pro-inflammatory response by 4hrs post H37Rv-infection. Whereas no significant change in cytokine concentration or transcription was observed during the first hour post mce1 H37Rv-infection, a significant decline of IL-1b, IL-9, IL-13, Eotaxin and GM-CSF combined with increased transcription of Il12b (25.1-fold) and Inb1 (17.9-fold) by 4hrs, indicated a pro-inflammatory response. The balance between pro-and anti-inflammatory responses during the early stages of infection may have significant bearing on outcome.

Investigation of the mechansim underlying the inflammatory phenotype in Group A ependymoma

Project description:Inflammatory response has been identified as a molecular signature of high-risk Group A ependymoma (EPN). To better understand the biology of this phenotype and aid therapeutic development, transcriptomic data from Group A and B EPN patient tumor samples, and additional malignant and normal brain data, were analyzed to identify the mechanism underlying EPN group A inflammation. Gene expression profiles of Group A and B EPN were contrasted to identify inflammatory transcriptional profiles (GSEA analysis). Candidate inflammatory mechanism genes were examined across a broader cohort of pediatric and adult brain tumor types and normal brain. Gene expression profiles were generated from surgical tumor and normal brain samples (n=149) using Affymetrix HG-U133plus2 chips (Platform GPL570).
