Bazedoxifene and conjugated estrogen combination improves gut-liver axis health [mouse liver]

Project description:Bazedoxifene and conjugated estrogens (CE+BZA) combination has been shown to prevent visceral adiposity and weight gain after menopause. However, its interaction with the microbiota has yet to be examined. In the present study, we use several –omics technologies to characterize the effects of various estrogens on the health of gut-liver axis. As reported in previous studies, CE+BZA combination is very effective at preventing ovariectomy-induced weight gain in mice fed a high-fat diet. Additionally, CE+BZA induces unique liver transcriptomic and blood metabolite profiles compared to estradiol, conjugated estrogens alone, and bazedoxifene alone. Several pathways and metabolites influenced are associated with lower rates of inflammation and overall benefits to gut and liver health. Finally, microbiome analysis shows that several bacterial species that potentially metabolize estrogens and affect their half-life in the body were significantly changed in CE+BZA treated mice. Our findings indicate a possible link between certain estrogens and gut microbiome and suggest a metabolic benefit of estrogens through manipulation of the gut-liver axis.

On the Strength of a 316L-Type Stainless Steel Subjected to Cold or Warm Rolling Followed by Annealing.

Project description:The ultrafine-grained microstructures and their effect on the yield strength of a 316L-type austenitic stainless steel processed by large strain cold/warm rolling and subsequent annealing were studied. A kind of continuous recrystallization developed during annealing, resulting in the evolution of uniform ultrafine-grained microstructures with relatively high residual dislocation densities. The development of such microstructure at 973 K led to excellent combination of tensile properties including high yield strength (?0.2 > 900 MPa) and satisfactory plasticity (? > 15%). A unique power law function between the annealed grain size and the dislocation density with a dislocation density exponent of -0.5 was obtained for these continuously recrystallized microstructures. A physically justified explanation of the observed structural/substructural strengthening is introduced.

E-cadherin regulates female tumor aggressiveness by controlling estrogen-sensitivity [ChIP-seq]

Project description:Sex disparities in cancer have been documented, but understanding of the underlying mechanisms is still limited, restricting precision medicine and the discovery of new therapeutic options. By analyzing overall cancer frequency, we found that women, between puberty and menopause, are more prone to develop cancer than men. We identified the cell adhesion molecule, E-cadherin, as a central node of the estrogen response in multiple cancers. Using original mouse melanoma model, a cancer with a higher incidence of women than in men during this period, we identify a previously unknown estrogen-sensitizing signaling pathway linking E-cadherin to the Estrogen Receptor  and to the Gastrin Releasing Peptide Receptor (GRPR), promoting melanoma aggressiveness in female only. Inhibition of this axis through GRPR targeting suppress metastasis in mice, representing a very promising therapeutic approach. Our results demonstrate that E-cadherin prevents carcinoma sensitivity to estrogens and identify a new therapeutic avenue to treat women suffering from multiple E-cadherin-dependent cancers.

Ischemic Postconditioning (IPostC) Protects Fibrotic and Cirrhotic Rat Livers after Warm Ischemia.

Project description:BackgroundDecreased organ function following liver resection is a major clinical issue. The practical method of ischemic postconditioning (IPostC) has been studied in heart diseases, but no data exist regarding fibrotic livers.AimsWe aimed to determine whether IPostC could protect healthy, fibrotic, and cirrhotic livers from ischemia reperfusion injury (IRI).MethodsFibrosis was induced in male SD rats using bile duct ligation (BDL, 4 weeks), and cirrhosis was induced using thioacetamide (TAA, 18 weeks). Fibrosis and cirrhosis were histologically confirmed using HE and EvG staining. For healthy, fibrotic, and cirrhotic livers, isolated liver perfusion with 90 min of warm ischemia was performed in three groups (each with n=8): control, IPostC 8x20 sec, and IPostC 4x60 sec. additionally, healthy livers were investigated during a follow-up study. Lactate dehydrogenase (LDH) and thromboxane B2 (TXB2) in the perfusate, as well as bile flow (healthy/TAA) and portal perfusion pressure, were measured.ResultsLDH and TXB2 were reduced, and bile flow was increased by IPostC, mainly in total and in the late phase of reperfusion. The follow-up study showed that the perfusate derived from a postconditioned group had much less damaging potential than perfusate derived from the nonpostconditioned group.ConclusionIPostC following warm ischemia protects healthy, fibrotic, and cirrhotic livers against IRI. Reduced efflux of TXB2 is one possible mechanism for this effect of IPostC and increases sinusoidal microcirculation. These findings may help to improve organ function and recovery of patients after liver resection.
