RNA-Seq profiling of circular RNAs and the oncogenic role of circPVT1 in cutaneous squamous cell carcinoma

ABSTRACT: Circular RNAs(circRNAs),which are a type of non-coding RNA with a special structure, have attracted the attention of researchers in recent years.As a high throughput gene screening method, RNA-seq has been widely used in tumor research.In present study,we assessed the expression pattern of circRNAs in CSCC via RNA-seq on CSCC and matched paracancerous samples. We further identified circPVT1 as being overexpressed in CSCC tissues and cells.Subsequently, we identified the characteristics of circPVT1 in A431 and SCL1 cells.Finally, we designed an siRNA for the junction of circPVT1 and confirmed that the knockdown of circPVT1 could inhibit the proliferation, migration, and invasion of CSCC cells.In summary, using RNA-seq we established the expression profile of circRNAs in CSCC, and the allowed us to examine of the role of circPVT1as an oncogene in this context, highlighting this circRNA as a potentialtarget for CSCC therapy.
