Project description:Spinocerebellar ataxia type 3 (SCA3) is one of the polyglutamine (polyQ) diseases, which are caused by a CAG repeat expansion within the coding region of the associated genes. The CAG repeat specifies glutamine, and the expanded polyQ domain with mutation confers dominant toxicity on the protein. Traditionally, studies have focused on protein toxicity in polyQ disease mechanisms. Recent findings, however, demonstrate that the CAG repeat RNA, which encodes the toxic polyQ protein, also contributes to the disease in Drosophila. To provide insight into the nature of the RNA toxicity, we extracted brain-enriched RNA from flies expressing a toxic CAG repeat mRNA (CAG100) and a non-toxic interrupted CAA/G mRNA repeat (CAA/G105) for microarray analysis. This approach identified a set of genes that are differentially expressed specifically in CAG100 flies.

Project description:Understanding the interactions of nanostructures with biological systems is essential to nanotoxicological research. Using a microarray-based toxicogenomics approach at early stage, this study investigated the relationship between particle size and toxicity of silica particles (SP) with diameters of 30, 70, and 300 nm (SP30, SP70, and SP300) as well as the mechanism of injury in mice. Two experiments with SiO2 particles of different sizes were considered in mice. One was aimed to investigate the dose-response relationship of SP70 toxicity at a dose of 10, 20, or 40 mg/kg (experiment 1), and the other set to study the size-response relationship of SP-induced toxicity using SP30, SP70, or SP300 (experiment 2). In experiment 2, two dosages each of SP30, SP70, and SP300 were performed. One was 10 mg/kg at three particle sizes, and the other was toxic doses of the three particle sizes, i.e., 10 mg/kg for SP30, 40 mg/kg for SP70, and 200 mg/kg for SP300. The toxic doses of the three particle sizes of SP were decided on the basis of the results of histopathological examinations and serum biochemical analysis in our previous study.n = 5

Four independent replicates of flies expressing CAG0, CAG100, or CAA/G105 by elav-GAL4 were collected at 3 days. The transgenes are DsRed with either (CAG0) no CAG repeat in the 3'UTR, (CAG100) a CAG repeat of 100 CAGs in the 3'UTR, or (CAA/G105) an interrupted CAA CAG repeat in the 3'UTR (ref: Li et al., Nature 453:1107) The transgenes were adjusted to match in mRNA expression such that CAG0 flies had one copy of the transgene, CAG100 flies had 5 copies, and CAA/G105 had two copies. Fly brain tissue (about 20 brains per sample, dissected from head capsule, eyes, lamina and outer medulla removed) was dissected in cold phosphate buffered saline (PBS) and stored in Trizol reagent (Invitrogen Corporation, Carlsbad, CA) at -80Ë?C. Total brain RNA was extracted and purified using TRIzol reagent (Invitrogen) and the RNeasy Mini system (Qiagen), and treated with RNase-free DNase I (Qiagen). To define genes whose expression is altered in response to a toxic CAG repeat RNA, we compared CAG100 flies with age-matched flies expressing CAG0. To exclude transcriptional changes in response to a non-toxic trinucleotide repeat, a second gene list was generated by comparing CAA/G105 flies with age-matched CAG0 flies.

Project description:Our previous studies indicated that long-term B-toxicity was mainly limited to leaves of Citrus species; alternations of cell wall structure in vascular bundles were involved in tolerance to B-toxicity. Here, miRNAs and their expression pattern were first identified from B-treated Citrus sinensis (tolerant) and C. grandis (intolerant) leaves with high-throughput sequencing in order to identify miRNAs that might be involved in tolerance to B-toxicity. Results: 51 (23 conserved and 28 novel) miRNAs in C. grandis and 20 (6 conserved and 14 novel) in C. sinensis were differentially expressed after B-toxic treatment, respectively. Illumina sequencing results were validated by stem-loop qRT-PCR, and 82.5% qRT-PCR data coincided with those from direct sequencing. Among the differentially expressed miRNAs, miR395a and miR397a were the most significantly up-regulated in B-toxic C. grandis leaves, yet both were down-regulated in B-toxic C. sinensis ones. With modified 5’-RACE, four auxin response factor genes were confirmed as the real targets of miR160a, and two laccase (LAC) genes as those of miR397a, respectively. Localization of cell wall polysaccharides indicated that up-regulation of LAC4 resulted in secondary deposition of cell wall polysaccharides in regions near the pits of vessel elements in C. sinensis, and that down-regulation of both LAC17 and LAC4, via up-regulating their negative regulator miR397a, led to poorly developed vessel elements in C. grandis.Our findings demonstrated that miR397a played a pivotal role in woody Citrus tolerance to B-toxicity by targeting LAC17 and LAC4, of which both were responsible for secondary cell wall synthesis.

Project description:Metals at high concentrations can exert toxic effects on microorganisms. It has been widely reported that lowering environmental pH reduces effects of cadmium toxicity in bacteria. Understanding the effects of pH-mediated cadmium toxicity on bacteria would be useful for minimizing cadmium toxicity in the environment and gaining insight into the interactions between organic and inorganic components of life. Growth curve analysis confirmed that cadmium was less toxic to Escherichia coli at pH 5 than at pH 7 in M9 minimal salts medium. To better understand the cellular mechanisms by which lowering pH decreases cadmium toxicity, we used DNA microarrays to characterize global gene expression patterns in E. coli in response to cadmium exposure at moderately acidic (5) and neutral (7) values of pH. Higher expression of several stress response genes including hdeA, otsA, and yjbJ at pH 5 after only 5 minutes was observed and may suggest that acidic pH more rapidly induces genes that confer cadmium resistance. Genes involved in transport were more highly expressed at pH 7 than at pH 5 in the presence of cadmium. Of the genes that showed an interaction between pH and cadmium effects, 46% encoded hypothetical proteins, which may have novel functions involved in mitigating cadmium toxicity.

Project description:Specific gene changes after exposure to the heart toxin, bis(2-chloroethoxy)methane, were analyzed to elucidate critical pathways associated with recovery from heart toxicity. Bis(2-chloroethoxy)methane was administered to male F344/N rats at 0, 200, 400, or 600 mg/kg by dermal exposure for up to 16 days. Heart toxicity occurred at day 2 of exposure in all three regions of the heart (atrium, ventricle, interventricular septum) and was characterized by myofiber vacuolation, necrosis, mononuclear-cell infiltration, and atrial thrombosis. Ultrastructural analysis revealed that the primary site of damage occured in the mitochondrion. Damaged mitochondria had disrupted cristae and loss of membrane structure, and distention of the myocardial sarcoplasmic reticulum. By day 5, even though daily dosing was continued, the heart toxic lesions begin to resolved. By day 16, even though BCEM treatment continued, the heart appeared normal. RNA was extracted from control and dosed rats at day 2 and day 5, and analyzed for gene change by PCR. Gene changes associated with promoting cell growth including up regulation of growth factors and cell adhesion molecules were up regulated. Temperature regulation was promoted by up regulation of uncoupling proteins. Genes controlling the action potential of the heart, including ion flow and G protein signals, were altered. Because mitochondria are damaged by BCEM exposure, preserving and restoring energy function to the heart is considered to be essential, and up regulation of components of the ATP synthase unit occurred by day 5. Significant gene changes included up regulation of transcription factors, growth factors, membrane bound G protein signals, and ion transport controls. Multiple gene changes in the heart may contribute to restoration of heart function after exposure to environmental cardiotoxins. Keywords: toxicity, recovery, cardiac, heart

Project description:Adult Sprague Dawley rats were treated i.g. with 50 mg/kg alpha-naphthylisothiocyanate (ANIT) or vehicle (corn oil). Hepatobiliary liver injury occurred at 24 h postdose in ANIT rats with repair at 120h. Livers were extracted from rats at 24h and 120 h post ANIT exposure. This study investigated differences in mRNA expression between the injury and repair phases in the context of ANIT exposure. 8 week male Sprague Dawley rats were administered ANIT or vehicle for quantification of hepatobiliary injury via clinical chemistry biomarkers and pathology between 6 and 168 h postdosing. Rats sacrificed at 24 h and 120h postdosing coincided with peak injury and injury resolution, respectively.

Project description:Aquatic organisms are exposed to many toxic chemicals and interpreting the cause and effect relationships between occurrence and impairment is difficult. Toxicity Identification Evaluation (TIE) provides a systematic approach for identifying responsible toxicants. TIE relies on relatively uninformative and potentially insensitive toxicological endpoints. Gene expression analysis may provide needed sensitivity and specificity aiding in the identification of primary toxicants. The current work aims to determine the added benefit of integrating gene expression endpoints into the TIE process. A cDNA library and a custom microarray were constructed for the marine amphipod Ampelisca abdita. Phase 1 TIEs were conducted using 10% and 40% dilutions of acutely toxic sediment. Gene expression was monitored in survivors and controls. An expression-based classifier was developed and evaluated against control organisms, organisms exposed to low or medium toxicity diluted sediment, and chemically selective manipulations of highly toxic sediment. The expression-based classifier correctly identified organisms exposed to toxic sediment even when little mortality was observed, suggesting enhanced sensitivity of the TIE process. The ability of the expression-based endpoint to correctly identify toxic sediment was lost concomitantly with acute toxicity when organic contaminants were removed. Taken together, this suggests that gene expression enhances the performance of the TIE process. Wild-collected Ampelisca abdita were exposed to either control (from sites in Long Island Sound, labeled LIS) sediment, toxic (from site on Elizabeth River, labeled ER) sediment, a series of mixtures of LIS and ER sediment, sediments manipulated to alter toxin bioavailability, or toxicant amended sediments. Lethality was scored, and survivors were subjected to mRNA expression analysis via oligo microarray.
