Expression data from Il-27 stimulated normal human epidermal keratinocytes (NHEK) and THP-1 cells

ABSTRACT: IL-27 plays a pivotal role in regulating complex immune responses. To evaluate the role of IL-27 in modulation in the expression of cellular and innate immunity-asscoiated genes in human epidermal keratinocytes (primary cells) and the monocytic THP-1 cell line (pre-treated with PMA) were stimulated with IL-27 and genome-wide expression of these cells were investigated utilizing microarray assay.

Project description:THP-1 cells were treated with phorbol 12-myristate 13-acetate (PMA) at 25μg/ml for 48h.Then remove PMA and rest for 24h to obtain M0 macropahges. Further applied LPS and IFNγ or IL-4 and IL-13 for 24h to obtain M1, M2 macrophages, respectively. During polarization, cells were growth within control medium(LN) or medium with extra lipid mixture(LH).M0,M1,M2 cells were collected and applied to RNA-sequencing. We found that differential activation of macrophage had distinct metabolic signature.Extra lipids supplement did not significantly alter transcriptomes of macrophages.

Transcriptomics of THP-1 macrophages treated with or without IL-26 for 24h

Project description:Purpose: The aim of this study was to obtain a comprehensive transcriptomic analysis of IL-26-exposed macrophages. Methods: THP-1 derived macrophage were generated by incubating THP-1 cells with PMA (50 ng/ml) for 24h.Then THP-1 macrophages were treated with IL-26 for 24h, which were collected and applied to RNA-sequencing. The constructed sequencing library was sequenced using an Illumina Novaseq 6000 system. The expression of transcripts was calculated as fragments per kilobase of exon model per million mapped reads (FPKM). Results: In total, we identified 1303 differentially expressed protein-coding genes between untreated and IL-26-treated macrophages, including 667 up-regulated and 636 down-regulated genes.
