Project description:Transcripts were measured in infraspinatus muscle before (PRE) and after 16 weeks of tendon release (TR) and 6 weeks of subsequent repair (END) in the operated and contralateral (END-CC) muscle. A group of animals received intramuscular injections of nandrolone starting with the surgery (N22W). In another group injection of nandrolone was started at the time of tendon repair (N6W). The CONTROL group received no nandrolone.

Project description:Reversal of fatty infiltration of pennate rotator cuff muscle after tendon release is hitherto impossible. The administration of nandrolone starting at the time of tendon release prevents the increase in fat content, but does not revert established fatty infiltration. We hypothesised that tendon release and myotendinous retraction cause alterations in lipid related gene expression leading to fatty muscle infiltration, which can be suppressed by nandrolone through its genomic actions if applied immediately after tendon release. The effects of infraspinatus tendon release and subsequent tendon repair at 16 weeks were studied in six Swiss Alpine sheep. In the interventional groups, 150mg nandrolone was administered weekly after tendon release until sacrifice (N22W, n=6) or starting at the time of repair (N6W, n=6). Infraspinatus volume, composition, expressed transcripts, lipids, and selected proteins were analyzed at baseline, 16 and 22 weeks. Tendon release reduced infraspinatus volume by 22% and increased fat content from 11% to 38%. These changes were not affected by repair. Fatty infiltration was associated with up-regulation of 227 lipid species, and increased levels of the adipocyte differentiation marker PPARG2 (peroxisome proliferator-activated receptor gamma 2). Nandrolone abrogated lipid accumulation, halved the loss in fiber area percentage, and up-regulated androgen receptor levels and transcript expression in the N22W but not the N6W group. The results document that nandrolone mitigates muscle-to-fat transformation after tendon release via a general down-regulation of lipid accumulation concomitantly with up-regulated expression of its nuclear receptor and downstream transcripts in skeletal muscle. Reduced responsiveness of retracted muscle to nandrolone as observed in the N6W group is reflected by a down-regulated transcript response.

Project description:Morphological studies of skeletal muscle tissue have provided detailed insights into the architecture of muscle fibers, the surrounding cells, and the extracellular matrix. However, a spatial proteomics analysis of the skeletal muscle, including the muscle-tendon transition zone, is lacking. Here, we prepared thin cryotome muscle sections along the longitudinal axis of the mouse soleus muscle and measured each muscle slice using short LC-MS gradients. We generated more than 3000 high-resolution longitudinal protein profiles of central to distal skeletal muscle regions and created a molecular network of different skeletal muscle regions that reveals the complex architecture of the muscle-tendon transition zone. Among the proteins that show an increasing profile from muscle to tendon, we find proteins related to neuronal activity, fatty acid biosynthesis, and the renin-angiotensin system (RAS). Blocking the RAS in cultured mouse tenocytes using losartan reduces the synthesis of extracellular matrix proteins, including collagen and fibronectin. Overall, our analysis of thin cryotome sections provides a spatial proteome of skeletal muscle and reveals that the RAS acts as an additional regulator of the matrix within muscle-tendon junctions.

Project description:Tendon tissue growth is promoted by mechanical stimulation, but the mechanism is not well understood. Piezo1, a mechanical stress-responsive channel receptor, is expressed in tendon cells, and the fact that tendon tissue growth was accelerated in Piezo1 gain of function mice suggests that Piezo1 plays an important role in tendon tissue growth. RNA-seq of tendon tissues from these mice showed increased expression of tendon-related genes and decreased expression of muscle-related genes, suggesting that Piezo1 may play a role in maintaining and enhancing the properties of tendon cells.

Project description:Amniotic derived cells are ideal seed cells for regenerative medicine protocols since they conjugate a remarkable plasticity to safety properties. This study investigated the role exerted by human amniotic cells during the process of tendon healing. In particular, in the present research it has been evaluated, by microarray technique, the presence of transcriptome variations in human amniotic epithelial (hAECs) and mesenchymal (hAMCs) stem cells when xenotransplanted into the preclinical ovine model of tendon defect, compared to freshly isolated ones. This analysis allowed to understand in which way, after 28 days transplantation, hAECs and hAMCs transcripts have been affected. Total of 4 experiments.Two-condition experiment, Human amniotic stem cells xenotransplantated in a sheep achilles tendon vs.HAEC/HAMC Biological replicates: 2 Xenotrasplatations. Project description:Despite their important roles in the musculoskeletal system, tendon and ligaments are much less studied comparing to bone, cartilage and muscle. The lack of knowledge in tendon biology severely hinders the understanding of the etiologies of tendon related diseases and development of efficient clinical treatments. In mouse, Scx gene, encoding a Twist family bHLH transcription factor, is expressed in progenitors of all tendons and ligaments, as well as in mature tenocytes. Previous studies show that inactivation of Scx gene results in absence or severe hypoplastia of force transmitting tendons, with muscle anchoring tendons and ligaments are less affected. Here we report a set of ChIPseq data from E13.5 forelimbs of novel Scx-2xFlag tagged mice in which a Scx-Flag fusion protein is expressed recapitulating endogenous Scx expression, and a set of RNAseq data of Scx-GFP positive cells from E13.5 Scx wildtype, E15.5 Scx wildtype and homozygous mutant forelimbs. Using RNAseq and ChIPseq assays, we identify several genes exhibiting Scx-dependent tendon expression druing differentiation, with Scx binding peaks located within their promoter/enhancer regions. Thus these genes may play critical roles in mediating Scx regulated tendon cell differentiation. Our results provide new insights in the mechanisms of tendon development.
