LncRNA-mRNA expression profile and functional network of vascular dysfunction in endotoxemic rats

ABSTRACT: In our study, we used a rat model of LPS intervention to extract blood vessels. Differentially expressed LncRNA and mRNA were screened in lps rat blood vessels and normal rat blood vessels by microarray analysis. And through CNC and ceRNA analysis, lncRNA, miRNA, and mRNA regulatory network diagrams were constructed using verified lncRNA. This has the potential to highlight specific molecular functions and mechanisms. Later, we performed functional enrichment analysis and annotation to further explore the importance of lncRNA in vascular cell injury in sepsis. This work helps to reveal the underlying mechanism of vascular cell damage caused by sepsis and provides a new therapeutic target for controlling vascular damage in sepsis.

Both miRNA and mRNA microarrays (Series GSE74755) were performed with rat VSMCs and reciprocally regulated pairs of miRNA and mRNA were selected after bioinformatic analysis.

Transcriptomic profiling of LPS-induced adrenal inflammation in sorted adrenal endothelial cells from C576L6 mice

Project description:Altogether, this is the first study presenting the global changes of the transcriptome in the adrenal microvascular cells. The results of RNA sequencing and subsequent bioinformatics analysis revealed that adrenal vascular endothelial cells are active players not only involved in maintenance of the adrenal gland microenvironment, but also they play a key role in pathogen recognition and initiation of inflammation. However, our results also strongly suggest that systemic administration of LPS, which recapitulates to some degree situation in people with meningococcal sepsis, may develop certain damage, including vascular inflammation, hypoxia, blood coagulation and leakage. That may contribute to the adrenal dysfunction.

Comparative proteomic analysis in microdissected renal vessels of hypertensive and normotensive rats

Project description:Systemic hypertension has a profound impact on the renal vascular physiology. In order to elucidate the biological pathways and macromolecules deregulated by hypertension renal vessels were obtained by Laser Capture Microdissection (LCM) from Spontaneously Hypertensive Rats (SHR) and age-matched controls (20 weeks). Proteomic analysis was performed aiming to detect early molecular alterations associated with hypertension at the renal vessels before the onset of vascular damage. Proteomic analysis identified 688 proteins, of which 58 were differentially expressed (15 up-regulated and 43 down-regulated in SHR). Many of these proteins are involved in vascular tone regulation by modulating the activity of endothelial Nitric Oxide Synthase (eNOS) (e.g. Xaa-Pro aminopeptidase 1 (XPP1), N(G) N(G)-dimethylarginine dimethylaminohydrolase 1 (DDAH1), Dehydropteridine reductase (DHPR)) or in blood pressure control by regulating the renin-angiotensin system (e.g. Glutamyl aminopeptidase/Aminopeptidase A (AMPE), Aminopeptidase N (AMPN)). Moreover, pathway enrichment analysis revealed that the eNOS activation pathway is deregulated only in SHR. Our study demonstrates that hypertension causes early proteomic changes in the renal vessels of SHR. These changes are relevant to vascular tone regulation and consequently may be involved in the development of vascular damage and hypertensive nephrosclerosis. Therefore, the identified proteins could be considered as therapeutic targets.

SARS-CoV-2-induced disruption of a vascular bed in a microphysiological system caused by type-I interferon from bronchial organoids

Project description:Blood vessels show various COVID-19-related conditions including thrombosis and cytokine propagation. Existing in vitro blood vessel models cannot represent the consequent changes in the vascular structure or determine the initial infection site, making it difficult to evaluate how epithelial and endothelial tissues are damaged. Here, we developed a microphysiological system (MPS) that co-culture the bronchial organoids and the vascular bed to analyze infection site and interactions. In this system, virus-infected organoids caused damage in vascular structure. However, vasculature was not damaged or infected when the virus was directly introduced to vascular bed. The knockout of interferon-related genes and inhibition of the JAK/STAT pathway reduced the vascular damage, indicating the protective effect of interferon response suppression. The results demonstrate selective infection of bronchial epithelial cells and vascular damage by cytokines and also indicate the applicability of MPS to investigate how the infection influences vascular structure and functions.

Citrulline supplementation leads to increased immune suppression and more severe outcomes in infected newborn piglets

Project description:Background: Preterm infants are at high risk of infection and sepsis, which is associated with arginine (ARG) deficiency. Optimal nutrition for preterm newborns suspected with infection is critical to prevent sepsis development. Citrulline (CIT) supplementation is known to prevent sepsis in adult rodents, by maintaining sufficient blood ARG levels and vascular stabilization. We hypothesized that CIT therapy improve sepsis outcomes in infected preterm newborns via CIT and ARG metabolism.Results: After infection, oral CIT supplementation led to higher mortality, blood bacterial load, and circulating and hepatic inflammation, compared with the infected controls. Intravenous CIT administration also showed increased inflammation and bacterial burdens without increased mortality. Liver transcriptomics and data from in vitro blood stimulation indicated that CIT induces enhanced immunosuppression, which may impair resistance response to bacteria at early stage of infection and later cause excessive inflammation and tissue damage. Conclusion: Early stage of CIT supplementation exacerbates sepsis severity in infected preterm pigs, likely via induced enhanced systemic immunosuppression.

Project description:In order to assess the alteration in lncRNA expression in rat lung carcinogenesis induced by NNK, we determined the lncRNA expression profiles in 3 rat lung tumor samples and matched normal lung tissues and 2 blood samples from the control and NNK treatment group in the 95th week using Arraystar Rat lncRNA Microarray. We induced lung cancer using 4-(methylnitrosamino)-1-(3-pyridyl)-1-butanone (NNK) in a rat model and determined the lncRNA expression profiles in lung cancer tissues and rat blood samples.

Transcriptome analysis of blood monocytes from sepsis patients

Project description:Blood monocytes serve as the first line of host defense and are equipped to recognize and respond to infection by triggering an immune-inflammatory response. While most information on these cells comes from in vitro studies in humans or in vivo studies in mice, little is known about monocytes under human disease conditions. We investigated the role of monocytes during sepsis and its resolution in humans. A transcriptomal and functional analysis of blood monocytes from patients during gram negative sepsis and at recovery was performed. Monocytes from sepsis patients showed upregulation of a large number of pro-inflammatory genes and cytokines/chemokines, consistent with an ongoing systemic inflammation. However, these cells showed impairment to ex vivo endotoxin (LPS) challenge, displaying a quantitative decrease in the number of LPS-inducible genes. Moreover, they downregulated the expression of several pro-inflammatory cytokine/chemokine genes, activation marker genes and transcription factors associated with monocyte/macrophage activation, upon ex vivo LPS stimulation. Functionally, they downregulated expression of inflammatory cytokines/chemokines and antigen presentation-related molecules and functions. In contrast, genes and functions related to phagocytosis, anti-microbial activity and tissue remodeling where remained unaffected or even enhanced . Collectively, our observations suggest a genetic and functional re-programming of these cells during human sepsis progression. Understanding the molecular mechanisms which regulate this re-programming will allow to devise strategies which could modulate the response of these cells and hence, disease progression. Blood monocytes from gram-negative sepsis patients during sepsis (Sepsis) and following their recovery (Recovery/Basal) as well as healthy donor (control) were isolated. Thereafter, these cells were treated ex vivo with or without LPS for 3h and analysed for transcriptomic study.

Project description:Increased numbers of endothelial cells are observed in peripheral blood of cancer patients. These circulating endothelial cells (CECs) may contribute to the formation of blood vessels in the tumor or reflect vascular damage caused by treatment or tumor growth. Characterization of these cells may aid in the understanding of the angiogenic process and may provide biomarkers for treatment efficacy of angiogenesis inhibitors. To identify markers typical for CECs in cancer patients we assessed global gene expression profiles of CD146 immunomagnetically enriched CECs from healthy donors and patients with metastatic breast, colorectal, prostate, lung and renal cancer.

Secretome of rat smooth muscle cells with stretch

Project description:The extracellular matrix (ECM) is fundamental to cellular and tissue structural integrity and provides mechanical cues to initiate diverse biological functions1. The quality and quantity of ECM determines tissue stiffness and controls gene expression, cell fate, and cell cycle progression in various cell types. ECM-cell interactions are mediated by focal adhesions (FAs), the main hub for mechanotransduction, connecting ECM, integrins and cytoskeleton. In the blood vessels, an additional layer of mechanical cues derived from pulsatile blood flow and pressure play a pivotal role in homeostasis and disease development. However, how cells sense and integrate different mechanical cues to maintain the blood vessel wall is largely unknown. Vascular ECM is synthesized by SMCs from mid-gestation to early postnatal period and determines the material and mechanical properties of the blood vessels14. Owing to the long half-life of ECM, blood vessels are generally thought to stand life-long mechanical stress without regeneration, particularly in large arteries. However, it is not completely known whether local micro-remodeling system exists for the maintenance of the vessel wall. To search for a potential remodeling factor(s), we performed cyclic stretch experiments (1 Hz; 20% strain) using rat vascular SMCs for 20 hours (h) with or without Brefeldin A (BFA), a protein transporter inhibitor. Conditioned media (CM) were analyzed using quantitative mass spectrometry.

Expression data from endthelial cells in ductus arteriosus and aorta in rat fetuses or neonates

Project description:Endothelial cells (Ecs) lining the blood vessels have been known to have a variety of functions and play a central role in homeostasis of the circulatory system. Since ductus arteriosus (DA) has unique characteristics comparing other vessels, we hypothesized that ECs were the primary target to investigate the molecular mechanisms of DA-specific vasoconstriction and vascular remodeling. Using fluorescence activated cell sorter, we isolated ECs from pooled tissues from the DA or the aorta of Wistar rat fetuses on the 21st day of gestation (e21) or 30 minutes after birth (day0). We prepared 3 sets of ECs in each tissues and time points. We aimed to compare the gene profiles of DA and the aorta at each developmental stage.
