MiR-199 Family Regulates Sonic Hedgehog Expression During Craniofacial Development

ABSTRACT: Through microarray analyses of three SHH conditions, we were able to identify the miR-199 family as a family of miRNAs that is regulated by SHH signaling from the brain.

The miR-17/92 Polycistron Is Up-regulated in Sonic Hedgehog-Driven Medulloblastomas and Induced by N-myc in Sonic Hedgehog–Treated Cerebellar Neural Precursors

Project description:Medulloblastoma is the most common malignant pediatric brain tumor, and mechanisms underlying its development are poorly understood. We identified recurrent amplification of the miR-17/92 polycistron proto-oncogene in 6% of pediatric medulloblastomas by high-resolution single-nucleotide polymorphism genotyping arrays and subsequent interphase fluorescence in situ hybridization on a human medulloblastoma tissue microarray. Profiling the expression of 427 mature microRNAs (miRNA) in a series of 90 primary human medulloblastomas revealed that components of the miR-17/92 polycistron are the most highly up-regulated miRNAs in medulloblastoma. Expression of miR-17/92 was highest in the subgroup of medulloblastomas associated with activation of the sonic hedgehog (Shh) signaling pathway compared with other subgroups of medulloblastoma. Medulloblastomas in which miR-17/92 was up-regulated also had elevated levels of MYC/MYCN expression. Consistent with its regulation by Shh, we observed that Shh treatment of primary cerebellar granule neuron precursors (CGNP), proposed cells of origin for the Shh-associated medulloblastomas, resulted in increased miR-17/92 expression. In CGNPs, the Shh effector N-myc, but not Gli1, induced miR-17/92 expression. Ectopic miR-17/92 expression in CGNPs synergized with exogenous Shh to increase proliferation and also enabled them to proliferate in the absence of Shh. We conclude that miR-17/92 is a positive effector of Shh-mediated proliferation and that aberrant expression/amplification of this miR confers a growth advantage to medulloblastomas.

Project description:To identify putative novel specific targets of miR-199-5p, miR-199a-3p and miR-214-3p, we overexpressed these miRNAs in human MRC5 pulmonary fibroblasts (CCL-171) using synthetic pre-miRNAs or a synthetic “negative” pre-miRNA control (miR-Neg). RNA samples were harvested 48 hours post-transfection and 3 independent experiments were carried out.

Impact of miR-199-5p overexpression and CAV1 silencing on human lung fibroblasts

Project description:To identify putative novel specific targets of mir-199-5p, we overexpressed miR-199a-5p as well as miR-21 and a siRNA targeted against CAV1 in human HFL1 pulmonary fibroblasts (CCL-153) by transfecting them with synthetic pre-miRNAs or a synthetic “negative” pre-miRNA as control (miR-Neg). RNA samples were harvested at 48 hours post-transfection and 2 independent experiments were carried out. Additional samples correspond to HFL1 cells treated or not with 10ng/ml TGFbeta for 48 hours in the absence of serum (2 independent experiments). 2 independent experiments performed in a one color design, corresponding to 7 conditions (miR-Neg, miR-199-5p, miR-21, si-Neg, si-CAV1, control, TGFbeta) and a total of 14 samples.

Sonic hedgehog signaling in craniofacial development.

Project description:Mutations in SHH and several other genes encoding components of the Hedgehog signaling pathway have been associated with holoprosencephaly syndromes, with craniofacial anomalies ranging in severity from cyclopia to facial cleft to midfacial and mandibular hypoplasia. Studies in animal models have revealed that SHH signaling plays crucial roles at multiple stages of craniofacial morphogenesis, from cranial neural crest cell survival to growth and patterning of the facial primordia to organogenesis of the palate, mandible, tongue, tooth, and taste bud formation and homeostasis. This article provides a summary of the major findings in studies of the roles of SHH signaling in craniofacial development, with emphasis on recent advances in the understanding of the molecular and cellular mechanisms regulating the SHH signaling pathway activity and those involving SHH signaling in the formation and patterning of craniofacial structures.

Sonic hedgehog-heat shock protein 90β axis promotes the development of nonalcoholic steatohepatitis (miRNA-Seq)

Project description:The incidence rate of nonalcoholic steatohepatitis (NASH) is on the rise with no effective clinical treatment available to date. Understanding the key molecular mechanism underlying the process of NASH can provide theoretical support for the development of safe and effective therapeutic drugs. Sonic hedgehog (SHH) and heat shock protein 90β (HSP90β) have been implicated in NASH but their molecular mechanisms of action remain elusive. HSP90β was found to be a key SHH downstream molecule that promoted NASH processes In hepatocytes, SHH reduced HSP90β ubiquitylation through deubiquitylase USP31, thus preventing HSP90β degradation and promoting hepatic lipid synthesis. As an important component protein in the formation of exosomes, HSP90β is high in high fructose, high fat, and high cholesterol (HFFC) diet-induced NASH mouse model, leading to secretion of exosomes enriched with miR-28-5p. miR-28-5p directly targets and decreases Rap1b levels, which in turn promote NF-κB transcriptional activity in macrophages and stimulate the expression of inflammatory factors. Genetic deletion, pharmacological inhibition of the SHH-HSP90β axis, or delivery of miR-28-5p to macrophages in the liver, impaired NASH symptomatic development. Importantly, analysis of NASH patient sera revealed a markedly higher abundance of miR-28-5p compared to healthy donors. Taken together, the SHH-HSP90β-miR-28-5p axis offers promising therapeutic targets against NASH, and serum miR-28-5p may serve as a novel NASH diagnostic biomarker.
