RNA-Seq analysis to compare immature myeloid cells from PARP1CKO and PARP2CKO mice with their corresponding littermate controls

ABSTRACT: We compared expression profiles of immature myeloid cells from PARP1CKO and PARP2CKO mice with their corresponding littermate controls by Next-generation sequencing (NGS). The results showed that genes regulated by PARP1 and PARP2 were largely distinct with only a small subset overlapping. PARP1 regulated genes are related to secretion, whereas PARP2 regulated genes are related to response to external stimulus and leukocyte migration. PARP2 deficiency down-regulated the expression of immune cytokines such as CCL3, CCL4, CCL6 and CCL9. In contrast, these genes were unaffected by PARP1 deficiency. This study has uncovered important functional and mechanistic distinctions between PARP1 and PARP2. The findings has also revealed specific roles of PARP2 in immature myeloid cells in regulating chemokine expression and immune response.
