The anti-fibrosis effect of iguratimod on skin tissues of systemic sclerosis patients


ABSTRACT: To evaluate the possibility of iguratimod working on SSc patients in clinical practice, we collected skin biopsies from three early SSc patients. Three grafts were divided equally into two irradiated nude mice and treated with iguratimod or vehicle solution subcutaneously for five weeks. Total RNA from cultured fibroblasts or xenografts were extracted by TRIzol.Differential expression analysis of two groups was performed using the DESeq2 from R package. we find the ensemble of genes encoding extracellular matrix and ECM-associated proteins, which was termed as matrisome, was enriched at the top of the GSEA list.In the ensemble, there were pro-fibrotic cytokine genes such as TGFB1 and FGFs, and WNTs; ECM degradation enzymes genes such MMPs and CTSs; ECM-associated chemokine such CCL13 and CCL18; and notably, remarkable amount of glycoprotein/ proteoglycan genes such as LRG1, PRG4, HAPLN1 and PODNL1.
