Project description:Senescence plays a key role in various physiological and pathological processes. We reported that injury-induced transient senescence correlates with heart regeneration, yet the multi-omics profile and molecular underpinnings of regenerative senescence remain obscure. Using proteomics and single-cell RNA-sequencing, here we report the regenerative senescence multi-omic signature in the adult mouse heart and establish its role in neonatal heart regeneration and Agrin-mediated cardiac repair in adult mice. We identified early growth response protein 1 (Egr1) as a regulator of regenerative senescence in both models. In the neonatal heart, Egr1 facilitates angiogenesis and cardiomyocyte proliferation. In adult hearts, Agrin-induced senescence and repair require Egr1, activated by the integrin/FAK-ERK/Akt1 axis in cardiac fibroblasts. We also identified cathepsins as injury-induced senescence-associated secretory phenotype (SASP) components that promote ECM degradation and potentially assist in reducing fibrosis. Altogether, we uncovered the molecular signature and functional benefits of regenerative senescence during heart regeneration, with Egr1 orchestrating the process.

Project description:The adult mammalian heart heals after myocardial infarction (MI) by deposition of scar tissue, leading to downstream arrhythmia, remodelling and heart failure1. In contrast, adult zebrafish and neonatal mouse hearts are capable of regenerating after injury. Macrophages are key mediators of tissue repair and appear to be required for both regeneration and healing by scar formation, but the mechanisms underlying these distinct roles are poorly understood2-4. Here we investigated how macrophages differentially influence the mode of repair by determining their responses in scar-free versus scar-induced healing, comparing ventricular resection with cryo-injured adult zebrafish hearts and neonatal versus adult mouse hearts after MI. Unbiased transcriptomics revealed molecular programmes implicating macrophages in the initiation and resolution of inflammation to dictate the kinetics of scarring during zebrafish regeneration and the activation of direct and indirect pathways to drive fibrosis in the adult mouse heart. Most notably we observed up-regulation of collagen isoforms in both zebrafish and mouse macrophages following injury. Adoptive transfer of macrophages, from resected zebrafish hearts into cryo-injured hosts and splenic monocyte-derived macrophages from adult mouse donors into neonatal hearts, enhanced scar formation and induced fibrosis, respectively, via cell autonomous production of collagen. In zebrafish, macrophage-specific targeting of collagen 4a binding protein and cognate collagen 4a1 followed by transfer led to significantly reduced scarring in cryo-injured hosts, as further evidence of a direct macrophage contribution to collagen deposition and scar formation. These findings contrast with the current model of scarring, whereby collagen is laid down exclusively by myofibroblasts, and implicate macrophages as critical regulators of heart repair.

Project description:Recent clinical studies indicate a strong link between AKI and progression of CKD. The increasing prevalence of AKI must compel the nephrology community to consider the long-term ramifications of this syndrome. Considerable gaps in knowledge exist regarding the connection between AKI and CKD. The 13th Acute Dialysis Quality Initiative meeting entitled "Therapeutic Targets of Human Acute Kidney Injury: Harmonizing Human and Experimental Animal Acute Kidney Injury" convened in April of 2014 and assigned a working group to focus on issues related to progression after AKI. This article provides a summary of the key conclusions and recommendations of the group, including an emphasis on terminology related to injury and repair processes for both clinical and preclinical studies, elucidation of pathophysiologic alterations of AKI, identification of potential treatment strategies, identification of patients predisposed to progression, and potential management strategies.

Project description:Myocardial infarction (MI) results from occlusion of blood supply to the heart muscle causing death of cardiac muscle cells. Following myocardial infarction (MI), scar formation acts to mechanically stabilise the injured heart to prevent rupture and death. While fibroblasts and macrophages are implicated in post-MI scar formation and maturation, their exact contributions to this process are poorly characterised, especially in the long-term (i.e., beyond 1-week post-MI). Here, we employ state-of-the-art spatially-targetted optical micro-proteomics (STOMP) to isolate proteomes of tissue fractions enriched for fibroblasts (sma+) and macrophages (cd68+) over a 6-week post-MI timecourse and compare them to whole-scar proteome. We illustrate dynamic, specific changes in sma- and cd68-enriched fraction protein composition over 1-6 weeks post-MI with some of these changes reflected in whole-infarct preparations. These results link specific cell populations to specific protein changes in whole infarct.

Project description:Mice with either a cranial bone defect, skin biopsy punch injury, quadriceps volumetric muscle loss defect or myocardial infarction (MI) were treated with Tregs, either via local hydrogel-mediated delivery (for bone, muscle and skin), or with systemic (intravenous) delivery (for heart). Control mice were treated with hydrogel alone (for bone, muscle and skin) or PBS (for heart). On day 4 and 7 post-MI, the injured tissues were harvested and the endogenous monocytes/macrophages were sorted and used for mini-bulk RNA sequencing, to compare the transcriptomic profiles of injured tissue monocytes/macrophages in Treg-treated and untreated mice post-injury.

Project description:Human UCB-MSCs showed donor-specific variation of therapeutic efficacy in improving LV systolic function, reducing infarct area, and preserving wall thickness after MI, even though there were no significant differences in MSC phenotypes. UCB-MSCs (M02) which showed better efficacy had better paracrine activity and unique gene expression profile than others. In DNA microarray, in contrast to M01, M02 showed unique gene expression profiles; up-regulated anti-apoptotic and down-regulated apoptotic gene expression.

Project description:Transcriptomic profile of recovered exogenous Tregs from injured bone, muscle, and skin of mice that were locally treated with Tregs, as well as heart, mediastinal lymph nodes (MLN) and spleen from mice with myocardial infarcts (MI) that were systemically treated with Tregs. Mice with tissue injuries were treated with exogenous Tregs that were sorted from spleens of Foxp3(IRES-mRFP) mice - C57BL6/J mice with bone, muscle and skin injuries were treated via hydrogel-mediated local delivery of Tregs soon after injury, while mice with myocardial infarcts (MI) were treated with systemic (intravenous) delivery of Tregs one day post-MI. Three days after Treg-delivery, the injured bone, muscle, and skin tissues, as well as heart, mediastinal lymph nodes and spleens from mice with MI were harvested, and the exogenous (delivered) Tregs were recovered by FACS sorting. These sorted exogenous Tregs recovered at day 3 post-Treg delivery (Day 3 recovered Tregs) were then used for mini-bulk RNA sequencing along with spleen Tregs before delivery as a control (Day 0 Spleen Tregs).
