A multiple reporter protein tolerant mouse model

ABSTRACT: We report the generation of a novel mouse strain that is immunologically tolerant to exogenously introduced reporter proteins ('Tol' strain). As a control, we test whether any transcriptomic changes can be observed between the Tol strain and wild-type littermate. This is tested for thymus, cerebrum and cerebellum.

Project description:SLC25A46 is an outer mitochondrial protein, member of the mitochondrial carrier family (SLC25), whereas mutations result in mitochondrial structure abnormalities leading to a wide spectrum of neurological disorders. SLC25A46 is known to interact with members of the MICOS (Mitochondrial Contact Site) complex involved in mitochondrial cristae organization, and components of the endoplasmic reticulum membrane. In the present work, our aim was to identify the interactome of SLC25A46 in cerebrum, cerebellum, heart and thymus from TgSLC25A46-FLAG transgenic mice through immunoprecipitation with anti-FLAG beads and proteomic analysis with LC-MS/MS.

Adaptive evolution provides a high-light stress tolerance in Synechocystis sp. PCC 6803

Project description:we obtained a HL tolerant (Tol) strain Synechocystis sp. PCC 6803 by adaptive evolution experiment that the cells were repeatedly subcultured for prolonged periods of time (52 days) under high light stress condition (7000 to 9000 μmol m-2 s-1). Although the growth of the parental strain almost stopped under 9000 μmol m-2 s-1, no growth inhibition was observed in the Tol strain. Furthermore, the growth rate was identical to that of parental strain under low light condition (40 μmol m-2 s-1). To further investigate the high light tolerant mechanisms in the Tol strain, the transcriptome was performed. The transcriptome data suggests the increase of isiA expression in the Tol strain under HL condition. The overexpression of isiA successfully enhanced the HL stress tolerance in the parental strain. The HL tolerant mechanism was different from previous reported mechanisms, such as a reduction of the light-harvesting antenna size. The tolerant strain would be an attractive host for bio-production under high light conditions.

MOLECULAR CHARACTERIZATION OF LIVER ALLOGRAFTS FROM OPERATIONALLY TOLERANT TRANSPLANT RECIPIENTS (Affymetrix)

Project description:In clinical organ transplantation complete cessation of immunosuppressive therapy can be successfully accomplished in selected recipients providing a proof-of-principle that allograft tolerance is attainable in humans. The intra-graft molecular pathways associated with human allograft tolerance, however, have not been comprehensively studied before. In this study we analyzed sequential liver tissue samples collected from liver recipients enrolled in a prospective multicenter immunosuppressive withdrawal clinical trial. Tolerant and non-tolerant recipients differed in the intra-graft expression of genes involved in the regulation of iron homeostasis.These results point to a critical role of iron homeostasis in the regulation of intra-graft alloimmune responses in humans and provide a set of novel biomarkers to conduct drug-weaning trials in liver transplantation. The complete database comprised the expression measurements of 54,675 genes for liver inmunotolerance groups: 9 Tolerant (TOL) 10 Non Tolerant (Non TOL).A subset of 5 tolerant samples (TOL POST) were taken 1 year after inmunosuppresive therapy.

Transcription profiling of prosaposin deficient mice show molecular alterations precede neuronal deficits

Project description:Prosaposin encodes, in tandem, four small acidic activator proteins (saposins) with specificities for glycosphingolipids hydrolases in lysosomes. To explore the molecular mechanism(s) of disease progression, temporal transcriptome microarray analyses of cerebrum and cerebellum tissues were conducted using mRNA from three prosaposin deficiency mouse models: PS-NA (hypomorphic prosaposin deficiency), PS-/- (prosaposin null) and 4L/PS-NA (a V394L/V394L glucocerebrosidase mutation and PS-NA) mice. Our results indicate that regionally specific gene expression abnormalities preceded the histological and behavioral changes and CEBPD is a candidate regulator of brain disease in prosaposin deficiency. The alterations of gene expression are detected at birth and are more profound in cerebellum than cerebrum. Experiment Overall Design: In order to increase the temporal resolution of expression profile in brain, the disease progression in those models were inverstigated in two regions of brains (cerebellum and cerebrum) at three or four time points according to the genotypes. PS-/-: new born (0d), 10 days (10d), 20 days (20d), 25 days (25d); PS-NA: new born, 4 weeks (4w), 12 weeks (12w), 18 weeks (18w); 4L/PS-NA: 4 weeks (4w), 12 weeks (12w), 18 weeks (18w). The data from those models were analyzed relative to the corresponding wild type at same time point (0d, 10d, 20d, 4w, 12w, 18w).
