Project description:In order to further determine the je virus (JEV) infection in C57BL / 6 mice after its miRNA levels change, we use the Agilent micrornas chip as a platform, to detect the uninfected group (control group) and the experimental mouse total RNA, it is found that the miRNA levels decrease infection group, and picked out the ten changes obviously micrornas verification, the result is the same.

Project description:Japanese Encephalitis Virus (JEV) modulates different proteins at different time points of infection to favour its propagation in the host cells. The dysregulation of intertwined pathways in the host has implications for virus pathogenesis. This study aims to decipher the global proteome of Japanese encephalitis infected THP-1 derived macrophages at 24 hours post-infection and 48 hours post-infection, which will further help to deduce the interwoven pathways regulated upon JEV infection.

Project description:Japanese encephalitis (JE) is an acute encephalitis syndrome contributed to Japanese encephalitis virus (JEV) infection. It is the chief cause of viral encephalitis in Asian. In recent years, association of JEV infection with neurological problems such as Guillain-Barré syndrome had reported. Nevertheless, its potential pathogenic mechanism has never been reported. Therefore, it is urgent to explore the relationship between peripheral nerve injury and JEV infection. Here, we use the liquid chromatography-tandem mass spectrometry (LC-MS/MS) technique to make out the protein expression levels of mice sciatic nerve between JEV infection group and the sham group. In general, 4303 proteins were identified by MS, and 187 differentially expressed proteins were found. There were 105 proteins up-regulated in the injured sciatic nerve, and 82 proteins were down-regulated. Functional enrichment analysis of differentially expressed proteins showed that the up-regulated proteins were mainly related to immune regulatory response, and the down-regulated proteins were related to ribosomal structural components and translation.

Project description:Japanese Encephalitis (JE) is an important causal of viral encephalitis in Eastern and Southeast Asia. Neuronal injury and varying degrees of neuroinflammation are often pathological hallmarks of JE, which result in high morbility and mortality rates. However, a clear understanding on neuron cells' responses to JEV infection remains elusive. This study, therefore, utilized the Affymetrix Human PrimeView array for a robust profiling of JEV infected SK-N-MC cells during the active phase of infection in order to investigate the molecular details of the host cell responses.

Project description:Japanese encephalitis virus (JEV) is a causative agent of encephalitis, mostly prevalent in Asia and South-Asian countries. Neuroinflammation is the hallmark of encephalitis, mediated in parts through the activation of brain-resident microglial cell, thus playing a critical role in determining pathogenesis. Deregulated activity of microglia can be lethal for the brain tissue. Interferon Regulatory Factor 8 (Irf8) mediates differentiation and transformation of microglia to reactive phenotypes and also regulates antiviral response through cross-talk with interferon pathway. In this study, we have conducted a comparative study between JEV infected C57BL/6 wild type and Irf8 Knockout (IRF8-/-) to identify the genes directly or indirectly regulated by IRF8.

Project description:Infection by neurotropic virus Japanese Encephalitis Virus (JEV) is characterized by profound neuronal cell death and neuroinflammation. Long non-coding RNAs (lncRNAs) are critical regulatory players in diverse biological processes, including viral pathogenesis. We use whole transcriptomic sequencing to identify a lncRNA JINR1 (JEV-induced non-coding RNA 1) induced upon JEV infection in neuronal cells. Transcription factor NF-κB triggers JINR-1 expression during JEV infection. Loss of JINR-1 impairs virus replication and reduces JEV-induced neuronal cell death and expression of genes involved in ER stress and neuroinflammation. Mechanistically, JINR1 inhibits the expression of mir-216-5p, which inhibits host factors GRP78 and c-JUN and the viral genome. In line with its role as a pro-viral host factor, JINR1 interacts with RBM10 and NF-κB to promote the expression of genes involved in ER stress and neuroinflammation. Our results suggest a role for JINR-1 in promoting JEV-induced cell death and neuroinflammation.

We used microarray analysis to identify cellular genes that are differentially regulated following infection of the brain with JEV (P3) or WNV (New York 99). Gene expression data for these flaviviruses was compared to that induced following infection of the brain with reovirus (Type 3 Dearing), an unrelated neurotropic virus. Although several studies have described gene expression changes following virus infection of the brain, this report is the first to directly compare large-scale gene expression data from different viruses. We found that a large number of genes were up-regulated in common to infections with all 3 viruses (fold change > 2, P < 0.001), including genes associated with interferon signaling, the immune system, inflammation and cell death/survival signaling. In addition, genes associated with glutamate signaling were down-regulated in common to infections with all 3 viruses (fold change > 2, P < 0.001). These genes may serve broad spectrum therapeutic targets for virus-induced CNS disease. A distinct set of genes were up-regulated following flavivirus-infection, but not following infection with reovirus. Project description:Japanese encephalitis virus (JEV) uses mosquitoes as the main carrier and pigs as the host. Over 60% of the world's population lives in the coverage region of JEV. Each year, 50000 to 170000 cases are reported to suffer the disease caused by JEV. The mortality rate in human is around 30%, and 50% survived patients have long-term neurological sequelae. JEV infection in pigs mainly occurs in the epidemic areas, infection with JEV can cause miscarriage and stillbirth in pigs. Infected piglets exhibit serious neurological diseases. Thus JEV poses a major risk to human and animal health. Epidemic Japanese encephalitis caused by JEV has spread in 24 countries, including Australia, Japan, India and most of the western Pacific and Southeast Asian countries. Based on the nucleotide sequences of JEV envelope protein, it can be divided into five genotypes, namely genotype I–genotype V. Genotype III was first detected in China, Genotype I has become the dominant type since it was isolated in Yunnan in 1977, then coexists with Genotype III. Understanding the distribution and prevalence of JEV genotypes have of great significance for the follow-up study, prevention and control of JEV. JEV activates the inflammatory response of macrophages. Results of proteomics showed that proteins of cytokine-mediated signal pathway and TNF signal pathway were activated at 6 hpi. Innate immune response, apoptosis signal pathway and TLR-related pathway were all activated at 12 hpi. Defense reaction and enzyme inhibitor activity related signal pathways were activated after infection of macrophages at 24 hpi.
