Analysis of global gene expressions in initial infection of Mycobacterium avium subsp. paratuberculosis using epithelial cells-peripheral blood mononuclear cells culture passage model.

ABSTRACT: Purpose: Understanding host-MAP interactions in initial stage of infection Methods: Native MAP and MDBK processed (4h) MAP were infected to bovine PBMCs. Total RNA after 24h and 72h were harvested. RNA-Seq was performed using total RNA of each group. Result: The gene expression patterns commonly activated in both groups suggest that Th17 activation is the main immunological response to MAP infection.

Project description:Mycobacterium avium subspecies paratuberculosis (MAP) causes chronic progressive granulomatous enteritis leading to diarrhea, weight loss, and eventual death in ruminants. Commercially available vaccines provide only partial protection against MAP infection and can interfere with the use of current diagnostic tests for bovine tuberculosis in cattle. In the current study, we characterized immune responses in calves to vaccines containing either MAP fusion protein particles (MAP antigens Ag85A202-347-SOD1-72 -Ag85B173-330-74F1-148+669-786 as a fusion), recombinant MAP (rMAP) fusion protein or commercially available Silirum(R) vaccine.

Project description:Purpose : Comparative analysis of host transcriptomes according to infection route following MAP infection Methods : Mice were infected with MAP by oral/intraperitoneal (IP) route. Total RNA were harvested from spleen and mesenteric lymph node after 6 weeks. RNA-seq was performed with total RNA of each sample. Results : Canonical pathway analysis showed that LXR/RXR activation pathway had negative z-score in IP group, whereas positive in oral group.

Transcriptional profiling of whole-blood in Mycobacterium avium subsp. paratuberculosis-infected cattle

Project description:To provide novel insights into understanding the host-immune response during the different stages of progression of the disease, we performed gene expression profiling in Mycobacterium avium subsp. paratuberculosis infection in cattle. We analyzed the transcriptional profiles of whole-blood cells of cattle identified and grouped according to the presence of MAP-specific antibodies and the MAP shed by them. The grouping was based on the results obtained by ELISA and PCR analyses as follows: 1) Test1 group: MAP-negative results obtained by ELISA and positive results obtained by PCR; 2) Test2 group: MAP-positive results obtained by ELISA and negative results obtained by PCR; 3) Test3 group: MAP-positive results obtained by ELISA and positive results obtained by PCR; 4) uninfected control: MAP-negative results obtained both by ELISA and PCR analysis.

Expression data of sheep exposed to Mycobacterium avium subsp. Paratuberculosis

Project description:In this study, consistent gene expression changes were analysed to identify gene-to-gene interaction and functional pathway relationships associated with the MAP exposure outcomes of either a paucibacillary or a multibacillary disease form or sheep resilient to disease. The results suggest that although groups of genes are differentially changed in the MAP exposed sheep regardless of clinical outcome, there are distinct variations in gene expression patterns between the paucibacillary and multibacillary forms of disease and of particular note was the finding that MAP exposed animals with evidence of recovery or resilience respond to MAP exposure with a pattern of gene and molecular pathway interactions distinct from those animals that progress to clinical paratuberculosis.

Cathepsin S in Mycobacterium avium subsp. paratuberculosis Infected Dairy Cows

Project description:In this study, samples of 16 dairy cows from a MAP infected farm were used. Serum, milk and fecal samples were collected. Categorizing these cows into two groups based on their MAP infection status different standard methods for detection MAP were applied. Healthy controls showed no positive results in enzyme-linked immunosorbent assay (ELISA) with serum and milk samples (cattletype MAP Ab, Qiagen, Hilden, Germany; In-direct, IDVet, Grabels, France) and after cultivation of fecal samples on commercial Her-rold´s Egg Yolk Agars (HEYM agar, Becton Dickinson, Heidelberg, Germany) for 12 weeks. Cows with positive results were grouped into MAP infected cows. Specifically, for mass spectrometry analysis serum of seven MAP infected cows and seven healthy controls were used. All animals were from the same farm and were kept under the same environmental conditions. For additional mass spectrometry analysis with a further control group sam-ples of 21 dairy cows from an uninfected farm were examined. All cattle from this farm showed negative results in ELISA with serum and milk samples. Additionally, there was never a positive result in regularly tested fecal samples and sock swab samples of this farm. For verification of differential CTSS expression in Western blot analysis five dairy cows from another infected farm were consultedincluded. MAP status of these cows was analyzed by cultivation of fecal samples on HEYM agar for 12 weeks and ELISA with se-rum samples. In detail, two cattle were categorized into healthy controls and three cattle into MAP infected cows. Project description:In order to gain a better understanding of MAP infection in immature macrophages, high-throughput sequencing technology was used to perform an analysis of mRNAs profiles of bovine monocyte derived macrophages upon MAP infection. The total numbers of raw reads collected from the MAP-infected and control MDM cells were 93.6 and 102 million raw reads per library. After removing low-quality sequences, Adaptor sequences and sequences containing more than 5% N (N means that base information cannot be determined), 91.7 and 100 million clean reads were obtained respectively. The analyses of differential expression revealed 618 DE mRNAs in MDM challenged with MAP compared to the control MDM, Among the DE mRNAs, 322 were upregulated and 296 were downregulated.Furthermore, The GO and KEGG pathway enrichment of these mRNA targets were performed.

Project description:In order to gain a better understanding of MAP infection in immature macrophages, high-throughput sequencing technology was used to perform an analysis of miRNAs profiles of bovine monocyte derived macrophages upon MAP infection.The total numbers of raw reads collected from the MAP-infected and control MDM cells were 14.3 and 17.0 million raw reads per library.The length distribution of the clean reads was focused on 21–24 nt in length and read counts of 22 nt were highest.A total of 510 mature miRNAs were identified, among these a total of 433 miRNAs were homologous to bovine, while 77 were predicted as novel miRNAs that not homologous to any species.The analyses of differential expression revealed 21 DE miRNAs in MDM challenged with MAP compared to the control MDM, Among the DE miRNAs, 14 were upregulated and 7 were downregulated.Furthermore, The GO and KEGG pathway enrichment of these miRNA targets were performed.
