Screening of genes involved in chromosome segregation in meiosis I

Project description:Prophase I of meiosis involves dynamic chromosome segregation processes including synapsis, meiotic recombination, and cohesion. Genetic defects in genes participating in these processes consistently cause reproduction failure in mice. To identify candidate genes responsible for infertility or recurrent pregnancy loss in humans, we performed expression profiling of male and female gonads of mice undergoing meiotic prophase I. Both male and female gonads at 15.5 days postcoitum, neonatal day 1 and 10 weeks postpartum were dissected from mice for RNA extraction and subjected to hybridization on Affymetrix microarrays.

Molecular basis of gender and reproductive status in breeding zebrafish

Project description:Sexual reproduction is a complex process involving multiple pathways. In this study, we aimed at investigating gene expression profiles underlying gender specificity in the zebrafish. To do so, we analyzed the transcriptome of the gonads of breeding fish and queried the gene expression profiles for significant differences between the genders and also between stages of gonad development. Major differences in gene expression profiles between genders were encountered at the level of the gonads. Tissue specificity of gene expression profiling from mussels in normal conditions.

Project description:We examined tissue specificity of gene expression in control mussels. Total RNA was isolated from gills (G), digestive gland (DG), tissues involved in contraction/motility (FML: foot/adductor muscles/ligaments) and reproduction (GM: gonads and mantle) producing four tissue-specific samples. Total RNA was reverse-transcribed and labeled using a Cy3-dCTP and Cy5-dCTP direct incorporation. Each sample was hybridized in competition with the total pool (equal RNA amounts from G, DG, GM and FML) and the relative abundance of each gene was measured by directly comparing fluorescent signals for each probe. We reversed labeling of pool tissues (reference) and single tissue sample in each experiment, thus carrying out two separate hybridizations for each tissue. Project description:In pigs, the peri-implantation period is recognized as determinant of successful reproduction, and it is controlled by neuroendocrinal interactions among signals originating from the female hypothalamic-pituitary-ovarian axis, uterus, and developing embryo. The transcriptomic activity of the pituitary gland may influence reproductive processes occurring in pigs during the peri-implantation period. The aim of this study was to determine alternations in gene expression in the porcine pituitary gland during the peri-implantation period (days 15-16 of pregnancy) in comparison with that during the respective days of the estrous cycle using a microarray approach.

Project description:To elucidate the function of NANOS2 to regulate the transcriptome in embryonic male germ cells, we performed expression microarray analysis of the embyornic gonads of the Nanos2+/-, Nanos2-/- male and wild type female from E12.5 to E15.5. The Nanos2+/- and Nanos2-/- female gonads at E15.5 were analyzed as a negative control. Biological duplicates were examined at each stage, genotype, and sex for each experiment.

Intra-pituitary follicle-stimulating hormone signaling regulates hepatic lipid metabolism

Project description:Inter-organ communication is a major hallmark of health and is often orchestrated by hormones released by the anterior pituitary gland. Pituitary gonadotropes secrete follicle-stimulating hormone (FSH) and luteinizing hormone (LH) to regulate gonadal function and control fertility. Whether FSH and LH also act on organs other than the gonads is debated. Here, we found that gonadotrope depletion in adult female mice triggers profound hypogonadism, obesity, glucose intolerance, fatty liver, and bone loss. The absence of sex steroids precipitates these phenotypes, with the notable exception of fatty liver, which results from ovary-independent actions of FSH. We uncover paracrine FSH action on pituitary corticotropes as a novel mechanism to restrain the production of corticosterone and prevent hepatic steatosis. Our data demonstrate that functional communication of two distinct hormone-secreting cell populations in the pituitary regulates hepatic lipid metabolism.
