Project description:This study examined the potential benefits of male specific-pathogen-free (SPF) White Leghorn embryos in cellular agriculture for sustainable and ethical poultry meat production-addressing traditional farming challenges, including disease outbreaks of Salmonella and Avian influenza. We isolated myogenic precursor cells (MPCs) from the thigh muscles (Musculus femoris) of 12.5-day-old embryos from 10 SPF White Leghorns that tested negative for Salmonella. We randomly selected MPCs from three males and three females, isolated them using a modified pre-plating (pp) method, and compared their in vitro development. After 1 h (pp1) and 2 h (pp2) of incubation, they were transferred to a new dish to remove fast-adhering cells and cultured (pp3). Isolated MPCs had a 69% positive reaction to Pax7. During proliferation, no differences were observed in PAX7, MYF5, or MYOD expression between the male and female MPCs. However, after five days of differentiation, the expression of late myogenic factors-MYOG and MYF6-significantly increased in all MPCs. Notably, MYOG expression was 1.9 times higher in female than in male MPCs. This impacted MYMK's expression pattern. Despite this, the myotube fusion index did not differ between the sexes. Muscle cells from male SPF-laying chicken embryos are promising for developing clean animal-cell-derived protein sources via resource recycling.

Project description:Environmental factors that enhance regeneration are largely unknown. We hypothesized that skin bacteria modulate regeneration. Here, we assessed low, medium, and high levels of bacterial burden in Wound Induced Hair follicle Neogenesis (WIHN), a rare adult organogenesis model. WIHN levels and stem cell markers indeed correlated with bacterial counts, being lowest in germ free (GF), intermediate in conventional specific pathogen free (SPF), and highest even in mice infected with pathogenic Staphylococcus aureus. We identified IL-1β and keratinocyte-dependent IL-1R-MyD88 signaling as necessary and sufficient for bacteria to promote regeneration. Finally, in a small clinical trial, we found that a topical broad-spectrum antibiotic slowed skin wound healing. These results counter conventional notions that infection inhibits regeneration and the need for full sterility of small wounds.

Project description:The widely reported anti-androgenic effects of refined sugar led to the exploration of safer alternatives. Saccharum officinarum molasses (SOM), a byproduct of sugar processing is gaining popularity as a substitute. This study investigated the effects of SOM and compared them to those of refined sugar on male reproductive functions. Blackstrap® Saccharum officinarum molasses were subjected to phytochemical screening and proximate analysis and fractionated to obtain methanol (SOMMF) and aqueous (SOMAqF) fractions. Twelve groups (n?=?5) of adult male Wistar rats received distilled water (Control); 0.8, 2.5, 7.9?g/kg SOM; 0.0064?g/kg sugar (Dangote®); 0.0064?g/kg sugar+7.9?g/kg SOM; 1.0, 3.2, 10.0?g/kg SOMMF and 0.6, 2.0, 6.4?g/kg SOMAqF, respectively. Administrations were done daily by oral gavage for eight weeks. Sperm profile and testicular and epididymal histology were assessed using microscopy. Serum testosterone was quantified using ELISA. Testicular malondialdehyde (MDA) was assayed by spectrophotometry. Data were analyzed using ANOVA at p?<?0.05 significance. Sperm count and viability reduced with 7.9?g/kg SOM, Sugar, 3.2 and 10.0?g/kg SOMMF, 2.0 and 6.4?g/kg SOMAqF. Abnormal sperms increased with 7.9?g/kg SOM, Sugar, 2.0 and 6.4?g/kg SOMAqF. Testosterone level reduced with 6.4?g/kg SOMAqF. Testicular MDA increased with SOM, 3.2 and 10.0?g/kg SOMMF and 6.4?g/kg SOMAqF. Seminiferous tubules and epididymal ducts of 7.9?g/kg SOM, Sugar and SOMAqF-treated rats showed anomalies. Saccharum officinarum molasses altered testicular and epididymal integrity via lipid peroxidation, thus reducing sperm quality and androgen levels in male Wistar rats.

Project description:RationaleAlcohol and nicotine co-dependence is common in humans, and nicotine increases alcohol drinking in humans without alcohol use disorder (AUD). Nevertheless, there is little basic research on the interactions between the reinforcing effects of these two drugs.ObjectivesThe aim of this study was to investigate the effects of chronic nicotine injections on oral alcohol self-administration in alcohol non-dependent rats.MethodsAfter stable alcohol self-administration was reached (baseline) and a period without alcohol access, adult male rats were treated with chronic nicotine or saline injections for 105 days during which time they were tested intermittently for alcohol self-administration. There were 3 experimental groups: (1) saline, rats treated with saline for 105 days; (2) early nicotine, rats treated with nicotine for 70 days, and then with saline for 35 days; and (3) late nicotine: rats treated with saline for 35 days, and then with nicotine for 70 days.ResultsOur results indicate that (1) chronic nicotine increases alcohol consumption regardless of whether exposure to alcohol was interrupted (early nicotine) or not (late nicotine) before the start of nicotine treatment, (2) the number of alcohol reinforcements correlates to blood-alcohol levels, and (3) alcohol self-administration rapidly decreases when nicotine is no longer available (early nicotine).ConclusionsThese discoveries may have clinical implications in social drinkers that use nicotine products, in that chronic nicotine can escalate alcohol drinking and cessation of nicotine exposure may decrease alcohol use.

Project description:In the present study, the effect of probiotics on the hematology of Wistar rats was examined. Locally isolated Lactobacillus plantarum MZ707748 (Pro 1), L. plantarum MZ710117 (Pro 2), Weisella confusa MZ727611 (Pro 3), and L. plantarum MZ735961 (Pro 4) were used. One strain of probiotic, L. acidophilus-14 (Pro 5), was purchased commercially. Different groups were designed as G1, G2, G3, G4, and 5, G5/PC consisting only pro 5 and NC & 0 day were untreated. Different groups have different probiotics like G1 containing Pro 1 and Pro 2, G2 comprising Pro 3 and Pro 4, G3 containing Pro 2, Pro 3 and Pro 5, G4 having Pro 1-5, and G5 containing Pro 5. A complete count of blood, serum chemistry, fecal analysis, and histopathological examination of the thymus and liver were done. Statistical differences were seen in the complete blood count parameters (p < 0.05). No difference was observed in AST, ALT, bilirubin, albumin, IL-6, and IgA (p > 0.05) except for TP, creatinine, and globulin (p < 0.05). Fecal strains of probiotic groups were antibiotic-resistant. In males, Lactobacillus helveticus OQ152020, Enterococcus lactis OQ1519891, E. faecium OQ152017, L. gasseri OQ152017, and E. lactis OQ152019 were isolated from positive control, G1, G2, G3, and G4 respectively. In females, Enterococcus sp. OP800231, Limosilactobacillus fermentum OQ151985, E. lactis OP800267, L. plantarum OP800244, and E. faecium OQ151988 were isolated from positive control, G1, G2, G3 and G4, respectively. It was concluded that all probiotic strains were safe to use and had beneficial effects on the hematology of Wistar rats.

Project description:Acute acrolein inhalation in male rats resulted in multi-tissue transcriptomic alterations that were observed through Illumina mRNA sequencing. Specifically, site-specific respiratory expression profile differences were noted between air- and acrolein-exposed groups. Nasal epithelial tissue demonstrated 452 differentially expressed genes (DEGs) (310 up-regulated and 142 down-regulated)and lung tissue demonstrated 95 DEGs (80 up-regulated and 15 down-regulated). Notable transcriptomic alterations were also observed in liver tissue of acrolein-exposed rats, with 1699 identified DEGs (788 up-regulated and 911 down-regulated). A variety of mRNA expression profile differences resulting from acute acrolein inhalation was observed in other peripheral tissues, including adipose, muscle, adrenals, hippocamus, and hypothalamus. Gene changes were largely representative of oxidative and inflammatory response in the nose, as well as xenobiotic metabolism changes in the lung. Liver changes, which were most numerous, included alterated metabolic signaling (Sirtuin and FXR signaling), as well as alterated oxidoreductive, GPCR, and glucocorticoid pathways. Together, these data demonstrate acrolein, a well-characterized respiratory irritant, induces systemic neuroendocrine immunological and metabolic stress.
