Targeted single cell RNA sequencing of T cells infiltrated in mouse gastric cancer.

ABSTRACT: YTN2 and YTN16 cells are chemically induced gastric cancer cell lines. YTN2 is spontaneously regressed in C57BL/6 mice in CD8+ T cell-dependent manner, while YTN16 grows progressively. To investigate tumor infiltrating T cells, we performed single-cell RNA-Seq of CD4+ and CD8+ T cells in YTN2 and YTN16 gastric tumor-bearing mice for 44 T cell-related genes (Supplementary Table 1).

Single-cell RNAseq of CD45+ cells in murine gastric cancer.

Project description:YTN2 and YTN16 were chemically induced gastric cancer cell lines. When inoculated subcutaneously, YTN2 spontaneously regresses while YTN16 grows progressively. By single-cell RNA-Seq and flow cytometry, we found more IL-17 producing cells in YTN16 than YTN2. Blockade of IL-17 signaling in YTN16 tumor-bearing mice suppressed tumor growth. To investigate tumor-infiltrating cells after IL-17 blockade, we performed single-cell RNA-Seq of CD45+ cells in YTN16 tumors.

Mass spectrometry analysis of the blood plasma and serum samples from healthy donors before and after a meal

Project description:Serum samples were taken from four healthy donors before a meal and 3, 5, 7, 9 h after a meal (Supplementary Table S6, Sheet 1). The analysis of the serum peptidome against the HuMiProt90 database showed that the amount of non-human peptides (potentially bacterial peptides) in the bloodstream significantly increased 5 h after a meal (Figure 3D, Supplementary Table S6, Sheet 3). The comparison of these data with the transit time of food in different parts of the gastrointestinal tract suggests that bacterial peptides enter the bloodstream predominantly in the small intestine. At the same time, phyla Proteobacteria, Firmicutes and Actinobacteria are the main contributors to the increase in (potentially) bacterial peptides 5 h after a meal (Supplementary Table S6, Sheets 5 and 6).
