Project description:Muscle stem cells (MuSCs) are required for muscle regeneration. In resting muscles, MuSCs are kept in quiescence. After injury, MuSCs undergo rapid activation, proliferation and differentiation to repair damaged muscles. Age-associated impairments in stem cell functions correlate with a decline in somatic tissue regeneration capacity during aging. However, the mechanisms underlying the molecular regulation of adult stem cell aging remain elusive. Here, we obtained quisecent MuSCs from young, old, geriatric mice for high resolution mass spectrometry Bruker timsTOF Pro. By comparison of young proteome to old MuSCs proteome or geriatric MuSC proteome, we identified the pathways that are differentially during aging.

Project description:Coordinating mechanical sensing and transcription activation is essential for muscle stem cell (MuSC) function during muscle regeneration. Here we report that MPP7, localized at the apical side of quiescent MuSCs, is an important regulator in MuSC activation, including proliferation and self-renewal. Mechanistically, MPP7 translocates to cell nucleus in response to the change of actin polymerization state with its binding partner AMOT (an actin binding protein) upon MuSC activation and acts to enhance the transcription activity of YAP and TAZ in the MuSC. RNA-seq of MuSCs isolated from control (Con), Mpp7 conditional knockout (cKO), Amot cKO, and Yap and Taz double cKO (YapTaz cKO) after 48 hrs in culture (for activation) in 2 replicas was used to determine the downtream genes regulated by Mpp7, Amot, Yap and Taz. Carm1 was one of the commonly shared down-regulated genes in all 3 cKOs and has been shown to be critical for MuSC self-renewal. We further demonstrated that the MPP7-PDZ domain interacts with TAZ indirectly via AMOT, and the MPP7-L27 domain cooperates with TAZ and another trsncription factor YY1 to regulate a select set of donwstream genes, including Carm1, for MuSC proliferation and self-renewal. Our data identified a coordinated molecular network from mechanical sensing to transcription regulation for MuSC function during muscle regeneration.

Project description:Adhesion between stem cells and their niche promotes stable niche localization and provides signaling information to sustain properties such as quiescence. Skeletal muscle stem cells (MuSCs) are localized between an adjacent myofiber and an enwrapping basal lamina. The most abundant cadherin in MuSCs, M-cadherin, is dispensable for MuSC function, whereas N-cadherin is required to prevent MuSCs from breaking quiescence in the absence of injury. Loss of both cadherins exacerbates this phenotype, yet the MuSCs remain in the niche and maintain regenerative function. These findings raise the possibility that cadherins are dispensable for anchorage of MuSCs to their niche. To address this question, we conditionally removed from MuSCs b- and g-catenin and, separately, aE- and aT-catenin, factors essential for cadherin-dependent adhesion. Catenin-deficient MuSCs break quiescence similarly to N-/M-cadherin-deficient MuSCs, but exit the niche, and are depleted via a single fate: precocious differentiation, reentry to the niche, and fusion to myofibers. These findings indicate that separable, cadherin-dependent functions are required in MuSCs for niche localization, quiescence, and stem cell maintenance.

Project description:Skeletal muscle tissues are comprised of muscle fibers, muscle stem cells (MuSCs), immune cells, endothelial cells, motor/sensory nerves with Schwann cells, and interstitial mesenchymal cells. Each cell population plays a unique role in muscle physiology and homeostasis. The mesenchymal progenitors in the skeletal muscle have been identified as a subpopulation that reside between muscle fibers, whose role is to coordinate muscle regeneration in a finely tuned manner by supporting the expansion and differentiation of MuSCs. These cells have been referred to as fibro/adipogenic progenitor cells (FAPs), termed according to their differentiation potentials in specific in vitro culture conditions and known to cause fibrous tissue and fatty degeneration under pathological environments and result in compromised muscle function. Project description:Adult muscle stem cells (MuSCs) transit from a quiescent state to a highly proliferative state to support muscle repair. Paxbp1 deletion in MuSCs blocked their cell cycle re-entry and caused subsequent muscle regeneration failures. To understand the underlying molecular mechanisms, we compared the transcriptomes of control and Paxbp1 mutant MuSCs by RNA-seq. Our data confirmed a marked repression of cell cycle machineries in Paxbp1 mutant MuSCs. We also uncovered down-regulation of mutliple metabolic pathways in mutant MuSCs. In summary, our study highlights an essential role of Paxbp1 in MuSCs growth and cell cycle re-entry.

We generated a network connecting age-affected ligands located in the niche and cell surface receptors on MuSCs. Thereby, we revealed signaling via Integrins, Lrp1, Egfr and Cd44 as the major cell communication axes perturbed through aging. We investigated the effect of Smoc2, a secreted protein that accumulates with aging, originating from fibro-adipogenic progenitors. Increased levels of Smoc2 contribute to the aberrant Itgb1/MAPK signaling observed during aging, thereby causing impaired MuSC functionality and muscle regeneration. By connecting changes in the proteome of MuSCs to alterations of their niche, our work will enable a better understanding of how MuSCs are affected during aging.

We found that the mammalian Hairless (Hr) gene is expressed in quiescent MuSCs. Using a mouse model in which Hr can be specifically ablated in MuSCs, we demonstrate that Hr expression is critical for MuSC function and muscle regeneration. We show that Hr is a histone demethylase antagonist that preserves Histone 3 Lysine 9 trimethylation (H3K9me3) and maintains heterochromatin structure. Loss of Hr results in reduced H3K9me3 levels, reduced heterochromatin, increased susceptibility of MuSCs to genotoxic stress, and the accumulation of DNA damage. Our study not only elucidates the molecular mechanism by which Hr regulates histone demethylases, but also demonstrates the importance of heterochromatin maintenance in stem cell function.
