Analysis of Foxp3 binding DNA in mouse bone marrow derived macrophages stimulated with brain derived DAMPs

Project description:Treg-specific transcription factor Foxp3 is also expressed in macrophages. Macrophages play an important role in the pathological process of stroke, and their Foxp3 is significantly up-regulated after stroke, and the underlying mechanism is still unclear. The purpose of this study was to investigate the function of Foxp3 as a transcription factor in macrophages. Ten million bone marrow-derived macrophages were treated with mouse brain-derived danger-associated molecular patterns, and experiments were performed 2 hours later using CST's Cut&Run kit and Foxp3 antibody. The obtained DNA was subjected to sequencing analysis. Our findings suggest that the expression of foxp3 is associated with the phagocytosis and digestion of macrophages, and the molecular may be a potential target for stroke. CUT & RUN and DNA product sequencing for Foxp3 in bone marrow-derived macrophages (BMDMs)

Furin deficiency efect on T cells

Project description:Analysis of T-cells lacking the proprotein convertase furin. Proprotein convertases promote the proteolytic maturation of proproteins. Furin is induced in activated T-cells. Results provide insight into the function of furin in T-cells. CD4+CD62L+CD44- naive, CD4+CD62L-CD44+ memory and CD4+CD25+FoxP3+ regulatory T cells were isolated from Fur flox/flox and CD4 cre Fur flox/flox mice. Naive T cells were activated via TCR. Total RNA was extracted from all cells and hybridized to Affymetrix microarrays.
