Modeling Alcohol-induced Neurotoxicity using Human Induced Pluripotent Stem Cell-derived Three-Dimensional Cerebral Organoids

ABSTRACT: We performed transcriptomic studies using microarray assays and conducted bioinformatics analyses to identify signaling pathways associated with ethanol-induced brain injury in cerebral organoids A human message RNA (mRNA) Expression Microarray assay (V4.0) and data analysis services were provided by Arraystar Inc. (Rockville, MD) to assess and compare the global abundance of 17,195 mRNAs across iPSC-derived 2-month cerebral organoids treated with and without ethanol.

Project description:To identify lncRNAs enriched in ECs, a microarray was performed to profile ~30,000 lncRNAs and ~26,000 coding transcripts using an Arraystar human LncRNA microarray v3.0 system (Arraystar, Rockville, MD). Three primary human EC lines and two non-EC lines at low passages, namely, human umbilical vein EC (HUVEC), human retinal EC (HREC), human choroidal EC, human dermal fibroblast cell (HDF) and human retinal pigment epithelial (RPE) cell lines, were used in the array. Analyses of long non-coding RNAs in human retinal endothelial cells following glucose treatment

Project description:The differential expression of lncRNAs in human retinal endothelial cells (HRECs) was investigated with a commericially available microarray (ArrayStar Human LncRNA Microarray V3.0; ArrayStar Inc., Rockville, Maryland USA). Approximately 28,035 lncRNAs and 21,407 coding transcripts were detected using this microarray. In addition to the array findings, lncRNAs were further constructured using transcriptome databases (including RefSeq, Gencode, UCSC). Sequences were selected using proprietary strategies and each transcript is represented by a specific exon or splice junction probe, which can accurately identify an individual transcript. Nevertheless, both postive and negative probes for housekeeping genes were additionally used as controls.
