ABSTRACT: Compare with normal nasopharyngeal epithelial cells, we found ACAT1 was decreased in NPC cells, we found that ACAT1 inhibited proliferation, colony formation, migration and invasion in NPC cells. Project description:Analysis of differential expression genes in NESG1-overexpressed and negative nasopharyngeal carcinoma. NESG1 is a candidate tumor suppressor in NPC. Results provide insight into the molecular pathogenesis of NESG1 in NPC. NESG1 cDNA was transfected into NESG1-negative NPC 5-8F cells and affymetrix microarrays HG-U133_Plus_2 were used to analyze the differential genes in NESG1-overexpressed NPC cells and their control NESG1-negative NPC cells.

Project description:Nasopharyngeal carcinoma (NPC) is a common cancer in southern China and South East Asia where more than 50,000 new cases are diagnosed each year. We used microarrays to identify down or upregulated genes in NPC compared with non-malignant controls. Experiment Overall Design: Snap frozen nasopharyngeal biopsies from 25 patients with histologically confirmed undifferentiated NPC were included in the microarray analysis. Controls were obtained from 3 patients with no evidence of malignancy.

Expression profiling of nasopharyngeal carcinoma cell lines

Project description:To identify the deregulated genes which may involved in the carcinogenesis of nasopharyngeal carcinoma (NPC). To identify potential tumor suppressor genes / oncogenes of NPC, we have employed whole genome microarray expression profiling as a discovery platform to analyze two NPC cell lines, TW01 and HONE1. For the purpose to filter out differential genes may not related to carcinogenesis of NPC, a gene expression database for premalignant nasopharyngeal epithelial cells was obtained from NP460hTert cells, and we used the commercial universal human RNA (stratagene) as common reference for all microarray experiments. A total of 1272 genes were commonly up-regulated (fold change > 2) and 1810 genes were commonly down-regulated (fold change < 0.5) in TW01 and HONE1 cell lines, whereas not deregulated in NP460hTert cells.

Genome-wide microRNA analysis of NPC and non-NPC specimens gene expression

Project description:Analysis of differential expression between NPC and non-NPC tissues at miRNA expression level. We used a miRNA microarray platform to investigate the profile between NPC tissues and normal nasopharyngeal tissues to evaluate the relationship between miRNA expression and NPC. Recognition of the aberrant miRNAs and enrichment pathway analysis of predicted target gene may provide some clues to estimate the mechanism of miRNA effects on NPC carcinogenesis mediated by miRNA-mRNA regulation. Total RNA was extracted from 8 NPC tissues compared to 4 nasopharyngeal tissues. We peformed the Illumina miRNA microarray to identify the differentially expressed miRNA between NPC tissues and normal nasopharyngeal tissues.
