Project description:LV hypertrophy is associated with Western diet consumption, while intake of n-3 polyunsaturated fatty acids is associated with anti-hypertrophic effects. We treated rats for 12 weeks with either a Control diet, a Western diet or a Western + DHA diet. For each of the 3 dietary treatments there were 2 pooled samples of heart tissue (with each pooled sample representing 5 rats) for a total of 6 arrays. Microarray analysis identified 66 differentially expressed transcripts. Pathways were identified using Ingenuity and DAVID software. Array results from two pooled samples (5 rats in each pool) for n=10 per treatment group were used for comparisons. Comparisons between Western vs. Control, Western + DHA vs. Control and Western + DHA vs. Western diets was subjected to analysis to generate log fold changes. A dietary treatment of 12 weeks was used in an effort to produce LVH while limiting the development of comorbidities. Microarray analysis was performed on pooled samples, followed by qRT-PCR and Western blot analysis. Groups were Control, Western and Western + DHA. Comparisons between groups are expressed as LogFC (i.e. LogFC_WESvCTRL, LogFC_DHAVCTRL, LogFC_DHAvWES), available in Series supplementary files.

Project description:The total abundance of phosphatidylcholine (PC) is known to influence lipoprotein production. However, the role of specific phospholipid species in lipid transport has been difficult to assess due to an inability to selectively manipulate membrane composition in vivo. Here we show that the LXR-regulated phospholipid remodeling enzyme lysophosphatidylcholine acyltransferase 3 (Lpcat3) is a critical determinant of membrane phospholipid composition and lipoprotein production. Mice lacking Lpcat3 in the liver show defects in lipoprotein production. The objective of generating this dataset was to analyze the effects of Lpcat3 loss of function on gene expression in mouse liver with a western diet challenge. This dataset compares gene expression in Lpcat3fl/fl and Lpcat3fl/fl Albumin-Cre liver samples from 16-week old male mice that were fed a western diet for 9 weeks since 7 weeks old. Each sample contains tissues from 5 mice.

Ldlr-/- huCETP transgenic mice were fed a low fat, cholesterol-containing western diet (LFWD, 9% fat, 0.15% cholesterol) starting at 8 weeks of age for 4, 8, 12, and 16 weeks (Supplementary Figure 1). At each time point, 10 mice were studied. Mice were euthanized at each time point and the aortic arch of each mouse was immediately collected and flash-frozen in liquid N2. Mice were fasted overnight prior to euthanasia. the aortic arches of 10 mice in the same study group were pooled for profiling, resulting in one pool per study group. Tissues were homogenized and total RNA was extracted. The control RNA pool was composed of equal aliquots of RNA derived from the 4 pools (40 samples) from the LFWD 4-, 8-, 12- and 16-week time points.

In contrast, myeloid cell responses towards innate stimuli remained broadly augmented. WD induced transcriptomic and epigenomic reprogramming of myeloid progenitor cells, leading to increased proliferation as well as enhanced innate immune and interferon responses towards in vivo LPS challenge. QTL analysis in human monocytes trained with oxidized low-density lipoprotein (oxLDL) and stimulated with LPS suggested inflammasome-mediated trained immunity. Consistently, Nlrp3-/-/Ldlr-/--deficient mice lacked WD-induced systemic inflammation or myeloid progenitor proliferation and reprogramming. Hence, NLRP3 mediates trained immunity following WD and could thereby arbitrate the potentially deleterious effects of trained immunity in inflammatory diseases.

Project description:Our understanding of how atherosclerosisr risk factors persist after risk factor mitigation is incomplete. Here, we use oral infection with Porphyromonas gingivalis (Pg) as a model risk factor. Atherosclerosis-prone LDLR Komice were infected with Pg or sham infected, and fed a high fat diet for 16 weeks. Their bone marrow was then injected into lethally irradiated naive LDLR KO mice. After 16 weeks of high fat diet feeding, macrophages were elicited into the peritoneal cavity with thioglycollate. DNA from these macrophages was isolated for WGBS sequencing. We hypothesize that oral infection with Pg caused methylation differences in genes that contribute to increased atherosclerosis.
