Project description:Cattle are often fed high concentrate diets to increase energy intake and improve overall animal performance. Such diets also cause changes in fermentation patterns and epithelial function. However, the molecular mechanisms involved in regulating epithelial function for cattle fed high concentrate diets have not been elucidated. In this study, we aimed to gain a broad overview of the involved molecular mechanisms by detecting differentially expressed genes (DEG) in rumen tissue from dairy cows fed a low concentrate (LC; 8%) compared to a high concentrate (HC; 64%) diet using a bovine-specific microarray platform containing 16,846 unique gene loci and 5,943 ESTs from the bovine genome. Feeding the HC diet increased the total volatile fatty acid concentration and markedly reduced ruminal pH, suggesting that the dietary treatments used did induce changes in ruminal fermentation. In response to changes in the ruminal environment, a total of 5,200 elements were detected as DEG in ruminal tissue with >1.5-fold expression change (P < 0.05) for cows fed HC relative to LC. Of the 5,200 DEG, 2,233 and 2,967 were up- and down-regulated, respectively. The GENECODIS analysis elucidated that relationships among the DEG represented 19 annotations characterized with GO molecular function and KEGG pathways with 26 DEG identified in multiple annotations such as calcium signaling and gap junction pathways. Among those DEG that were identified numerous times, catalytic subunit of cAMP-dependent protein kinase (PRKACB) was down-regulated in ruminal tissue from cows fed HC, suggesting that this gene may have important roles including regulation of cell proliferation and differentiation, and intracellular pH regulation. Project description:Purpose: The present study was designed to identify both differentially expressed (DE) genes in the liver tissues of fattening Merino lambs and differences in metabolites accumulated in plasma, thus trying to identify modified metabolic pathways as a consequence of milk restriction during the suckling period. Methods: Twenty-four male Merino lambs were assigned to 2 different groups (n=12 per dietary treatment). The first group (ad libitum, ADL) was kept permanently with the dams whereas the other group (restricted, RES) was milk restricted. When they reached 15 kg of live body weight, all the lambs were offered the same complete pelleted diet at the same level to ensure no differences in dry matter intake. All the lambs were slaughtered with 27 kg. For transcriptomic analysis, 4 liver samples representative from each group (8 samples in total) were selected for RNA sequencing methodology (RNA-seq). Results: 38 DE annotated genes were identified by RNA-seq, with 23 DE genes being down-regulated and 15 up-regulated in the liver of RES relative to the ADL group (P < 0.10). In general, those genes and pathways involved in protein synthesis or protease inhibitors were down-regulated in the RES group, whereas those related to proteolytic degradation were up-regultated, thus suggesting a higher catabolism of proteins in these lambs. Contrarily, RES lambs showed over-expression of xenobiotic metabolism pathways, whereas those genes related to β-oxidation of fatty acids were down expressed. Conclusions: According to the data obtained, early feed restriction during the suckling period of Merino lambs promoted long-term effects on hepatic transcriptomic profile and plasma metabolic profile wich might have modified fatty acids metabolism, catabolism of proteins and detoxification of xenobiotics, thus reducing feed efficiency of fattening period.

Project description:This study aimed to evaluate the effect of including soybean molasses (SM) on performance, blood parameters, carcass traits, meat quality, fatty acid and muscle (longissimus thoracis) transcriptomic profiles of castrated lambs. Twenty Dorper × Santa Inês lambs (20.06 ± 0.76 kg BW) were assigned to a randomized block design, stratified by BW, with the following treatments: CON - 0 g/kg of SM and SM20 - 200 g/kg of SM on DM basis, allocated in individual pens. The diet consisted of 840 g/kg concentrate and 160 g/kg corn silage for 76 days, with the first 12 days as an adaptation period and the remaining 64 days on the finishing diet. The SM20 diet increased blood urea concentration (P = 0.03) while reduced glucose concentration (P = 0.04). Lambs fed SM showed higher subcutaneous fat deposition (P = 0.04) and higher subcutaneous adipocyte diameter (P < 0.01), in addition to reduced meat lipid oxidation (P < 0.01). Soybean molasses reduced the quantity of branched-chain fatty acids in longissimus thoracis (P = 0.05) and increased the quantity of saturated fatty acids (P = 0.01). In the transcriptomic analysis, 294 genes were identified as differentially expressed, which belong to pathways such as oxidative phosphorylation, citric acid cycle, and monosaccharide metabolic process. In conclusion, diet with SM increased carcass fat deposition, reduced lipid oxidation, and changed the energy metabolism, supporting its use in ruminant nutrition.

Project description:Cistus ladanifer L. is a common shrub endemic to the Mediterranean that is highly concentrated in condensed tannins (CT). CT form complexes with dietary protein that resist microbial degradation in the rumen, which enhances dietary protein utilization in ruminant diets. The objective of this study was to evaluate the utilization of CT in the diet of lambs on the proteomes of muscle, hepatic and adipose tissues. Twenty-four white merino ram lambs were divided in three groups (n=8) fed on different diets: control (16% crude protein - CP), reduced protein (12% CP) and reduced protein treated with CT extract. At the end of the trial, lambs were slaughtered and the longissimus lumborum muscle, hepatic and peri-renal adipose tissues sampled. A two-way approach was used for shotgun proteomic analysis: 2D-DIGE (gel-based) and nanoLC-MS (gel-free). In the muscle, control lambs had lower abundance proteins that partake in the glycolysis pathway. With dietary 12 % CP, lambs had higher abundance of Fe-carrying proteins in the hepatic tissue. Lambs with dietary CT had higher abundance of hepatic flavin reductase. In the adipose tissue, control lambs had lower abundance of fatty-acid synthase. In conclusion, CT inclusion influences specific pathways in lamb tissues.

Project description:Feeding animals with either concentrates or alfalfa grazing has been proven to reduce the oxidative process that occurs in meat products. Indoor-kept lambs were fed a standard concentrate (n=7, C) before slaughtering all animals at 22–24 kg of live weight. Simultaneously, 7 unweaned lambs grazed in alfalfa paddocks (ALF) with their dams. Global transcriptomic data of liver with the Affymetrix® Ovine Gene 1.1 microarray was used. When ALF group was compared with C group, were identified 96 genes differentially expressed. Among these genes 92 were down- regulated and 4 were up- regulated. The clusters corresponding to gene expression profiles from treatments were clearly separated from each other. These differentially expressed genes were selected for a functional analysis by using DAVID. Three major gene clusters associated with “sterol biosynthesis (EBP, MVD, HMGCR, CYP51A1, HMGCS1, NR0B2, C14ORF1, FDFT1, SQLE, DHCR7, SC5DL, DHCR24, NSDHL) , “lipid biosynthetic process (ACACA, CYP51A1, FADS1, FADS2, SCD y SC5DL)”, “cholesterol metabolic process (EBP, MVD, HMGCR, CYP51A1, SQLE, DHCR7, HMGCS1, NR0B2, DHCR24, FDFT1, NSDHL)” were found.

Project description:The objective of this study was to examine the effect of dietary restriction and subsequent re-alimentation induced compensatory growth on the global gene expression profile of ruminal epithelial papillae. Holstein Friesian bulls (n=38) were assigned to one of two groups: restricted feed allowance (RES; n=19) for 125 days (Period 1) followed by ad libitum access to feed for 55 days (Period 2) or (ii) ad libitum access to feed throughout (ADLIB; n=19). All bulls received the same diet of 70% concentrate 30% grass silage through out the experimental trial,with the amount of feed provided different dependent on each treatment group. At the end of Period 1, 9 animals from each treatment group were slaughtered, with 10 animals from each treatment slaughtered at the end of Period 2. Rumen epithelium was collected from all animals within thirty minutes of slaughter. RNA was extracted and rumen epithelium gene expression was examined using RNAseq technology on all samles collected (end of Period 1: 9 samples each from ADLIB and RES groups; end of Period 2: 10 samples each from ADLIB and RES groups). Genes identified as differentially expressed in response to both dietary restriction and subsequent compensatory growth included those involved in processes such as cellular interactions and transport, protein folding and gene expression, as well as immune response. This information can be exploited in genomic breeding programmes to assist selection of cattle with a greater ability to compensate following a period dietary restriction.

Project description:Purpose: The present study was designed to identify both differentially expressed (DE) genes and differences in proteins accumulated in the liver tissues of suckling female lambs, thus trying to identify modified metabolic pathways as a consequence of milk restriction during the suckling period. Methods: Forty Assaf lambs (average BW 4.7 kg) were penned individually, twenty of them were fed milk replacer (200 g dry matter/L) ad libitum (ADL; 192 mL/kg LBW) whereas the other group (restricted, RES) only received 120 mL/kg LBW. When they were 35 days old, four animals per group were slaughtered (8 lambs in total) and a piece of liver was excised for transcriptomic analysis. The liver transcriptome analysis was carried out using RNA sequencing methodology (RNA-seq). Results: 386 DE genes were identified by RNA-seq, 198 of them being annotated genes in the KEGG pathway. Positive values of log2-fold change (log2FC) indicated that 210 genes were up-regulated in the liver of RES relative to the ADL group, whereas negative log2FC values denoted the down-regulation of 176 genes (P < 0.05). Conclusions: According to the data obtained, a restricted milk intake during the suckling period of replacement lambs affects hepatic transcriptome and proteome associated with an altered metabolism of lipids and proteins, thus reducing feed efficiency of replacement period.
