Characterization of Arabidopsis transcriptome responses to different aphid species reveals genes that contribute to host susceptibility and nonhost resistance

Project description:Background: Aphids are economically important pests and that display exceptional variation in host range. The underlying mechanism of diverse aphid host ranges are not well understood, but it is likely that molecular interactions are involved. With significant progress being made towards understanding host responses upon aphid attack, the mechanisms underlying nonhost resistance remain to be elucidated. Here, we investigated and compared Arabidopsis host and nonhost responses to aphids at the transcriptome level using three different aphid species. Results: Gene expression analyses revealed a surprising level of overlap in the overall gene expression changes during host and nonhost interactions with regards to the sets of genes differentially expressed and the direction of expression changes. Despite the overlap in transcriptional responses across interactions, there was a stronger repression of genes involved in metabolism and oxidative responses specifically during the host interaction. In addition we indentified a set of genes with opposite gene expression patterns during host versus nonhost interactions. Aphid performance assays on Arabidopsis mutants selected based on our transcriptome analyses identified genes involved in host and nonhost interactions. Conclusions: Understanding how plants respond to aphid species that differ in their ability to infest plant species, and identifying the genes and signaling pathways involved, is essential for the development of novel and durable aphid control in crop plants. Our work is an important step forward to provide such essential insights in that we identified novel genes contributing to host susceptibility, host defences as well nonhost resistance to aphids.

Morphological and transcriptomic analysis reveal robust nonhost resistance response during chickpea interaction with Alternaria brassicae

Project description:Alternaria brassicae is a necrotrophic fungal pathogen which infects brassica crops and lead to huge loss in crop production. There is a need to exploit the novel resistance mechanism against A. brassicae. Under field condition, chickpea is the potential nonhost plant for A. brassicae. At molecular level, it is not known how the NHR in chickpea operates against A. brassicae. In present study, we did the transcriptomic analysis in chickpea plants exposed to nonhost pathogen, A. brassicae by using microarray. Chickpea plants were spray inoculated with the spore suspension of A. brassicae. The leaf samples were harvested after 24 hpi and 48 hpi from pathogen treated plants and from the mock-treated control plants. The Transcriptome analysis were done from the leaf samples obtained at both the time-points by microarray using Agilent ChickpeaGXP_8X60K chip. Our result suggested the robust transcriptional reprogramming leading to defense response against A. brassicae.

Identification of candidate genes for nonhost resistance to powdery mildew in wheat

Project description:To identify the nonhost resistance genes, a time-series based global expression profiling was performed in wheat using Agilent gene expression 44K microarray array. Seven-day-old wheat plants of cv. Renan was inoculated with spore density of 50-80 conidia mm-2 of adapted (Bgt) or non-adapted (Bgh) powdery mildew pathogen, and the abaxial epidermis of inoculated primary leaves or from non-inoculated control leaves was peeled at 6, 12, 24 and 74 h after inoculation. Total RNA was extracted by using the RNeasy plant mini kit with on-Column DNase digestion (Qiagen, Hilden, Germany), and hybridized to a 44K Agilent oligonucleotide custom array of wheat.

Project description:Powdery mildew is a very common plant disease and only few plants are immune. Host interactions have been identified and characterized for the pathosystems barley-B. graminis f. sp. tritici (Bgt) and wheat-B. graminis f. sp. hordei (Bgh), whereas no data are reported about powdery mildew and nonhost plants, such as rice. On the other hand rice nonhost resistance is widely unexploited and only few expression data are available. To characterize rice response during nonhost interaction with Bgh, a global expression analysis was performed by using the GeneChipM-BM-. Rice Genome Array. To describe rice gene expression profiles during nonhost interaction, 2 week-old rice plantlets were inoculated with Bgh. Treated (inoculated) and control (mock) samples were collected 24 hours post-inoculation for GeneChipM-BM-. Rice Genome Array hybridization. For transcript proM-oM-,M-^Aling experiments with powdery mildew, entire leaves were sampled from rice plantlets 2 weeks old (cv. Nipponbare). Treated and control (mock) rice leaves were collected 24 hours post inoculation. A Novel Pear Scab (Venturia nashicola) Resistance Gene, Rvn3, from Interspecific Hybrid Pear (Pyrus pyrifolia × P. communis).

Project description:Asian pear scab is a fungal disease caused by Venturia nashicola. The identification of genes conferring scab resistance could facilitate the breeding of disease-resistant cultivars. Therefore, the present study aimed to identify a scab-resistance gene using an interspecific hybrid population ((Pyrus pyrifolia × P. communis) × P. pyrifolia). Artificial inoculation of V. nashicola was carried out for two years. The segregation ratio (1:1) of resistant to susceptible individuals indicated that resistance to V. nashicola was inherited from P. communis and controlled by a single dominant gene. Based on two years phenotypic data with the Kruskal-Wallis test and interval mapping, 12 common markers were significantly associated with scab resistance. A novel scab resistance gene, Rvn3, was mapped in linkage group 6 of the interspecific hybrid pear, and co-linearity between Rvn3 and one of the apple scab resistance genes, Rvi14, was confirmed. Notably, an insertion in pseudo-chromosome 6 of the interspecific hybrid cultivar showed homology with apple scab resistance genes. Hence, the newly discovered Rvn3 was considered an ortholog of the apple scab resistance gene. Since the mapping population used in the present study is a pseudo-BC1 population, pyramiding of multiple resistance genes to pseudo-BC1 could facilitate the breeding of pear cultivars with durable resistance.
