RNA-seq of induced pluripotent stem cell or somatic stem cell-derived corneal epithelial cell sheet

ABSTRACT: Our previous study indicate that corneal epithelial cell sheet (CEPS) derived from stem cells, including induced pluripotent stem cells (iPSCs), has low immunogenicity. Therefore, we hypothesized that CEPS express immunosuppressive molecules. To address this hypothesis, we performed RNA-seq of human iPSC-derived CEPS (iCEPS) and somatic stem cell-derived CEPS (sCEPS) with or without human recombinant IFN-g (rIFN-g) treatment.

Differentiating human embryonic stem cells into corneal epithelial-like cells

Project description:Transplantation of ex vivo expanded limbal stem cells (LSC) is the main treatment for limbal stem cell deficiency though the clinical problem of donor tissues shortage. Recently, as the development of tissue engineering, embryonic stem cells (ESC) derived corneal epithelial-like cells (ESC-CEC) has become a new direction to this issue.Our group successfully induced ESC into corneal epithelial-like cells, and in the present study we explored various aspects of physiological properties of ESC-CEC. The experiment included three samples: hES, the human embryonic stem cell line H1, RA_SB, the corneal epithelial-like cells derived from hES by differentiation with RA and SB, epithelial_cell, the primary human limbal stem cells from cadaver eyes. hES, the human embryonic stem cell line H1, RA_SB, the corneal epithelial-like cells derived from hES by differentiation with RA and SB, epithelial_cell, the primary human limbal stem cells from cadaver eyes.

Directed differentiation of human embryonic stem cells to corneal endothelial cell-like cells: A transcriptomic analysis

Project description:The corneal endothelium is composed of a monolayer of corneal endothelial cells (CECs), which is essential for maintaining corneal transparency. Human embryonic stem cells (hESCs) hold the promise of providing an abundant donor source for generating CEC cells for cell replacement therapies. Here we demonstrate that CEC-like cells can be efficiently derived from human ESCs. In addition, we performed global gene expression profiling of stem-cell-derived CEC cells, incorporating with adult CEC cells, fetal CEC cells and other human tissue type data. Our data indicate that hESC-derived CEC-like cells closely resemble human fetal CEC cells.

Pluripotent stem cell-derived corneal endothelial cells as an alternative to donor corneal endothelium in keratoplasty

Project description:Here, we evaluate the efficacy of cryopreserved human embryonic stem cell (hESC)-derived corneal endothelial cells (CECs) to form a functional monolayer of corneal endothelium (CE) in mammals (rabbits) and non-human primates (monkeys). We injected cryopreserved hESC-derived CECs in rabbits and monkeys either immediately after removing 8 mm of the central portion of the CE or a few days later when corneal edema developed. All clinical models developed deturgesced and clear corneas 2-3 weeks following the CEC injection and remained comparable to the CE of the untreated eye. Confocal scanning microscopy confirmed an intact structure of hexagonal/polygonal cells and immunohistochemical analysis illustrated a monolayer expressing barrier and pump function proteins in the regenerated CE. The necropsy examination confirmed no remarkable change in multiple tissues examined for teratoma formation. In conclusion, our data demonstrate the efficacy of cryopreserved hESC-derived CECs to form a functional CE on the denuded Descemet’s membrane.

Single-cell RNA-seq of corneal epithelial cells

Project description:Stem cells are essential for maintaining homeostasis and regenerating damaged tissue. In the corneal epithelium, the location and molecular markers of corneal epithelial stem cells are not fully understood. Recent advances in single-cell RNA sequencing has offered unprecedented opportunity to unveil transcriptional differences across corneal cell types. We report the application 10x Genomics Chromium technology for high-throughput single-cell RNA sequencing of transparent central corneal epithelium, and identified a cluster of facultative stem/progenitor cells that exhibits colony-forming ability and self-renewal capacity. This study provides new insight into facultative adult stem cell biology.

Single-cell transcriptomic analysis of corneal organoids during development

Project description:Corneal organoids are useful tools for disease modeling and tissue transplantation however they have not yet been well studied during maturation. We characterized human iPSC derived corneal organoids at 1, 2, 3 and 4 months of development using single-cell RNA sequencing to determine the cellular heterogeneity at each stage. We found pluripotent cell clusters committed to epithelial cell lineage at 1 month early corneal epithelial, endothelial and stromal cells markers at 2 months, keratocytes as the largest cell population at 3 months, and a large epithelial cell population at 4 months. We compared organoid to fetal corneal development at different stages and found that 4 month organoids closely resemble the corneal cellular complexity of the fetal (16 post conception week) and adult cornea. Using RNA velocity pseudotime trajectory analysis, we found that eye field primordial cells and limbal stem cells appear to give rise to corneal epithelial cells during development.

Gene expression of sheep epidermal stem cells and corneal epithelia cells

Project description:Corneal epithelial cells (CECs) are required for corneal transparency and visual function, and corneal injuries may cause corneal blindness. Skin epidermal stem cells (SESCs), which share the same origin with CECs and have the potential of multi-directional differentiation are ideal seed cells for tissue engineered corneal construction to treat corneal blindness. To identify critical genes and pathways that modulate transdifferentiation from SESCs to CECs, we isolated and cultured the sheep SESCs and CECs and then compared gene expression of SESCs and CECs using microarray.
