Gene expression profile induced by moderate physical exercise in heart left ventricles in rats

Project description:Physical exercise training is a known protective factor against cardiovascular morbidity and mortality. Nevertheless, the underlying specific molecular mechanisms still remain uncompletely explored. To identify molecular mechanisms by which exercise training induces this favorable phenotype a genomic approach was used in an animal model of mild exercise previously demonstrated by our group to induce cardioprotection. Our data indicate that mild exercise by inducing no or few permanent changes in gene expression profile is able to determine cardioprotection without induction of cardiac hypertrophy. Development of a hepatic cryoinjury model to study liver regeneration

Project description:We developed a new model to study liver regeneration based on cryoinjury. Hepatic cryoinjury induced a localised necrotic and apoptotic lesion characterised by inflammation and infiltration of innate immune cells. Following this initial phase, we observed fibrosis, which resolved as regeneration re-established homeostasis in 30 days. Importantly, this approach enables the comparison of healthy and injured parenchyma with an individual animal, providing unique advantages to previous models. These datasets include Bulk RNA-seq of livers collected at sham, 1 dpci, 3 dpci, and 7 dpci.

Project description:Contrary to mammals, zebrafish regenerate their heart upon cryoinjury of the ventricular apex. Regeneration is preceeded by a transient fibrotic response. Here we compare the expression profile of fibroblast-like cells at 7 different time points of fibrosis resolution. Using a postnb:CreERT2; ubb:loxP-GFP-loxP-mCherrycz1701 double transgenic line, we permanently label cells that expressed postnb at 3 and 4 days post injury (dpi) with mCherry by administration of 4-OHT. We sequenced mCherry-labelled cells obtained from the ventricular apex at 7 and 60 dpi.

Morphine alleviates pain after heart cryonjury in zebrafish without impeding regeneration

Project description:Nociceptive response belongs to a basic animal behavior facilitating adaptability and survival upon external or internal stimuli. Fish, similarly to higher vertebrates, also possess nociceptive machinery. Current protocols involving procedures performed on adult zebrafish including heart cryoinjury do not, however, take into account the adverse effects including pain that may potentially arise from these methodologies. Here, we assess the effect of two analgesics, lidocaine and morphine, followed after the heart cryoinjury in zebrafish. Monitoring swimming behaviour together with histology and gene expression analysis at the single cell level using scRNA sequencing and RNAscope fluorescent in situ hybridization technology, we show morphine, but not lidocaine, significantly improves animal welfare 6 hours post-cryoinjury, without impairing heart regeneration process. Altogether, morphine should be considered as the analgesic of choice to reduce post-surgical pain in adult zebrafish.

Changes in the transcriptome of circulating neutrophils and skeletal muscle in response to endurance exercise

Project description:Analysis of the changes in the transcriptome of circulating neutrophils and skeletal muscle from standardized resting conditions (baseline; pre-EXTRI) to 3, 48 and 96 hours after an experimental exercise trial (EXTRI; 1 hour of cycling followed by 1 hours of running) in 8 healthy, endurance-trained, male subjects. It was hypothesized that the time-course dependent transcriptomic changes would reflect the molecular and signalling mechanisms by which neutrophils regulate and counter-regulate inflammation, and by which skeletal muscle responds, regenerates, and phenotypically adapts to intense, prolonged exercise involving muscle damage. Results provide an important insight in the signalling pathways underlying the transcriptional activation and priming of circulatory neutrophils in response to physiological stress, in particular muscle-derived damage-associated molecular patterns. Furthermore, the study provides novel data on the skeletal muscle transcriptome beyond 48 hours after strenuous endurance exercise, and indicates important muscular remodelling processes at 96 hours post-EXTRI.

Moderate-Intensity Physical Exercise Affects the Exercise Performance and Gut Microbiota of Mice.

Project description:The gut microbiota is closely associated with the health of the host and is affected by many factors, including exercise. In this study, we compared the gut microbial changes and exercise performance over a 14-week period in mice that performed exercise (NE; n = 15) and mice that did not perform exercise (NC; n = 15). Mice were subjected to stool collection and exercise tests one week prior to adaptive training and after 2, 6, 10, and 14 weeks of exercise. Bacteria associated with the stool samples were assessed via Illumina-based 16S rRNA gene sequencing. While there was no significant difference in body weight between the groups (p > 0.05), the NE group had a significantly higher exercise performance from weeks 2-14 (p < 0.01) and lower fat coefficient (p < 0.01) compared with the NC group. The Shannon index of the gut microbiota in the NC group was higher than that in the NE group at weeks 6 and 10, and the Chao1 index was higher than that in the NE group at week 14. Exercise performance positively correlated with the relative abundance of Phascolarctobacterium. Grouped time series data analysis demonstrated that Bifidobacteria, Coprococcus, and one unnamed genus in the Clostridiales order were significantly increased in the NE group, which correlated with increased glucose, flavonoid, arginine, and proline metabolism. In conclusion, moderate-intensity treadmill exercise significantly increased the exercise performance of mice and changed the core bacteria and bacterial metabolic activity. These results provide a reference for studying the effects of exercise intervention and exercise performance on the gut microbiota of mice.
